Het is essentieel om de &quot;stevigheid&quot; van MSC&#39;s te behouden voorafgaand aan de verrijking van neurale voorvaders via hypoxische preconditioning en neurosfeercultuur. Uit onze ervaring kunnen multipotente MSC&#39;s betrouwbaar worden geïdentificeerd door hun langwerpige fibroblastachtige morfologie. In tegenstelling hiermee, MSCs die een meer platgemaakte, vierhoekige morfologie, met prominente cytoskeletale stressvezels hebben aangenomen, nemen niet gemakkelijk neurale celpartijen vast en dienen weggegooid...

De transplantatie van neurale progenitoren en hun derivaten toont belofte als een behandelingsstrategie na traumatische zenuwletsel 1 , 2 en neurodegeneratie 3 , 4 . Voorafgaand aan de klinische toepassing is het essentieel om te verzekeren: i) een methode om toegang te krijgen tot en uit te breiden op een autologe bron van stam- / stamcellen en ii) een middel om hen naar relevante volwassen cellen te brengen 3 . Onze interesse in celtherapie voor ruggenmergbesering heeft ons geleid tot het zoeken naar een robuuste, autologe celbron van neurale progenitoren uit volwassen weefsels. Een subpopulatie van MSC&#39;s komt uit de neurale kam en is gemakkelijk toegankelijk vanuit de mergholte. Deze cellen zijn neurale voorvaders die neuronen en glia 5 kunnen genereren. Diermodellen van cerebrale ischemie tonen aan dat hypoxie de proliferatie bevordert<html> Inferatie en multipotentie van neurale voorlopers in de hersenen 6 . Dit was de basis voor het gebruik van hypoxische pre-conditionering als middel om uit te breiden op merg-afgeleide neurale progenitoren. De transplantatie van Schwann-cellen in het gewonde ruggenmerg bevordert regeneratie 2 . SCLC&#39;s kunnen worden gegenereerd door MSC&#39;s door middel van supplementatie met gliogene factoren ( dwz β-Heregulin, bFGF en PDGF-AA) maar tonen fenotypische instabiliteit. Bij de terugtrekking van groeifactoren keren ze terug naar een fibroblastachtig fenotype 7 . Fenotypische instabiliteit is ongewenst bij celtransplantatie vanwege het risico op afwijkende differentiatie en carcinogenese. Aangezien de Schwann-celprecursoren geassocieerd zijn met axonbundels binnen de embryonale perifere zenuw 8 , werden we geleid tot cocultuur-SCLC&#39;s met gezuiverde embryonale DRG-neuronen 7 ,Ass = &quot;xref&quot;&gt; 9. Resultaat rijpe Schwanncellen zijn lotgevonden en tonen functie in vitro 7 , 9 en in vivo 10 . Ons protocol voor de verrijking van neurale progenitoren van MSC&#39;s is eenvoudig en efficiënt en resulteert in een toename van het celnummer voor latere analyses. De afleiding van door Schotse cellen geformuleerde schadecellen via het coculture platform zorgt voor de studie van glialifferentiatie en voor het genereren van stabiele en functionele Schwann-cellen voor potentiële klinische toepassing.

hypoxic preconditioning of marrow-derived progenitor cells as a source for the generation of mature schwann cells

Dit manuscript beschrijft een middel om te verrijken voor neurale voorliefhebbers van de MSC-populatie en daarna te leiden naar het volwassen Schwann-cellot. Wij ondervonden ratten- en menselijke MSC&#39;s voor transiente hypoxische condities (1% zuurstof gedurende 16 uur), gevolgd door uitbreiding als neurospheren bij substratum met een lage bindingstijd, met supplementatie van de epidermale groeifactor (EGF) / basis fibroblastgroeifactor (bFGF). Neurospheren werden gepolijst op poly-D-lysine / laminine-beklede weefselkweekplastic en gekweekt in een gliogene cocktail die β-Heregulin, bFGF en bloedplaatjes afgeleide groeifactor (PDGF) bevat om Schwann-celachtige cellen te genereren (SCLC&#39;s). SCLC&#39;s werden gedurende 2 weken door middel van cocultuur door middel van cocultuur geregeld met gezuiverde Dorsal root Ganglia (DRG) neuronen verkregen uit E14-15 zwangere Sprague Dawley ratten. Volwassen Schwann cellen tonen aanhoudendheid in S100p / p75 expressie en kunnen myeline segmenten vormen. Cellen die op deze manier zijn gegenereerd hebben potentieel aPplicaties bij autologe celtransplantatie na ruggenmergletsel, evenals bij ziekte-modellering.

Een overzicht van de belangrijkste fasen in ons protocol is geïllustreerd in figuur 1 . Samenvattend worden rat- en menselijke MSC&#39;s geselecteerd door aanhechting aan weefselkweekplastic. Uitgebreide MSC&#39;s zijn voorgeschreven met hypoxie en zijn dan onderworpen aan neurosfeervormende condities. Neurospheren zijn geplateerd en mogen onderscheiden in SCLC&#39;s.
