<html> Fotoakustisk tomografi (PAT) er en hybridbildingsmodalitet som har mange anvendelser i både kliniske og prekliniske studier 1 , 2 , 3 , 4 , 5 . I PAT bestråler nanosekund laserpulser biologisk vev. Opptaket av innfallslampe av vevskromophorene fører til en lokal temperaturstigning, som deretter gir trykkbølger utstrålet i form av lydbølger. En ultralydsdetektor samler de fotoakustiske signaler på forskjellige stillinger rundt prøven. De bildeakustiske signalene (PA) blir rekonstruert ved hjelp av ulike algoritmer (for eksempel en forsinkelses- og sum-algoritme) 6 for å generere bildeakustisk bilde. Denne hybridbildingsmodaliteten gir høyoppløselig, dypvevsbilding og høy optisk absorpsjonskontrast 7 ,Class = &quot;xref&quot;&gt; 8. Nylig ble det oppnådd en ~ 12 cm billeddybde 9 i kyllingbrystvev ved hjelp av en lengre bølgelengde (~ 1.064 nm) og et eksogent kontrastmiddel kisfosfftalocyanin. Denne dybdefølsomheten er mye høyere enn dybdekänsligheten til andre optiske metoder, for eksempel konfokal fluorescensmikroskopi, tofoton fluorescensmikroskopi, 10 optisk koherensomografi, 11 etc. Ved bruk av mer enn en bølgelengde kan PAT demonstrere strukturelle og funksjonelle forandringer i organer . For mange menneskelige sykdommer har små dyrmodeller vært veletablert 12 , 13 , 14 , 15 . For avbildning av små dyr er det vist flere modaliteter. Av alle disse tilnærmingene har PA bildebehandling fått oppmerksomhet ganske raskt på grunn av fordelene nevnt ovenfor. PAT har vist sitt potensial for å avdekke blodkar i vev og organer ( dvs. hjerte, lunger, lever, øyne, milt, hjerne, hud, ryggmargen, nyre, etc. ) av små dyr 4 , 16 , 17 , 18 . PAT er en veletablert modalitet for små-dyr hjernedimensjonering. PA-bølger er produsert på grunn av lysabsorbsjonen av kromoforene, slik at flerbølgelengde PAT tillater kartlegging av total hemoglobinkonsentrasjon (HbT) og oksygenmetning (SO 2 ) 19 , 20 , 21 , 22 . Hjerne-neurovaskulær avbildning ble oppnådd ved hjelp av eksogene kontrastmidler 12 , 23 , 24 . PA-modalitet kan bidra til å gi en bedre forståelse av hjernens helse vedGi informasjon på molekylære og genetiske nivåer. For små-dyr avbildning, er Nd: YAG / OPO lasere mye brukt som PAT excitasjonskilder. Disse lasere leverer ~ 5 ns nær-infrarøde pulser med energi (~ 100 mJ ved OPO-utgangsvinduet) ved en 10 Hz-repetisjonshastighet 25 . PA-systemet utstyrt med slike lasere er kostbart og omfangsrikt og gir mulighet for lavhastighets bildebehandling med enkeltelement-ultralydtransdusere (UST) på grunn av den lave repetisjonshastigheten til laserkilden. En typisk A-line-oppkjøpstid i slike PA-systemer er ~ 5 min per tverrsnitt 25 . Et bildebehandlingssystem med en så lang målingstid er ikke ideell for bildebehandling av små dyr, fordi det er vanskelig å kontrollere de fysiologiske parametrene for fullkroppsbilding, tidsoppløst funksjonell bildebehandling, etc. Ved å vedta flere enkeltelement-UST-er, array Baserte UST-er, eller en laser med høy repetisjonshastighet, er det mulig å øke bildehastigheten til PAsystemer. Ved å bruke bare ett enkeltelement UST til å samle alle PA-signaler rundt prøven, begrenser bildehastigheten til systemet. Flere single-element USTs arrangert i sirkulær eller halvcirkelformet geometri er demonstrert for høyhastighets, svært følsomme bildebehandlingsteknikker. Array-baserte USTs 26 , som for eksempel lineære, halv-sirkulære, sirkulære og volumetriske arrays, har blitt brukt for real-time avbildning 1 . Disse array-baserte UST-ene vil øke bildehastigheten og redusere målfølsomheten, men de er dyre. Imaginghastigheten til PA-systemer som bruker array-baserte UST-er er imidlertid fortsatt begrenset av laserens repetisjonshastighet. Pulserende laserteknologi avansert for å lage pulserende laserdioder (PLD) med høy repetisjon. 7.000 rammer / s B-skanning fotoakustisk avbildning ble demonstrert med PLDer ved hjelp av en klinisk ultralydplattform 27 . Slike PLDer kan forbedre bildehastigheten til thE PAT-system, til og med med single-element UST sirkulær skanning geometri. Single-element UST er billigere og svært sensitive, i motsetning til array-baserte USTs. I løpet av de siste tiårene ble det rapportert lite forskning om bruk av høyrepeterings-rate PLD som excitasjonskilde for PA-bildebehandling. En fiberbasert nær-infrarød PLD ble demonstrert for PA-bildebehandling av fantomer 28 . In vivo avbildning av blodkar på en ~ 1 mm dybde under den menneskelige huden ble demonstrert ved bruk av lav-energi PLD 29 . Et PLD-basert optisk oppløsningsfotoakustisk mikroskop (ORPAM) ble rapportert. Ved bruk av PLD-er ble det vist på ~ 1,5 cm dyp bildebehandling ved en rammeplass på 0,43 Hz 30 . Svært nylig ble det rapportert et PLD-PAT-system som ga bilder på så kort som ~ 3 s og ved en ~ 2 cm billeddybde i biologisk vev 25 , 31 . Denne studien viste at et slikt lavkost, kompakt system kan gi høy-quaLikhetsbilder, selv ved høye hastigheter. PLD-PAT-systemet kan brukes til fotoakustisk avbildning med høy rammebasert hastighet (7000 fps), bildebehandling av blodkar, fingeravbildning, 2 cm dyp vevsbilde, liten dyrs hjernedannelse etc. Enkeltbølgelengden og Lavpuls-energipulser fra PLD begrenser applikasjonen til flerspektral og dypvevsbilding. Forsøk har blitt utført på små dyr ved bruk av det samme PLD-PAT-systemet som brukes til prekliniske applikasjoner. Hensikten med dette arbeidet er å gi den visualiserte eksperimentelle demonstrasjonen av PLD-PAT-systemet for in vivo 2D tverrsnittshjernedimensjonering av små dyr.

