Vestibulaire schwannomen (VS&#39;s) zijn de meest voorkomende neoplasma&#39;s van de cerebellopontinehoek, gediagnosticeerd bij 1 op elke 100.000 individuen. Hoewel niet-metastatische, beïnvloeden deze tumoren een levenskwaliteit van een patiënt 1 , 2 , 3 , 4 , 5 , 6 ernstig. Geïnteresseerden leven meestal met gehoorverlies, tinnitus en een gevoel van gehoorvervulling. Symptomen worden steeds meer belemmerend aangezien de tumor groeit, waardoor balansproblemen, gelaatsverlamming en vermindering van andere craniale zenuwfuncties ontstaan. Beheersopties voor VS zijn in wezen beperkt tot waakzaam wachten op statische tumoren en stereotactische radiotherapie of chirurgische resectie voor groeiende tumoren <sUp class = &quot;xref&quot;&gt; 8. De chirurgische verwijdering van deze tumoren in onderzoeksgebonden ziekenhuizen biedt de mogelijkheid om vers tumorweefsel te verzamelen en te analyseren die tijdens de patiëntenoperaties worden verzameld. a unified methodological framework for vestibular schwannoma research

Vestibulaire schwannomen zijn de meest voorkomende neoplasma&#39;s van de cerebellopontinehoek, die 6-8% van alle intracraniale groeizen vormen. Hoewel deze tumoren sensorineuraal gehoorverlies veroorzaken in 95% van de getroffen individuen, blijven de moleculaire mechanismen die tot dit gehoorverlies leiden, onduidelijk. Dit artikel schetst de stappen in ons laboratorium om de inzameling en verwerking van diverse primaire menselijke weefselmonsters te vergemakkelijken voor downstream onderzoeksapplicaties die integraal zijn bij de studie van vestibulaire schwannomen. Specifiek beschrijft dit werk een verenigd methodologisch kader voor de verzameling, verwerking en cultuur van Schwann- en schwannomacellen uit chirurgische monsters. Dit is geïntegreerd met parallelle verwerkingsstappen die nu essentieel zijn voor huidig ​​onderzoek: de verzameling van tumor- en zenuwafscheidingen, het behoud van RNA en de extractie van eiwitten uit verzamelde weefsels, de fixatie van weefsel voor de bereiding van secties, eenD de blootstelling van primaire humane cellen aan adeno-geassocieerde virussen voor toepassing op gentherapie. Daarnaast benadrukt dit werk de translabyrinthine chirurgische benadering om deze tumor te verzamelen als een unieke kans om menselijk zintuiglijk epitheel van het binnenoor en periliem te verkrijgen. Tips voor het verbeteren van de experimentele kwaliteit worden verstrekt en gemeenschappelijke valkuilen worden gemarkeerd.

Primaire menselijke VS-cellen in cultuur, zoals vastgesteld in sectie 5, kunnen in veel downstream-onderzoeksaanvragen als informatieve modellen voor ziekteverwante processen worden behandeld ( Figuur 1 ).
