Klebsiella pneumoniae 는 호기성과 혐기성 조건 모두에서 잘 자라는 박테리아입니다. K. pneumoniae 는 많은 화학 물질 생산에 사용되는 중요한 산업 미생물입니다. 1,3- 프로판 디올은 폴리 트리 메틸렌 테레 프탈레이트를 합성하는 모노머로서 주로 사용되는 유용한 화학 물질입니다. 폴리 트리 메틸렌 테레 프탈레이트는 1,2- 프로판 디올, 부탄디올 또는 에틸렌 글리콜 1 보다 우수한 특성을 나타내는 생분해 성 폴리 에스테르입니다. 1,3- 프로판 디올은 K. pneumoniae 가 산소 제한 조건에서 글리세롤을 기질로 사용하여 생산됩니다 2 . 2,3- 부탄디올 및 그 유도체는 플라스틱, 용매 생산 및 합성 고무 분야에서 응용 분야를 가지며 바이오 연료로 사용될 잠재력을 가지고있다 3 . 글루코스를 기질로하는 2,3- 부탄디올은 야생형 4 주 대사 산물입니다. 2,3- 부탄디올은 synthesiz피루브산에서 에드. 첫째, 두 분자의 pyruvate가 응축되어 α-acetolactate를 생성한다. 이 반응은 α- 아세토 락 테이트 신타 제에 의해 촉매된다. α-acetolactate는 α-acetolactate decarboxylase에 의해 acetoin으로 전환된다. 부탄디올 탈수소 효소에 의해 촉매 될 때 R- 아세토 인은 2,3- 부탄디올로 더 감소 될 수있다. K. pneumoniae에 적합한 효율적인 유전자 대체 방법이 연구되었으며, 많은 돌연변이 체가 5 , 6 , 7 구축되었다. 2,3- 부탄디올 탈수소 효소 활성을 잃은 budC 돌연변이 체는 배양액에서 높은 수준의 아세토 인을 축적합니다. 아세트 인은 식품의 풍미를 향상시키는 첨가제로 사용됩니다 8 . α- acetolactate decarboxylase를 코딩하는 budA 가 돌연변이되면 2- 케토 글루 콘산이 배양액에 축적된다. 2- 케토 글루 콘산은 에리 토르 브산 (isoascorbic acid)의 합성에 사용되며,, 식품 산업에서 사용되는 산화 방지제 9 . 2- 케토 글루 콘산은 포도당 산화 경로의 중간체이며; 페리 플라 즘 공간에 위치한이 경로에서 글루코스는 글루 콘산으로 산화 된 다음 2- 케토 글루 콘산으로 더 산화된다. 페리 플라 즘에서 생성 된 글루 콘산 및 2- 케토 글루 콘산은 추가 대사를 위해 세포질로 수송 될 수있다. 2- 케토 글루 콘산의 축적은 산성 조건에 달려 있으며, 공기 보충이 높을수록 2- 케토 글루 콘산 생산이 유리합니다 10 . Gluconate dehydrogenase ( 갓 에 의해 코드화 됨) 글루 콘산의 2- 케톨 구연산으로의 전환을 촉매한다. K. pneumoniae 의 gad 돌연변이 체는 2- ketogluconic acid 대신에 고농도의 gluconic acid를 생산했으며,이 과정은 또한 산성 조건에 달려있다. 글루 콘산은 벌크 유기산으로 시멘트 1 의 물성을 높이기위한 첨가제로 사용됩니다.1. 글루 콘산으로의 글루코스 산화는 글루코오스 탈수소 효소에 의해 촉매된다. Xylose는 포도당 탈수소 효소의 적절한 기질이기도합니다. 기질로 키 실로 오스를 사용하면 키모로 오스 산이 생성됩니다. 바이오 매스를 원료로 사용하는 화학 생산은 생명 공학 분야에서 가장 중요한 주제입니다 13 . 바이오 매스의 주성분은 셀룰로오스, 헤미셀룰로오스 및 리그닌이다. 그러나 이러한 거대 분자 화합물은 대부분의 미생물 ( K. pneumoniae 포함)에 의해 직접적으로 대사 될 수 없습니다. 바이오 매스의 셀룰로오스와 헤미셀룰로오스는 반드시 가수 분해되어 포도당과 크 실로 오스가되어야하며 미생물에 의해 사용될 수 있습니다. 리그 노 셀룰로오스에 리그닌이 존재하면 효소에 의한 바이오 매스 가수 분해를 방지하는 보호막이 형성됩니다. 따라서, 리그닌 및 헤미셀룰로오스를 제거하고 셀룰로오스의 결정 성을 감소시키는 전처리 공정은 마이크에 의한 바이오 매스 이용 중에 항상 수행된다roorganisms. 많은 전처리 방법이 개발되었습니다 : 산성, 알칼리성, 암모니아 및 증기 전처리가 일반적입니다. 대나무는 열대 및 아열대 지역에 풍부하며 중요한 바이오 매스 자원입니다. 여기서는 대나무 가수 분해물의 제조 및 대나무 가수 분해물을 이용한 화학적 생산이 제시된다

대나무는 중요한 바이오 매스이며 대나무 가수 분해 물은 화학 물질 생산을위한 공급 원료로 Klebsiella pneumoniae에서 사용됩니다. 여기서, 대나무 분말을 NaOH로 전처리하고 중성 pH로 세척 하였다. 셀룰로오스를 전처리 된 대나무 분말에 첨가하여 글루코오스 30g / L 및 크실 로스 15g / L를 함유하는 가수 분해물을 생성시키고 화학 생산 용 매질을 제조하기위한 탄소 공급원으로 사용 하였다. 미세 혐기 조건에서 배양되었을 때 야생형 K. pneumoniae에 의해 12.7 g / L 2,3- 부탄디올이 생성되었다. 호기 조건에서 K. pneumoniae 의 budC 돌연변이 체에 의해 13.0g / L의 R- acetoin이 생산되었다. 25.5 g / L 2- 케 토글 루콘 산과 13.6 g / L 크 실로 닉산의 혼합물은 높은 공기 보충을 통한 2 단계, pH 조절 발효에서 K. pneumoniae 의 budA 돌연변이 체에 의해 생산되었다. 발효의 첫 번째 단계에서, 배양 물은 중성 pH로 유지되었다; 세포 성장 후, 발효n 단계는 문화가 산성화되는 것을 허용하는 두 번째 단계로 진행되었다.
