<html> La tomographie photoacoustique (PAT) est une modalité d&#39;imagerie hybride qui a de nombreuses applications dans les études cliniques et précliniques 1 , 2 , 3 , 4 , 5 . En PAT, les impulsions laser à la nanoseconde irradient le tissu biologique. L&#39;absorption de la lumière incidente par les chromophores des tissus conduit à une élévation de la température locale, qui donne alors des ondes de pression émises sous forme d&#39;ondes sonores. Un détecteur d&#39;ultrasons collecte les signaux photoacoustiques à diverses positions autour de l&#39;échantillon. Les signaux photoacoustiques (PA) sont reconstruits en utilisant différents algorithmes (comme un algorithme de retard et de somme) 6 pour générer l&#39;image photoacoustique. Cette modalité d&#39;imagerie hybride offre une image haute résolution et des tissus profonds et un contraste d&#39;absorption optique élevé 7 ,Class = &quot;xref&quot;&gt; 8. Récemment, une profondeur d&#39;imagerie de ~ 12 cm 9 a été obtenue dans le tissu de poitrine de poulet à l&#39;aide d&#39;une longueur d&#39;onde plus longue (~ 1 064 nm) et d&#39;un agent de contraste exogène appelé phosphate de phtalocyanine. Cette sensibilité à la profondeur est beaucoup plus élevée que la sensibilité de profondeur d&#39;autres méthodes optiques, telles que la microscopie de fluorescence confocale, la microscopie à fluorescence à deux photons, la tomographie par cohérence optique 10 , 11, etc. En utilisant plus d&#39;une longueur d&#39;onde, PAT peut démontrer des changements structurels et fonctionnels dans les organes . Pour de nombreuses maladies humaines, les modèles de petits animaux ont été bien établis 12 , 13 , 14 , 15 . Pour l&#39;imagerie des petits animaux, plusieurs modalités ont été démontrées. Sur toutes ces approches, l&#39;imagerie PA a rapidement attiré l&#39;attention en raison des avantages mentionnés ci-dessus. PennsylvanieT a montré son potentiel pour imager les vaisseaux sanguins dans les tissus et les organes ( c.-à-d., Le cœur, les poumons, le foie, les yeux, la rate, le cerveau, la peau, la moelle épinière, les reins, etc. ) des petits animaux 4 , 16 , 17 , 18 . PAT est une modalité bien établie pour l&#39;imagerie cérébrale des petits animaux. Les ondes PA sont produites en raison de l&#39;absorption de la lumière par les chromophores, de sorte que le PAT à longueur d&#39;onde multiple permet la cartographie de la concentration totale d&#39;hémoglobine (HbT) et de la saturation en oxygène (SO 2 ) 19 , 20 , 21 , 22 . L&#39;imagerie neurovasculaire du cerveau a été réalisée à l&#39;aide d&#39;agents de contraste exogènes 12 , 23 , 24 . La modalité de PA peut aider à mieux comprendre la santé du cerveau parFournissant des informations aux niveaux moléculaire et génétique. Pour l&#39;imagerie des petits animaux, les lasers Nd: YAG / OPO sont largement utilisés comme sources d&#39;excitation PAT. Ces lasers fournissent des impulsions infrarouges proches de ~ 5 ns avec de l&#39;énergie (~ 100 mJ à la fenêtre de sortie OPO) à un taux de répétition de ~ 10 Hz 25 . Le système de PA équipé de ces lasers est coûteux et volumineux et permet l&#39;imagerie à basse vitesse avec des transducteurs à ultrasons à un seul élément (UST) en raison du faible taux de répétition de la source laser. Un temps typique d&#39;acquisition de ligne A dans de tels systèmes de PA est de ~ 5 min par section transversale 25 . Un système d&#39;imagerie avec un temps de mesure si long n&#39;est pas idéal pour l&#39;imagerie des petits animaux, car il est difficile de contrôler les paramètres physiologiques pour l&#39;imagerie corporelle complète, l&#39;imagerie fonctionnelle résolue dans le temps, etc. En adoptant plusieurs UST à un seul élément, Ou un laser à haute répétition, il est possible d&#39;augmenter la vitesse d&#39;imagerie de PASystèmes. L&#39;utilisation d&#39;un seul UST à un seul élément pour collecter tous les signaux PA autour de l&#39;échantillon limitera la vitesse d&#39;imagerie du système. Plusieurs UST à un seul élément disposés en géométrie circulaire ou semi-circulaire sont démontrés pour des techniques d&#39;imagerie haute vitesse et très sensibles. Les UST 26 basés sur les tableaux, tels que les tableaux linéaires, semi-circulaires, circulaires et volumétriques, ont été utilisés avec succès pour l&#39;imagerie en temps réel 1 . Ces UST basés sur le tableau augmentent la vitesse d&#39;imagerie et réduisent la sensibilité à la mesure, mais ils coûtent cher. Cependant, la vitesse d&#39;imagerie des systèmes PA qui utilisent des UST basées sur le tableau est encore limitée par le taux de répétition du laser. La technologie laser pulsée est avancée pour produire des diodes laser pulsées à haute répétition (PLD). 7 000 images / s L&#39;imagerie photoacoustique B-scan a été démontrée avec des PLD utilisant une plate-forme d&#39;échographie clinique 27 . De tels PLD peuvent améliorer la vitesse d&#39;imagerie deE système PAT, même avec une géométrie de balayage circulaire UST à un seul élément. Les UST à un élément sont moins coûteux et très sensibles, contrairement aux UST basés sur les tableaux. Au cours de la dernière décennie, peu de recherches ont été rapportées sur l&#39;utilisation de PLD à taux de répétition élevé comme source d&#39;excitation pour l&#39;imagerie PA. Un PLD à proximité d&#39;infrarouge à base de fibres a été démontré pour l&#39;imagerie PA de fantômes 28 . L&#39;imagerie in vivo des vaisseaux sanguins à une profondeur de ~ 1 mm au-dessous de la peau humaine a été démontrée en utilisant des PLD à faible énergie 29 . Un microscope photoacoustique à résolution optique PLD (ORPAM) a été signalé. En utilisant des PLD, une image profonde de ~ 1,5 cm à une cadence de 0,43 Hz a été démontrée 30 . Très récemment, un système PLD-PAT a été signalé qui a fourni des images aussi courtes que ~ 3 s et à une profondeur d&#39;imagerie de ~ 2 cm dans un tissu biologique 25 , 31 . Cette étude a prouvé qu&#39;un tel système compact et peu coûteux peut fournir une qualité élevéeLite images, même à grande vitesse. Le système PLD-PAT peut être utilisé pour l&#39;imagerie photoacoustique à haute cadence (7 000 images par seconde), l&#39;imagerie des vaisseaux sanguins, l&#39;imagerie des doigts, l&#39;imagerie tissulaire de 2 cm de profondeur, l&#39;imagerie cérébrale des petits animaux, etc. La longueur d&#39;onde unique et Les impulsions à faible impulsion d&#39;énergie de PLD limitent son application à l&#39;imagerie multi-spectrale et à tissus profonds. Des expériences ont été réalisées sur de petits animaux utilisant le même système PLD-PAT utilisé pour des applications précliniques. Le but de ce travail est de fournir la démonstration expérimentale expérimentée du système PLD-PAT pour l&#39;imagerie cérébrale en 2D in vivo in vivo de petits animaux.

