Las modalidades ópticas de imágenes de alta resolución, como la tomografía de coherencia óptica, la microscopía confocal y la microscopía multifotónica, tienen numerosos beneficios. Sin embargo, la resolución espacial disminuye significativamente a medida que aumenta la profundidad de formación de imágenes. Esto se debe a la naturaleza difusa del transporte ligero en tejidos blandos 1 , 2 . La integración de la excitación óptica y la detección de ultrasonidos proporciona una solución para superar el reto de la imagen óptica de alta resolución en tejidos profundos. La microscopía fotoacústica (PAM) es una modalidad de este tipo que puede proporcionar imágenes más profundas que otras modalidades de imagen óptica. Se ha aplicado con éxito a imágenes estructurales, funcionales, moleculares y de células in vivo 3 , 4 , 5 , 6 , 7 , 8 </sup&gt; , 9 , 10 , 11 , 12 , 13 combinando el fuerte contraste de absorción óptica con la alta resolución espacial del ultrasonido. En PAM, un pulso láser corto irradia el tejido / muestra. La absorción de luz por los cromóforos ( por ejemplo, melanina, hemoglobina, agua, etc. ) produce un aumento de la temperatura, que a su vez produce la producción de ondas de presión en forma de ondas acústicas (ondas fotoacústicas). Las ondas fotoacústicas generadas pueden ser detectadas por un transductor ultrasónico de banda ancha fuera del límite del tejido. Utilizando óptica débil y acústica de enfoque apretado, imágenes de tejidos profundos se puede lograr en la microscopía fotoacústica de resolución acústica (AR-PAM) [ 14 , 15 , 16] . En ar-PAM, se ha demostrado una resolución lateral de 45 μm y una profundidad de imagen de hasta 3 mm 15 . Con el fin de resolver acústicamente capilares individuales (~ 5 μm), se requieren transductores ultrasónicos que funcionen a frecuencias centrales de&gt; 400 MHz. A tales frecuencias altas, la profundidad de penetración es inferior a 100 μm. El problema causado por el enfoque acústico apretado puede ser resuelto usando el enfoque óptico apretado. La microscopía fotoacústica de resolución óptica (OR-PAM) es capaz de resolver capilares únicos, o incluso una sola célula 17 , y una resolución lateral de 0,5 μm se ha logrado 18 , 19 , 20 , 21 , 22 , 23 , 24 . El uso de un nanojet fotónico puede ayudar a lograr una resolución más allá de la resolución limitada por difracciónN 25 , 26 . En OR-PAM, la profundidad de penetración es limitada debido al enfoque de la luz, y puede imagen hasta ~ 1,2 mm dentro del tejido biológico [ 23] . Por lo tanto, AR-PAM puede imagen más profunda, pero con una resolución más baja, y OR-PAM puede imagen con una resolución muy alta, pero con una profundidad de imagen limitada. La velocidad de formación de imágenes del sistema AR y OR-PAM depende principalmente de la velocidad de repetición del impulso de la fuente láser 27 . La combinación de AR-PAM y OR-PAM será de gran beneficio para las aplicaciones que requieren una alta resolución y una imagen más profunda. Se ha hecho poco esfuerzo para combinar estos sistemas. Por lo general, se utilizan dos escáneres de imágenes diferentes para la formación de imágenes, lo que requiere que la muestra se mueva entre ambos sistemas, lo que dificulta la realización de imágenes in vivo . Sin embargo, la imagen híbrida con AR y OR PAM permite la creación de imágenes con resoluciones escalables aNd profundidades. En un enfoque, se utiliza un haz de fibras ópticas para suministrar luz tanto para AR como para OR PAM. En este enfoque, se utilizan dos láseres separados (un láser de alta energía a 570 nm para el AR y un láser de baja energía y alta frecuencia de repetición a 532 nm para el OR), lo que hace que el sistema resulte inconveniente y caro 28 . La longitud de onda del láser OR-PAM es fija, y muchos estudios, como la saturación de oxígeno, no son posibles utilizando este sistema combinado. Los estudios comparativos entre AR y OR PAM tampoco son posibles debido a la diferencia en las longitudes de onda del láser entre AR y OR. Además, AR-PAM utiliza iluminación de campo brillante; Por lo tanto, las señales fotoacústicas fuertes de la superficie de la piel limitan la calidad de la imagen. Por esta razón, el sistema no puede utilizarse para muchas aplicaciones de bioimagen. En otro enfoque para realizar AR y OR PAM, el enfoque óptico y ultrasónico se desplaza, lo que hace que el enfoque de la luz y el enfoque ultrasónico no estén alineados. Utilizando esta técnica, el AR-PAM y el OR-PAM pueden lograr resoluciones de sólo 139 μm y 21 μm, respectivamente, lo que lo convierte en un sistema de baja resolución. En todos estos casos, AR-PAM no utilizó iluminación de campo oscuro. El uso de la iluminación de campo oscuro puede reducir la generación de señales fotoacústicas fuertes de la superficie de la piel. switchable acoustic and optical resolution photoacoustic microscopy for in vivo small-animal blood vasculature imaging

La microscopía fotoacústica (PAM) es una modalidad de imagen in vivo de rápido crecimiento que combina óptica y ultrasonido, proporcionando una penetración más allá de la trayectoria libre media óptica (~ 1 mm en la piel) con alta resolución. Al combinar el contraste de absorción óptica con la alta resolución espacial del ultrasonido en una sola modalidad, esta técnica puede penetrar tejidos profundos. Los sistemas de microscopía fotoacústica pueden tener una baja resolución acústica y una sonda profunda o una alta resolución óptica y una sonda poco profunda. Es difícil lograr una alta resolución espacial y una gran penetración en profundidad con un único sistema. Este trabajo presenta un sistema AR-OR-PAM capaz tanto de imágenes de alta resolución a profundidades poco profundas como de imágenes de tejido profundo de baja resolución de la misma muestra in vivo . Una resolución lateral de 4 μm con una profundidad de imagen de 1,4 mm utilizando enfoque óptico y una resolución lateral de 45 μm con una profundidad de imagen de 7,8 mm utilizando enfoque acústico fueron exitosasDemostrado utilizando el sistema combinado. En este caso, se realiza una proyección de imagen vascular de sangre de animal pequeño in vivo para demostrar su capacidad de formación de imágenes biológicas.

El esquema del sistema AR-OR-PAM se muestra en la Figura 1 . En esta configuración, todos los componentes se integraron y montaron en una configuración de jaula óptica. El uso de un sistema de jaula hace que la cabeza de exploración AR-OR-PAM sea compacta y fácilmente ensamblada, alineada e integrada en una única etapa de escaneado.
