<html> Photoacoustic Tomography (PAT) is een hybrid imaging modaliteit die veel toepassingen heeft in zowel klinische als preklinische studies 1 , 2 , 3 , 4 , 5 . In PAT bestralen nanoseconde laserpulsen biologisch weefsel. De absorptie van incidentielicht door de weefselchromoforen leidt tot een lokale temperatuurstijging, die dan drukgolven uitzendt in de vorm van geluidsgolven. Een ultrasone detector verzamelt de foto-akoestische signalen op verschillende posities rond het monster. De foto-akoestische (PA) signalen worden gereconstrueerd met behulp van verschillende algoritmes (zoals een vertragings- en som-algoritme) 6 om het foto-akoestische beeld te genereren. Deze hybrid imaging modaliteit biedt hoge resolutie, diepgeweven beeldvorming en hoge optische absorptie contrast 7 ,Class = &quot;xref&quot;&gt; 8. Onlangs is een beeldvormende diepte van 12 cm bereikt in kippenborstweefsel met behulp van een langere golflengte (~ 1.064 nm) en een exogeen contrastmiddel, die fosforfthalocyanine bevat. Deze diepte gevoeligheid is veel hoger dan de diepte gevoeligheid van andere optische methoden, zoals confocal fluorescentiemicroscopie, twee-foton fluorescentiemicroscopie, 10 optische coherentietomografie, 11 enz. Met meer dan één golflengte kan PAT structurele en functionele veranderingen in organen aantonen . Voor veel mensenziekten zijn kleine dierenmodellen goed gevestigd 12 , 13 , 14 , 15 . Voor het afbeelden van kleine dieren zijn diverse modaliteiten aangetoond. Uit al die benaderingen heeft PA imaging eerder aandacht gekregen vanwege de bovengenoemde voordelen. VADERT heeft zijn potentieel voor het beelden van bloedvaten in de weefsels en organen ( dwz hart, longen, lever, ogen, milt, hersenen, huid, ruggengraat, nier, enz. ) Van kleine dieren 4 , 16 , 17 , 18 weergegeven . PAT is een gevestigde modaliteit voor beeldvorming van kleine dieren. PA-golven worden geproduceerd door de lichtabsorptie door de chromophoren, zodat meerdere golflengten PAT de mapping van de totale hemoglobine concentratie (HbT) en zuurstofverzadiging (SO2) 19 , 20 , 21 , 22 mogelijk maken . Brein-neurovasculaire beeldvorming werd bereikt met behulp van exogene contrastmiddelen 12 , 23 , 24 . PA modaliteit kan bijdragen tot een beter begrip van de gezondheid van de hersenen doorInformatie verstrekken op de moleculaire en genetische niveaus. Voor beeldvorming met kleine dieren worden Nd: YAG / OPO lasers veel gebruikt als PAT excitatie bronnen. Deze lasers leveren ~ 5 ns bijna-infraroodpulsen met energie (~ 100 mJ bij het OPO-uitvoervenster) bij een herhalingssnelheid van 10 Hz. Het PA-systeem uitgerust met dergelijke lasers is kostbaar en omvangrijk en maakt het mogelijk om met lage-snelheid beeldvorming met een-element-ultrageluid transducers (UST) te maken vanwege de lage herhalingssnelheid van de laserbron. Een typische A-lijn aanwinstijd in dergelijke PA-systemen is ~ 5 min per doorsnede 25 . Een beeldvorming systeem met zo&#39;n lange metingstijd is niet ideaal voor beeldvorming met kleine dieren, omdat het moeilijk is om de fysiologische parameters te controleren voor beeldvorming in het hele lichaam, tijdoplossend functioneel beeldvorming, enz. Door meerdere single-element USTs te selecteren, Gebaseerde UST&#39;s, of een laser met hoge repetitiesnelheid, is het mogelijk om de beeldsnelheid van PA te verhogensystemen. Door slechts één single-element UST te gebruiken om alle PA signalen te verzamelen rond het monster, wordt de beeldsnelheid van het systeem beperkt. Meerdere single-element UST&#39;s die in cirkelvormige of halfcirkelvormige geometrie worden geregeld, worden aangetoond voor high-speed, zeer gevoelige beeldvormingstechnieken. Array-gebaseerde USTs 26 , zoals lineaire, halve cirkelvormige, cirkelvormige en volumetrische arrays, zijn succesvol gebruikt voor real-time beeldvorming 1 . Deze array-gebaseerde UST&#39;s zullen de beeldsnelheidsnelheid verhogen en de meetgevoeligheid verminderen, maar ze zijn duur. Echter, de beeldsnelheid van PA systemen die gebruik maken van array-gebaseerde UST&#39;s is nog steeds beperkt door de herhalingssnelheid van de laser. Pulsed-laser technologie is geavanceerd om hoge-repetitie-rate pulserende-laser diodes (PLD&#39;s) te maken. 7000 frames / s B-scan foto-akoestische beeldvorming werd aangetoond met PLD&#39;s met behulp van een klinisch ultrasound platform 27 . Dergelijke PLD&#39;s kunnen de afbeeldingssnelheid van th verbeterenE PAT-systeem, zelfs bij single-element UST circulaire scan geometrie. Single-element UST&#39;s zijn minder duur en zeer gevoelig, in tegenstelling tot array-gebaseerde UST&#39;s. In het afgelopen decennium werd weinig onderzoek gerapporteerd over het gebruik van PLD&#39;s met hoge herhalingsgraad als de excitatiebron voor PA-beeldvorming. Een vezelgebaseerde bijna-infrarood PLD werd aangetoond voor PA-beeldvorming van fantoom 28 . De in vivo beeldvorming van bloedvaten bij een ~ 1 mm diepte onder de menselijke huid werd aangetoond met behulp van energiezuinige PLD&#39;s 29 . Een PLD-gebaseerde optische resolutie foto-akoestische microscoop (ORPAM) werd gemeld. Met behulp van PLD&#39;s werd ~ 1,5 cm diepbeeldvorming bij een beeldsnelheid van 0,43 Hz aangetoond 30 . Zeer recent werd een PLD-PAT-systeem gemeld dat beelden op zo kort als ~ 3 s en op een ~ 2 cm beelddiepte in biologisch weefsel 25 , 31 leverden. Deze studie bleek dat zo&#39;n goedkoop, compact systeem hoge qua kan leverenLity beelden, zelfs bij hoge snelheden. Het PLD-PAT-systeem kan worden gebruikt voor fotocoustic imaging met hoge beeldsnelheid (7000 fps), beeldschermbeelden op het bloed, vingergewichtbeelden, 2 cm diepe weefselbeelden, beeldvorming met kleine dieren, enz. De enkelvoudige golflengte en Low pulse-energy pulsen van PLD beperken de toepassing ervan tot multispectrale en diepe weefselbeelden. Er zijn experimenten uitgevoerd op kleine dieren met hetzelfde PLD-PAT systeem dat wordt gebruikt voor preklinische toepassingen. Het doel van dit werk is het visualiseren van de experimentele demonstratie van het PLD-PAT-systeem voor in-vivo 2D doorsnedebeelden van kleine dieren.

