Il trapianto di progenitori neurali e dei loro derivati ​​dimostra la promessa come strategia di trattamento a seguito di lesioni traumatiche 1 e 2 e neurodegenerazione 3 , 4 . Prima dell&#39;applicazione clinica, è essenziale assicurare: i) un metodo per accedere e espandere ad una fonte autologa di cellule staminali / progenitori e ii) un mezzo per indirizzarli a tipi di cellule mature, rilevanti 3 . Il nostro interesse per la terapia cellulare per lesioni del midollo spinale ci ha portato a cercare una fonte di cellule autologhe robuste e robuste di progenitori neurali dai tessuti adulti. Una sottopopolazione di MSC provengono dalla cresta neurale e sono facilmente accessibili dalla cavità del midollo. Queste cellule sono progenitori neurali che possono generare neuroni e glia 5 . I modelli animali di ischemia cerebrale dimostrano che l&#39;ipossia promuove la prol<html> Iferazione e multipotenza di progenitori neurali all&#39;interno del cervello 6 . L&#39;instabilità fenotipica è indesiderabile nel trapianto cellulare a causa del rischio di differenziazione aberrante e di carcinogenesi. Poiché i precursori di cellule di Schwann sono associati a fasci dell&#39;assone all&#39;interno del nervo periferico embrionale 8 , ci sono stati condotti SCLC di coculture con neuroni embrionali DRG purificati 7 ,Ass = &quot;xref&quot;&gt; 9. Le cellule Schwann mature risultanti sono fate-committed e dimostrano la funzione in vitro 7 , 9 e in vivo 10 . Il nostro protocollo per l&#39;arricchimento dei progenitori neurali da MSC è semplice ed efficiente e porta ad un aumento del numero di cellule per i successivi dosaggi. La derivazione di cellule Schwann fatte tramite destino attraverso la piattaforma di coculture consente lo studio della differenziazione gliale e la generazione di cellule Schwann stabili e funzionali per una possibile applicazione clinica.

Questo manoscritto descrive un mezzo per arricchire i progenitori neurali dalla popolazione delle cellule stromali del midollo (MSC) e quindi di indirizzarli al destino cellulare di Schwann. Abbiamo sottoposto a MSC topi e umani situazioni ipossiche transitorie (ossigeno 1% per 16 h) seguito da espansione come neurosfere su substrato di basso affinamento con fattore di crescita epidermica (EGF) / fattore di crescita fibroblastico base (bFGF). Le neurosfere sono state seminate in plastica di coltura tissutale rivestita con poli-D-lisina e laminina e coltivate in un cocktail gliogenico contenente β-Heregulin, bFGF e fattore di crescita derivato da piastrine (PDGF) per generare cellule simili a cellule Schwann (SCLC). Gli SCLC sono stati indirizzati verso l&#39;impegno di destino mediante cocultura per 2 settimane con neuroni gangliali radici dorsali purificati (DRG) ottenuti da E14-15 ratti incinte Sprague Dawley. Le cellule Schwann mature dimostrano persistenza nell&#39;espressione S100β / p75 e possono formare segmenti di mielina.
