Klebsiella pneumoniaeは、好気性および嫌気性条件下で良好に生育する細菌である。 K. pneumoniaeは、多くの化学物質を生産するために使用される重要な産業微生物です。 1,3-プロパンジオールは、ポリトリメチレンテレフタレートを合成するためのモノマーとして主に使用される貴重な化学物質である。ポリトリメチレンテレフタレートは、1,2-プロパンジオール、ブタンジオールまたはエチレングリコール1よりも優れた特性を示す生分解性ポリエステルです。 1,3-プロパンジオールは、酸素制限条件下で基質としてグリセロールを用いて肺炎桿菌によって産生される2 。 2,3-ブタンジオールおよびその誘導体は、プラスチック、溶剤製造および合成ゴムの分野で用途があり、バイオ燃料として使用される可能性がある3 。グルコースを基質とすると、2,3-ブタンジオールは野生型株4の主要代謝産物である。 2,3-ブタンジオールはシンセシスピルビン酸から得た。まず、ピルビン酸の2分子が凝縮してα-アセト乳酸を産生する。この反応はα-アセト乳酸シンターゼによって触媒される。 α-アセトラクテートは、α-アセトラクテート脱炭酸酵素によってアセトインに変換される。ブタンジオール脱水素酵素によって触媒されると、 R-アセトインはさらに2,3-ブタンジオールに還元され得る。 K. pneumoniaeに適した効率的な遺伝子置換法が検討されており、多くの変異体が構築されている5,6,7。その2,3-ブタンジオール脱水素酵素活性を失ったbudC変異体は、培養液中に高レベルのアセトインを蓄積する。アセトインは、食品の風味を向上させるための添加剤として使用されます8 。 α-アセト乳酸デカルボキシラーゼをコードするbudAが変異すると、2-ケトグルコン酸がブロス中に蓄積する。 2-ケトグルコン酸は、エリソルビン酸（イソアスコルビン酸）の合成に使用され、食品業界で使用される酸化防止剤9 。 2-ケトグルコン酸はグルコース酸化経路の中間体であり;ペリプラズム空間に位置するこの経路では、グルコースはグルコン酸に酸化され、さらに酸化されて2-ケトグルコン酸になる。ペリプラズムで産生されたグルコン酸および2-ケトグルコン酸は、さらなる代謝のために細胞質に輸送することができる。 2-ケトグルコン酸の蓄積は酸性条件に依存し、より高い空気補給は2-ケトグルコン酸産生に有利である10 。 gadによってコードされるグルコン酸脱水素酵素は、 グルコン酸の2-ケトグルコン酸への変換を触媒する。 K. pneumoniaeのgad突然変異体は、2-ケトグルコン酸の代わりに高レベルのグルコン酸を産生し、このプロセスもまた酸性条件に依存する。グルコン酸はバルク有機酸であり、セメント1の性質を高める添加剤として使用される1。グルコン酸へのグルコース酸化は、グルコース脱水素酵素によって触媒される。キシロースは、グルコースデヒドロゲナーゼの適切な基質でもある。キシロースが基質として使用される場合、 肺炎桿菌はキシロン酸12を産生する。 バイオマスを原料とした化学生産は、バイオテクノロジーの重要なテーマです13 。バイオマスの主成分は、セルロース、ヘミセルロースおよびリグニンである。しかしながら、これらの巨大分子化合物は、ほとんどの微生物（ K. pneumoniaeを含む）によって直接的に異化されることはできない。バイオマス中のセルロースおよびヘミセルロースは、グルコースおよびキシロースに加水分解されなければならず、微生物によって使用されなければならない。リグノセルロース中のリグニンの存在は、酵素によるバイオマス加水分解を防止する保護バリアを作り出す。したがって、リグニンおよびヘミセルロースを除去し、セルロースの結晶化度を低下させる前処理工程は、常にマイクによるバイオマス利用中に行われるルーニズム。多くの前処理方法が開発されている：酸性、アルカリ性、アンモニアおよび蒸気の前処理が一般的である。 竹は熱帯および亜熱帯地域で豊富であり、重要なバイオマス資源です。ここでは、竹の加水分解物の調製と竹の加水分解物を用いた化学生産が提示されている

竹は重要なバイオマスであり、竹の加水分解物は化学生産の原料としてクレブシエラ・ニューモニエ（Klebsiella pneumoniae）によって使用される。ここでは、竹粉をNaOHで前処理し、中性pHになるまで洗浄した。前処理した竹粉にセルラーゼを添加して30g / Lのグルコースと15g / Lのキシロースを含む加水分解物を生成させ、炭素源として化学製造用の培地を調製した。微好気条件で培養すると、12.7g / Lの2,3-ブタンジオールが野生型K.ニューモニエによって産生された。好気的条件では、 K. pneumoniaeの budC突然変異体により13.0g / LのR-アセトインが生成された。 25.5g / Lの2-ケトグルコン酸と13.6g / Lのキシロン酸との混合物を、高空気補給を伴う2段階pH制御発酵におけるK.pneumoniaeのbudA突然変異体によって産生させた。発酵の第1段階では、培養物を中性pHに維持した。細胞増殖後、発酵nは第2段階に進み、その間に培養物は酸性になった。
