De circadiane klok regelt een brede waaier van biologische processen uit een diurnale expressie van genen te slapen gedrag in een voorspelbare ritme met een frequentie van ongeveer 24 h. epidemiologische studies suggereren sterk dat chronische verstoring van circadiane ritme verhoogt het risico op borst- en prostaatkanker in werknemers in ploegendienst, met inbegrip van verpleegkundigen en vlucht bemanningen1,2,3. Deze bevindingen worden bevestigd door knaagdieren onderzoeken, die aantonen dat de blootstelling aan constant licht, licht-at-night of lichte cycli die verhoging van de jetlag tumor incidentie nabootsen en versnelt4,5van de groei van de tumor. Gebaseerd op gegevens uit zowel menselijke en knaagdier studies, het Internationaal Agentschap voor kankeronderzoek ploegenarbeid als een waarschijnlijke menselijk carcinogeen agens (Type 2A) in 20106ingedeeld.
Eerder, we laten zien dat een enkele kankerverwekkende dosis van de borstklier tumor specifiek carcinogeen, N- nitroso-N- methylurea (NMU), verstoord de circadiane expressie van genen die grote circadiane (CGs) (b.v., periode 2 , Per2) en verschillende circadiane-gecontroleerde genen (CCGs), met inbegrip van DNA-beschadiging responsieve toets en gerepareerd (DDRR) genen in de melkklier doel (maar niet in de lever). Inzicht in de mechanismen die een verstoring van de circadiane door toxische stoffen in het milieu en de pathogenese link kan leiden tot mechanistically gebaseerde benaderingen van ziektepreventie. Echter studies gericht op het definiëren van de interacties tussen de blootstelling en het circadiane ritme worden gewoonlijk uitgevoerd in vivo. Een typische in vivo experiment onderzoeken die de impact op de circadiane ritme grote aantallen dieren, vereist zoals weefsels uit ten minste drie controle en drie blootgestelde dieren moeten worden verzameld elke 3-4 h over een periode van 24 of 48 h. Ontwikkeling van een gevalideerde in vitro -systeem dat in vivo observaties en mechanismen recapituleert zou dus niet alleen het verminderen van het aantal dieren vereist, maar ook drastisch verminderen experimentele kosten en de eis dat onderzoekers werken voortdurend over een periode van 24-48 h. Bovendien kan een gevalideerde in vitro -systeem worden gebruikt voor high throughput screening van verbindingen en/of genetische wijziging die invloed hebben op het circadiane ritme, of haar reactie op milieustressoren of toxische stoffen. Daarom zijn de strategische combinatie van in vitro en in vivo modellen en experimenten nodig om het verkrijgen van verschillende inzichten met verschillende focus.
Bij zoogdieren bestaan circadiane oscillatoren niet alleen in gespecialiseerde neuronen van de SCN, maar ook in de meest perifere celtypes. Deze moleculaire klokken zijn vergelijkbaar met die in de gevestigde fibroblast cellijnen en primaire fibroblasten van embryo's of volwassen dieren; Er is echter behoefte aan een weefsel type-specifieke cellulaire modellen11. Bijgevolg, traditionele studies van motorische activiteit in vivo, SCN explants ex vivoen cel-gebaseerde in vitro testen in vereeuwigd fibroblast cellen worden veel gebruikt om te studeren van cel-autonome circadiane gebreken. Echter, er is geen bewijs dat aangeeft dat een in vitro fibroblast cel-gebaseerde bepaling circadiane mechanismen recapituleren kan en reacties in cellen van andere perifere organen in vivo. Verschillende soorten cellen kunnen verschillende patronen van genexpressie, xenobiotische metabolisme en DDRR, en het verband tussen giftigheid en circadiane genexpressie kunnen celtype specifieke en/of gemoduleerd door verschillende fysiologische parameters. Daarnaast hebben circadiane oscillatoren in fibroblast gebaseerde systemen niet volledig geëvalueerd voor reacties op toxische stoffen in het milieu, stressoren en preventieve agenten die link blootstelling aan mechanismen voor de ontwikkeling van de ziekte en preventie. Dus, is er een behoefte aan facile, gevalideerde cel-systeemeigen, in vitro bioluminescentie testen om te studeren orgel specifieke milieu circadiane disruptors. Renilla luciferase (rLuc) en firefly luciferase zijn 30-61 kDa monomeer eiwitten die vereisen geen posttranslationele verwerking voor enzymatische activiteit en kunnen functioneren als een genetische verslaggever onmiddellijk na de vertaling. Zodra het substraat aan het enzym luciferase koppelt, genereert de biochemische reactie gekatalyseerd een lichtflits. Luciferase constructies zijn zo grote schaal gebruikt als een gen expressie verslaggever systeem in vitro en in vivo. Echter in de circadiane ritme studies, het nut van de verslaggever luciferase wordt beperkt door de relatief lange halveringstijd van het eiwit luciferase (T-1/2 = 3.68 h) ten opzichte van de periode (met name voor de korte periode) voor veranderingen in het circadiane gen expressie; echter heb tal van studies door de jaren heen met succes gebruikt het luciferase-gen in de pGL3 vector, waaruit blijkt dat het snel afbreekbaar luciferase niet mogelijk nodig voor het melden van circadiane ritmes, vooral voor de ritmes met een langere periode, zoals 24 h. Dus, een verslaggever plasmide met behulp van gedestabiliseerd luciferase vector, pGL [Luc2P/Neo], waarin de hPEST (een opeenvolging van de destabilisatie eiwit) is ontwikkeld, waardoor we kunnen gebruiken als een vector circadiane verslaggever voor onze huidige in vitro bioluminescentie assay. Het eiwit dat gecodeerd wordt door Luc2P heeft een veel kortere halfwaardetijd (T-1/2 = 0.84 h) en vandaar, sneller en met een grotere magnitude reageert op veranderingen in transcriptionele activiteit dan wild-type, ikndicating dat het kan worden gebruikt voor het bewaken van de ritmische uitdrukking van luciferase geregeld door de promotor van de PER2 nauwkeurig in real-time16.

in vitro bioluminescence assay to characterize circadian rhythm in mammary epithelial cells

De circadiane ritme is een fundamentele fysiologische proces aanwezig in alle organismen dat biologische processen, variërend van genexpressie slapen gedrag regelt. In gewervelde dieren, wordt circadiane ritme geregeld door een moleculaire oscillator die functioneert in zowel de suprachiasmatic nucleus (SCN; centrale pacemaker) en de afzonderlijke cellen bestaande uit de meest perifere weefsels. Belangrijker, is verstoring van circadiane ritme door blootstelling aan licht-at-night, milieustressoren en/of toxische stoffen geassocieerd met verhoogd risico van chronische ziektes en veroudering. De mogelijkheid om agenten die centrale en/of perifere biologische klokken kunnen verstoren, en agenten die kunnen voorkomen of verzachten van de gevolgen van de verstoring van circadiane, te identificeren heeft aanzienlijke gevolgen voor de preventie van chronische ziekten. Hoewel knaagdier modellen kunnen worden gebruikt als aanduiding van de posities en agenten die veroorzaken of verstoring van circadiane voorkomen/verzachten, vereisen deze experimenten grote aantallen dieren. In vivo studies ook vereisen aanzienlijke middelen en infrastructuur, en vereisen van onderzoekers aan het werk van de hele nacht. Dus, is er dringend behoefte aan een celtype passende in vitro systeem naar scherm voor milieu circadiane disruptors en smaakversterkers in celtypes uit verschillende organen en ziekte staten. We hebben een vector die transcriptie van de gedestabiliseerd luciferase in eukaryote cellen onder de controle van de menselijke periode 2 gen promotor drijft gebouwd. Deze constructie circadiane verslaggever was stabiel naar menselijke borstklier epitheliale cellen transfected en circadiane responsieve verslaggever cellen werden geselecteerd om de Bioluminescentie in vitro assay. Hier presenteren we een gedetailleerd protocol vaststellen en valideren van de bepaling. We leveren verdere details voor bewijs van concept experimenten aantonen van het vermogen van onze in vitro -assay te recapituleren de effecten in vivo van verschillende chemische stoffen op de cellulaire biologische klok. De resultaten wijzen erop dat de bepaling aangepast aan een verscheidenheid van celtypes worden kan te screenen voor zowel milieu disruptors en chemopreventive versterkers van circadiane klokken.
