Die Transplantation von neuronalen Vorläufern und deren Derivaten zeigt Versprechen als Behandlungsstrategie nach traumatischer Nervenverletzung 1 , 2 und Neurodegeneration 3 , 4 . Vor der klinischen Anwendung ist es wichtig, i) eine Methode für den Zugang und die Erweiterung einer autologen Quelle von Stamm- / Vorläuferzellen und ii) ein Mittel, um sie auf relevante, reife Zelltypen zu lenken, zu gewährleisten. Unser Interesse an der Zelltherapie bei Rückenmarksverletzungen führte uns dazu, eine robuste, autologe Zellquelle neuraler Vorläufer aus adulten Geweben zu suchen. Eine Subpopulation von MSCs stammt aus dem Neuralwappen und ist leicht aus der Markhöhle zugänglich. Diese Zellen sind neuronale Vorläufer, die Neuronen und Glia erzeugen können 5 . Tiermodelle der zerebralen Ischämie zeigen, dass Hypoxie die Prol<html> Iferation und multipotenz der neuronalen progenitoren im gehirn 6 Dies war die Grundlage für die Verwendung der hypoxischen Vorkonditionierung als Mittel zur Ausweitung auf margen-abgeleitete neuronale Vorläufer. Die Transplantation von Schwann-Zellen in das verletzte Rückenmark fördert die Regeneration 2 . SCLCs können aus MSCs mittels Supplementierung mit gliogenen Faktoren ( dh β-Heregulin, bFGF und PDGF-AA) erzeugt werden, zeigen aber phänotypische Instabilität. Nach dem Abzug der Wachstumsfaktoren kehren sie zu einem fibroblastenähnlichen Phänotyp 7 zurück . Phänotypische Instabilität ist bei der Zelltransplantation aufgrund des Risikos einer abweichenden Differenzierung und Karzinogenese unerwünscht. Da Schwann-Zellvorläufer mit Axonbündeln innerhalb des embryonalen peripheren Nervs 8 assoziiert sind, wurden wir zu kokulturen SCLCs mit gereinigten embryonalen DRG-Neuronen 7 ,Ass = &quot;xref&quot;&gt; 9 Die resultierenden reifen Schwann-Zellen sind schicksalhaft engagiert und zeigen die Funktion in vitro 7 , 9 und in vivo 10 . Unser Protokoll für die Anreicherung von neuronalen Progenitoren aus MSCs ist einfach und effizient und führt zu einer Erhöhung der Zellzahl für nachfolgende Assays. Die Ableitung von Schicksal-engagierten Schwann-Zellen über die Coculture-Plattform ermöglicht die Untersuchung der Glia-Differenzierung und die Erzeugung von stabilen und funktionellen Schwann-Zellen für eine mögliche klinische Anwendung.

hypoxic preconditioning of marrow-derived progenitor cells as a source for the generation of mature schwann cells

Dieses Manuskript beschreibt ein Mittel, um neuronale Progenitoren aus der Pop-Pop-Pop-Population (MSC) zu bereichern und danach auf das reife Schwann-Zellschicksal zu lenken. Wir unterwarfen Ratten- und menschliche MSCs vorübergehenden hypoxischen Zuständen (1% Sauerstoff für 16 h), gefolgt von einer Expansion als Neurosphären auf Niedrig-Attachment-Substrate mit epidermalem Wachstumsfaktor (EGF) / basischer Fibroblastenwachstumsfaktor (bFGF) -Anpassung. Neurosphären wurden auf Poly-D-Lysin / Laminin-beschichtete Gewebekultur-Plastik ausgesät und in einem gliogenen Cocktail mit β-Heregulin, bFGF und Thrombozyten-Wachstumsfaktor (PDGF) kultiviert, um Schwann-Zell-ähnliche Zellen (SCLCs) zu erzeugen. Die SCLCs wurden für 2 Wochen mit gereinigten Dorsalwurzelganglien (DRG) -Neuronen, die von E14-15-schwangeren Sprague-Dawley-Ratten erhalten wurden, über die Kokultur gerichtet. Reife Schwann-Zellen zeigen die Persistenz bei der S100β / p75-Expression und können Myelinsegmente bilden. Auf diese Weise erzeugte Zellen haben das Potential aAnwendungen in autologen Zelltransplantationen nach Rückenmarksverletzungen sowie bei der Krankheitsmodellierung.

Eine Übersicht über die wichtigsten Stufen in unserem Protokoll ist in Abbildung 1 dargestellt. Zusammenfassend werden Ratten- und menschliche MSCs durch die Einhaltung von Gewebekulturplastik ausgewählt. Expandierte MSCs sind mit Hypoxie vorkonditioniert und unterliegen dann Neurosphären-bildenden Bedingungen. Neurosphären sind plattiert und können in SCLCs differenzieren.
