Since the endosymbionts within whiteflies cannot be cultured in vitro, dissection and assembling bacteriocytes is an effective way to obtain enough genetic material of endosymbionts. Bakterier som bildar ett intimt symbiotiskt förhållande med relativa värdar är utbredd i arthropods 1 . Endosymbionterna har visat sig påverka värden hos värdar, såsom näringsmetabolism, reproduktion, respons på miljöbelastningar 2 , 3 , 4 etc. , i nästan alla utvecklingsstadier 5 . Mekanismen som ligger bakom föreningarna är dock fortfarande i stort sett okänd. Genomik har prioritet och betydelse när man studerar potentiella funktioner och roller hos bakterier. Vissa grundläggande uppgifter, det vill säga den taxonomiska statusen, funktionella gener, metabolismvägarna, sekretionssystemen, kan härledas från genom-sekvenser, vilket lyser på symbionternas möjliga roller i symbios. Med utvecklingen av hög genomströmningssekvensering har ett stort antal bakteriegener sekvenserats wMed olika funktioner avslöjade 6 . Endosymbionter är av vital betydelse för hemipteraner, såsom bladlöss 7 , bedbugs 8 , psyllids 9 , bruna planthoppers 10 och cicadas 11 . Till exempel har Buchnera i bladlus , som obligatorisk symbiont, visat sig vara involverad i essentiell aminosyrorbiosyntes, tillsammans med generna från bladlusgenomet 12 . Vidare avslöjas transkriptionsreglering av Buchnera 13 . I psyllider sekvenseras Carsonella och rankas det minsta bakteriegenomet som någonsin hittats 14 . Alla dessa kännetecken för endosymbioner är baserade och härledda från genometsekvenserna. Eftersom dessa endosymbioner inte kan odlas in vitro har flera sätt applicerats för att isolera adekvata bakterier för sequencing. I bladlus extraheras endosymbioner genom centrifugering och filtrering och utsätts för ytterligare genomisk och transkriptomisk analys 5 . I bruna planthoppare sekvenseras endosymbioner tillsammans med hela insektsgenomet 10 . Whitefly B. tabaci är ett artskomplex som innehåller mer än 35 morfologiskt oskiljbara arter (kryptiska arter), bland vilka två invasiva arter har invaderat över hela världen och orsakat enorma skador på jordbruksproduktionen 15 . Av anmärkning har endosymbioner inom B. tabaci- arter visat betydelse vid utvecklingen av skadegörarna 16 . Hittills har åtta endosymbioner identifierats i whiteflyen, inklusive den obligatoriska symbionten, Candidatus Portiera aleyrodidarum och sju sekundära symbionter Hamiltonella , Rickettsia , Arsenophonus , Cardinium , &lt;Em&gt; Wolbachia, Fritschea och Hemipteriphilus definierade 17 , 18 . Till skillnad från de tidigare beskrivna hemiptererna är vitflödet B. tabaci en extremt liten insekt med en längd på endast 1 mm. De flesta endosymbioner är begränsade till bakteriocyter 19 (specialiserade celler innehållande symbionter, vilka vidare bildar bakteriom i B. tabaci ). Dessutom kan dessa endosymbioner inte odlas in vitro . Det enda sättet att erhålla endosymbionter från B. tabaci är att dissekera bakterien ut. Det finns dock svårigheter i dissektionen. För det första kopplar den bräckliga bakterien alltid med andra vävnader av vitflugan, som är svår att separera. För det andra begränsar den lilla storleken av whitefly isoleringen av tillräckligt med bakterier. För det tredje klarar endosymbionter i bakterien, vilket gör det ytterst komplicerat att förvärva en enda bakterieart. <p clAss = &quot;jove_content&quot;&gt; Här rapporterar vi ett enkelt och billigt protokoll för att isolera whitefly endosymbionter för efterföljande metagenom-sekvensering. Bakterie symbionter bildar ett intimt förhållande med sina värdar och ger i högsta grad fördelar till värdarna. Genomisk information är kritisk för att studera funktioner och utveckling av bakteriella symbioner i deras värd. Eftersom de flesta symbionter inte kan odlas in vitro är metoder för att isolera en adekvat mängd bakterier för genom-sekvensering väldigt viktiga. I Whitefly Bemisia tabaci har ett antal endosymbioner identifierats och förutspås vara av betydelse vid utveckling och reproduktion av skadedjur genom flera tillvägagångssätt. Mekanismen som ligger bakom föreningarna förblir dock i stort sett okänd. Hindern beror delvis på det faktum att endosymbionterna i whitefly, mestadels begränsad i bakteriocyter, är svåra att separera från värdcellerna. Här rapporterar vi ett steg för steg protokoll för identifiering, extraktion och rening av endosymbioner från whitefly B. tabaci huvudsakligen genom dissektiPå och filtrering. Endosymbiont-prov framställda med denna metod, även om det fortfarande är en blandning av olika endosymbiont-arter, är lämpliga för efterföljande genom-sekvensering och analys av de möjliga rollerna hos endosymbioner i B. tabaci . Denna metod kan också användas för att isolera endosymbioner från andra insekter.
