Het gebruik van röntgenkristallografie voor de bepaling van hoge resolutie structuren van biologische macromoleculen heeft de afgelopen twee decennia een constante progressie beleefd. De groeiende opname van röntgenkristallografie door niet-deskundige onderzoekers illustreert de democratisering van deze benadering op vele gebieden van de levenswetenschappen 1 . Historisch gezien zijn kristallen met afmetingen onder ~ 10 μm beschouwd als uitdagend, indien niet onbruikbaar, voor structuurbepaling. De toenemende beschikbaarheid van toegewijde microfocus bundellijnen voor synchrotron stralingsbronnen wereldwijd en technologische vooruitgang, zoals de ontwikkeling van gereedschappen om microkristallen te manipuleren, hebben veel van deze barrières verwijderd die het brede gebruik van röntgen microkristallografie hebben verbannen. Vooruitgang in seriële röntgen microkristallografie 2 , 3 en micro elektronen diffractie 4 haHet is gebleken dat het gebruik van micro- en nanokristallen voor het bepalen van de structuur niet alleen haalbaar is, maar ook soms voor het gebruik van grote kristallen 5 , 6 , 7 . Deze vooruitgang werd eerst toegepast op de studie van peptiden 8 en natuurlijke kristallen geproduceerd door insectenvirussen 9 , 10 . Ze worden nu gebruikt voor een breed scala van biologische macromoleculen, waaronder de moeilijkste systemen zoals membraanproteïnen en grote complexen 11 . Om de analyse van deze microkristallen te vergemakkelijken, zijn ze geanalyseerd in meso , in het bijzonder membraaneiwitten 12 en in microfluidica chips 13 . De beschikbaarheid van deze nieuwe microkristallografie methodologieën heeft de mogelijkheid geboden om in te gebruikenVivo kristallisatie als een nieuwe route voor structurele biologie 14 , 15 , 16 die een alternatief biedt voor klassieke in vitro kristallogenese. Helaas, zelfs wanneer in vivo kristallen kunnen worden geproduceerd, blijven verschillende obstakels zoals de afbraak of verlies van liganden tijdens de zuivering van cellen, moeilijkheid bij manipulatie en visualisatie van de kristallen bij de synchrotronbundel en vervelende röntgendiffractie-experimenten. Als alternatieve kristallen zijn ook direct in de cel geanalyseerd zonder enige zuiveringstap 17 , 18 , 19 . Een vergelijkende analyse suggereert dat dergelijke in cellulo- benaderingen efficiënter kunnen zijn dan de analyse van gezuiverde kristallen en levert data van hogere resolutie 20 op . Dit protocol is inGeneigd om onderzoekers nieuw te helpen bij microcrystallografie van eiwitten. microcrystallography of protein crystals and in cellulo diffraction

De komst van microfocus van hoge kwaliteit bij veel synchrotronfaciliteiten heeft de routineanalyse van kristallen kleiner dan 10 μm in hun grootste dimensie toegestaan, die een uitdaging vertegenwoordigde. We presenteren twee alternatieve workflows voor de structuurbepaling van eiwitmicrokristallen door röntgenkristallografie met een speciale focus op in vivo gekweekte kristallen. De microkristallen worden ofwel geëxtraheerd uit cellen door sonicatie en gezuiverd door differentiële centrifugatie, of geanalyseerd in cellulose na celsortering door stromingscytometrie van kristalbevattende cellen. Eventueel worden gezuiverde kristallen of kristalbevattende cellen geweekt in zware atoomoplossingen voor experimentele fase. Deze monsters worden vervolgens op diffra- tieve experimenten op soortgelijke wijze bereid door toepassing op een micromesh-ondersteuning en flashkoeling in vloeibare stikstof. We beschrijven en vergelijken serie-diffractie-experimenten van geïsoleerde microkristallen en kristal-Bevattende cellen met behulp van een microfocus synchrotron beamline om datasets te produceren die geschikt zijn voor fasering, modelbouw en verfijning. Deze werkstromen worden geïllustreerd met kristallen van het Bombyx mori cypovirus 1 (BmCPV1) polyhedrine geproduceerd door infectie van insectencellen met een recombinant baculovirus. In deze casestudy is celluloanalyse efficiënter dan analyse van gezuiverde kristallen en levert een structuur in ~ 8 dagen van expressie tot verfijning.

Een overzicht van beide alternatieve methoden voor structuurbepaling met behulp van in vivo microkristallen wordt gepresenteerd ( Figuur 1 ). Polyhedra kan gemakkelijk worden gezuiverd door sonicatie en centrifugatie.