a high-performance compact photoacoustic tomography system for in vivo small-animal brain imaging

In vivo har små dyr bildebehandling en viktig rolle å spille i prekliniske studier. Fotoakustisk tomografi (PAT) er en fremvoksende hybridbildingsmodalitet som viser stort potensial for både prekliniske og kliniske anvendelser. Konvensjonelle optiske parametriske oscillatorbaserte PAT (OPO-PAT) systemer er store og dyre og kan ikke gi høyhastighets bildebehandling. Nylig har pulsed-laser dioder (PLDs) blitt demonstrert som en alternativ eksitasjonskilde for PAT. Pulserende laserdiode PAT (PLD-PAT) har blitt demonstrert for høyhastighets bildebehandling på fotoakustiske fantomer og biologiske vev. Dette arbeidet gir en visualisert eksperimentell protokoll for in vivo hjernedimensjonering ved bruk av PLD-PAT. Protokollen inkluderer den kompakte PLD-PAT-systemkonfigurasjonen og dens beskrivelse, dyreforberedelse for hjernedimensjonering, og en typisk eksperimentell prosedyre for 2D tverrsnitt rottehjernebilder. PLD-PAT-systemet er kompakt og kostnadseffektivtFektive og kan gi høyhastighets bildebehandling av høy kvalitet. Brainbilder samlet inn vivo ved forskjellige skannehastigheter presenteres.

In vivo hjernedannelsesresultater som viser evnen til det beskrevne PLD-PAT-systemet, vises i denne delen. For å demonstrere høyhastighets bildebehandlingskapasiteten til PLD-PAT-systemet ble in vivo hjernedimensjonering av to forskjellige sunne rotter utført.