a high-performance compact photoacoustic tomography system for in vivo small-animal brain imaging

In vivo speelt kleine dierenbeelden een belangrijke rol in preklinische studies. Photoacoustic tomography (PAT) is een opkomende hybrid imaging modaliteit die groot potentieel voor zowel preklinische als klinische toepassingen aantoont. Conventionele optische parametrische oscillator-gebaseerde PAT (OPO-PAT) systemen zijn omvangrijk en duur en kunnen geen snelle beeldvorming bieden. Onlangs zijn pulsed-laser diodes (PLD&#39;s) succesvol aangetoond als een alternatieve excitatiebron voor PAT. Pulsed-laser diode PAT (PLD-PAT) is succesvol aangetoond voor high-speed beeldvorming op photoacoustic phantoms en biologische weefsels. Dit werk biedt een visualized experimenteel protocol voor in vivo hersenbeeldvorming met behulp van PLD-PAT. Het protocol bevat de compacte PLD-PAT-systeemconfiguratie en de beschrijving ervan, dierbereiding voor hersenbeeldvorming en een typische experimentele procedure voor 2D doorsnede-beeldvorming van de rattenhuid. Het PLD-PAT-systeem is compact en kost-efFectief en kan high-speed beeldvorming van hoge kwaliteit leveren. Brainbeelden die in vivo bij verschillende scansnelheden worden verzameld, worden gepresenteerd.

De in vivo hersenbeeldresultaten die de mogelijkheden van het beschreven PLD-PAT-systeem tonen, worden in deze sectie getoond. Om de high-speed beeldvormingsmogelijkheden van het PLD-PAT-systeem te demonstreren, werd de in vivo hersenafbeelding van twee verschillende gezonde ratten uitgevoerd.
