Les schwannomes vestibulaires (VS) sont les néoplasmes les plus fréquents de l&#39;angle de la cerebellopontine, diagnostiqués chez 1 sur 100 000 individus. Bien que non métastatiques, ces tumeurs affectent sévèrement la qualité de vie du patient 1 , 2 , 3 , 4 , 5 , 6 . Les personnes touchées vivent généralement avec une perte auditive, un acouphène et un sentiment de plénitude auditive. Les symptômes deviennent de plus en plus débilitants à mesure que la tumeur augmente, provoquant des problèmes d&#39;équilibre, une paralysie faciale et une altération d&#39;autres fonctions du nerf crânien. Des complications potentiellement mortelles dus à la compression du tronc cérébral peuvent également entraîner 7 . Les options de gestion des VS sont essentiellement limitées à l&#39;attente vigilante de tumeurs statiques et de radiothérapie stéréotaxique ou de résection chirurgicale pour les tumeurs croissantes <sUp class = &quot;xref&quot;&gt; 8. L&#39;élimination chirurgicale de ces tumeurs dans les hôpitaux affiliés à la recherche présente l&#39;opportunité d&#39;acquérir et d&#39;analyser le tissu tumoral frais recueilli lors des chirurgies du patient. Une approche chirurgicale spécifique de VS, la résection de translabérine, peut même offrir un accès à l&#39;épithélium sensoriel humain précieux de l&#39;oreille interne et du périlymph. Parce que les VS proviennent d&#39;un nerf sensoriel périphérique ( c.-à-d. Cet article présente un cadre méthodologique unifié pour le traitement simultané d&#39;un seul échantillon VS ou GAN humain primaire pour de multiples applications en aval: culture spécifique au type de cellule, extraction de protéines, préservation de l&#39;ARN, collecte de la sécrétion tumorale et fixation des tissus. Ce travail décrit également la collecte et le traitement chirurgicaux du liquide céphalo-rachidien humain (CSF) et du perilymph lors de la résection VS de translabyrinthine, car ces tissus étroitement apparentés peuvent servir de sources importantes de biomarqueurs pour VS. Enfin, ce protocole présente des étapes pour la transduction virale de cellules VS humaines primaires en culture comme une nouvelle application de ce tissu pour une utilisation en thérapie génique.

a unified methodological framework for vestibular schwannoma research

Les schwannomes vestibulaires sont les néoplasmes les plus courants de l&#39;angle de la cerebellopontine, ce qui représente 6 à 8% de toutes les croissance intracrânienne. Bien que ces tumeurs provoquent une perte auditive sensiveureure chez 95% des personnes affectées, les mécanismes moléculaires qui sous-tendent cette perte auditive demeurent insaisissables. Cet article décrit les étapes établies dans notre laboratoire pour faciliter la collecte et le traitement de divers échantillons primaires de tissus humains pour les applications de recherche en aval intégrées à l&#39;étude des schwannomes vestibulaires. Plus précisément, ce travail décrit un cadre méthodologique unifié pour la collecte, le traitement et la culture des cellules Schwann et Schwannoma à partir d&#39;échantillons chirurgicaux. Ceci est intégré aux étapes de traitement en parallèle, maintenant considérées comme essentielles pour la recherche actuelle: la collecte des sécrétions tumorales et nerveuses, la préservation de l&#39;ARN et l&#39;extraction des protéines des tissus collectés, la fixation du tissu pour la préparation des sections,D l&#39;exposition des cellules humaines primaires aux virus adéno-associés pour l&#39;application à la thérapie génique. En outre, ce travail met en évidence l&#39;approche chirurgicale translymérine pour collecter cette tumeur comme une occasion unique d&#39;obtenir l&#39;épithélium sensoriel humain de l&#39;oreille interne et de la périlymph. Des conseils pour améliorer la qualité expérimentale sont fournis et les pièges courants sont mis en évidence.

Les cellules VS humaines primaires en culture, telles qu&#39;établies dans la section 5, peuvent être traitées comme des modèles informatifs pour les processus associés à la maladie dans de nombreuses applications de recherche en aval ( Figure 1 ).
