Since the endosymbionts within whiteflies cannot be cultured in vitro, dissection and assembling bacteriocytes is an effective way to obtain enough genetic material of endosymbionts. Before dissection, the species of whitefly and endosymbionts involved should be explicitly confirmed. The whitefly B. tabaci is a species complex with more than 35 morphologically indistinguishable species and different cryptic species may contain different endosymbionts. Bakterier som danner et intimt symbiotisk forhold med relative verter er utbredt i leddyr 1 . Endosymbionter har blitt vist å påvirke aspekter av verter, som næringsmetabolisme, reproduksjon, respons på miljøspenninger 2 , 3 , 4 etc. , i nesten alle utviklingsstadier 5 . Imidlertid er mekanismen som ligger til grunn for foreningene fortsatt stort sett ukjent. Genomics er prioritert og viktig når man studerer de mulige funksjonene og rollene til bakterier. Enkelte grunnleggende opplysninger, det vil si den taxonomiske statusen, funksjonelle gener, metabolismeveier, sekresjonssystemer, kan utledes av genometsekvenser som skjuler lysene på symbionets mulige roller i symbiose. Med utviklingen av høy gjennomstrømningssekvensering har et stort antall bakteriegener blitt sekvensert wMed forskjellige funksjoner avslørt 6 . Endosymbionter er av vital betydning i hemipterans, som bladlus 7 , bedbugs 8 , psyllids 9 , brown planthoppers 10 og cicadas 11 . For eksempel har Buchnera i bladlus, som obligatorisk symbiont, vist seg å være involvert i essensiell aminosyrebiosyntese, sammen med gener fra bladlusgenomet 12 . Videre er transkriptjonell regulering av Buchnera også avslørt 13 . I psyllider blir Carsonella sekvensert og rangert som det minste bakteriegenomet som noensinne er funnet 14 . Alle disse kjennetegnene for endosymbionter er basert og utledes fra genometsekvensene. Fordi disse endosymbionter ikke kan dyrkes in vitro , har flere tilnærminger blitt anvendt for å isolere tilstrekkelige bakterier for sequencing. I bladlus ekstraheres endosymbioner gjennom sentrifugering og filtrering og underkastes videre genomisk og transkriptomisk analyse 5 . I brune plantehoppere sekvenseres endosymbionter sammen med hele insektsgenomet 10 . Whitefly B. tabaci er et artskompleks som inneholder mer enn 35 morfologisk uoppdagelige arter (kryptiske arter), blant hvilke to invasive arter har invadert over hele verden og forårsaket enorm skade på landbruksproduksjonen 15 . Av oppmerksomhet har endosymbioner innenfor B. tabaci- arter vist betydning i utviklingen av skadedyrene 16 . Til nå er det identifisert åtte endosymbionter i whiteflyen, inkludert den obligatoriske symbionten, Candidatus Portiera aleyrodidarum og syv sekundære symbionter Hamiltonella , Rickettsia , Arsenophonus , Cardinium , &lt;Em&gt; Wolbachia, Fritschea og Hemipteriphilus definert 17 , 18 . I motsetning til hemipteransene beskrevet tidligere, er whitefly B. tabaci et ekstremt lite insekt med en lengde på 1 mm. De fleste endosymbionter er begrenset til bakteriocytter 19 (spesialiserte celler som inneholder symbionter, som ytterligere danner bakteriom i B. tabaci ). I tillegg kan disse endosymbionene ikke dyrkes in vitro . Den eneste måten å skaffe endosymbionter fra B. tabaci er å dissekere bakterien ut. Det er imidlertid problemer med disseksjonen. For det første kobler den skjøre bakterien alltid med andre vev av whitefly, som er vanskelig å skille fra. For det andre begrenser den lille størrelsen på whitefly isolasjonen av nok bakteriom. Bakterie symbionter danner et intimt forhold til sine verter og gir i de fleste tilfeller fordeler til vertene. Genomisk informasjon er kritisk for å studere funksjonene og utviklingen av bakterielle symbionter i verten. Ettersom de fleste symbionter ikke kan dyrkes in vitro , er metoder for å isolere en tilstrekkelig mengde bakterier for genom-sekvensering svært viktige. I Whitefly Bemisia tabaci har en rekke endosymbioner blitt identifisert og antas å være av betydning for utvikling og reproduksjon av skadedyrene gjennom flere tilnærminger. Imidlertid forblir mekanismen som ligger til grunn for foreningene stort sett ukjente. Hindringen kommer delvis fra det faktum at endosymbionter i whitefly, mest forstyrret i bakteriocytter, er vanskelig å skille fra vertscellene. Her rapporterer vi en trinnvis protokoll for identifisering, ekstraksjon og rensing av endosymbionter fra whitefly B. tabaci hovedsakelig ved dissektiPå og filtrering. Endosymbiont-prøver fremstilt ved denne fremgangsmåte, selv om de fortsatt er en blanding av forskjellige endosymbiont-arter, er egnede for etterfølgende genom-sekvensering og analyse av de mulige roller av endosymbionter i B. tabaci . Denne metoden kan også brukes til å isolere endosymbioner fra andre insekter.