a high-performance compact photoacoustic tomography system for in vivo small-animal brain imaging

L&#39; imagerie animale chez les petits animaux a un rôle important à jouer dans les études précliniques. La tomographie photoacoustique (PAT) est une modalité d&#39;imagerie hybride émergente qui présente un grand potentiel pour les applications précliniques et cliniques. Les systèmes PAT (OPO-PAT) basés sur les oscillateurs paramétriques optiques classiques sont volumineux et coûteux et ne peuvent pas fournir une image haute vitesse. Récemment, les diodes à laser pulsé (PLD) ont été démontrées avec succès comme une source d&#39;excitation alternative pour PAT. La diode laser à impulsions PAT (PLD-PAT) a été démontrée avec succès pour l&#39;imagerie à grande vitesse sur des fantômes photoacoustiques et des tissus biologiques. Ce travail fournit un protocole expérimental visualisé pour l&#39;imagerie cérébrale in vivo en utilisant PLD-PAT. Le protocole comprend la configuration compacte du système PLD-PAT et sa description, la préparation des animaux pour l&#39;imagerie cérébrale et une procédure expérimentale typique pour l&#39;imagerie cérébrale de rat à 2D en coupe transversale. Le système PLD-PAT est compact et coûteuxEfficace et peut fournir une image haute vitesse et de haute qualité. Les images du cerveau recueillies in vivo à différentes vitesses de balayage sont présentées.

Les résultats d&#39;imagerie cérébrale in vivo qui démontrent les capacités du système PLD-PAT décrit sont présentés dans cette section. Pour démontrer les capacités d&#39;imagerie à grande vitesse du système PLD-PAT, l&#39;imagerie cérébrale in vivo de deux rats sains différents a été réalisée.
