a unified methodological framework for vestibular schwannoma research

Vestibulære schwannomer er de mest almindelige neoplasmer af cerebellopontinvinklen og udgør 6-8% af alle intrakraniale vækst. Skønt disse tumorer forårsager sensorineurelt høretab i op til 95% af de ramte individer, forbliver de molekylære mekanismer, der ligger til grund for dette høretab, unnvikende. Denne artikel beskriver de trin, der er etableret i vores laboratorium for at lette indsamling og behandling af forskellige primære humane vævsprøver til downstream-forskningsapplikationer, der er integreret i studiet af vestibulære schwannomer. Specifikt beskriver dette værk en samlet metodologisk ramme for indsamling, behandling og kultur af Schwann- og schwannomaceller fra kirurgiske prøver. Dette er integreret med parallelle behandlingstrin, der nu betragtes som afgørende for nuværende forskning: indsamling af tumor- og nervesekretioner, bevaring af RNA og ekstraktion af protein fra indsamlede væv, fiksering af væv til fremstilling af sektioner, enD eksponering af primære humane celler til adeno-associerede vira til anvendelse på genterapi. Desuden fremhæver dette arbejde den translabyrintiske kirurgiske tilgang til at samle denne tumor som en unik mulighed for at opnå menneskelig sensorisk epithel fra det indre øre og periliem. Der gives tips til forbedring af eksperimentelle kvalitet og almindelige faldgruber fremhævet.

Primære humane VS-celler i kultur som fastlagt i afsnit 5 kan behandles som informative modeller for sygdomsassocierede processer i mange downstream-forskningsapplikationer ( Figur 1 ).
