I denne undersøgelse, vi induceret HI hjerneskade i en neonatal P7-10 CD-1 mus og identificeret hjernen læsion med relevante kognitive og motoriske underskud. Neonatal HI hjerneskade opstår i barndommen (ca to patienter pr. 1.000 børn)1,2,3,4,5. Undersøgelser vedrørende neonatal HI hjerneskade er vigtige, og hjælp en etablerede neonatal HI hjerne skade musen model kan lette i vivo prækliniske forskning på HI hjerneskade.
Traditionelle HI modeller anvendes på voksne rotter6. For den nyfødte model, er ris-Vannucci metode almindeligt anvendt på P7 rotter7,8. Men da rotter og mus er lidt anderledes9,10, selv om de begge gnavere, vi udførte en modificeret ris-Vannucci metode på CD-1 hvalpe på P7-10, baseret på tidligere undersøgelser, der viste, at P7-10 er perioden byder umodne oligodendrocytes, svarende til menneskets sigt P011,12. Neonatal HI mus modellen er etableret gennem begge ligation af den ensidige halspulsåren og musene eksponering for hypoxi med 8% ilt i P7-10 unger.
Musene underkastet proceduren viste forskellige grader af hjernelæsioner i området posterolateral i højre hjernehalvdel. For at identificere den kognitive og motoriske underskud, neurobehavioral vurderinger baseret på PAT, blev stigen walking test og greb styrke test udført. neurobehavioral assessments in a mouse model of neonatal hypoxic-ischemic brain injury

Vi udført ensidige carotis arterieokklusion på CD-1 mus til at oprette en neonatal hypoksiske iskæmisk (HI) model og undersøgt virkningerne af neonatal HI hjerneskade ved at studere neurobehavioral funktioner i disse mus, sammenlignet med ikke-opererede (dvs. normal) mus. I løbet af undersøgelsen, blev ris-Vannucci metode brugt til at fremkalde neonatal HI hjerneskade i postnatal dag 7-10 (P7-10) mus. HI operation blev udført på hvalpene af ensidige carotis arterie ligatur og eksponering for hypoxi (8% O2 og 92% N2 til 90 min). En uge efter operationen, de skadede hjerner blev vurderet med det blotte øje gennem semi-transparent kraniet og blev kategoriseret i undergrupper baseret på fravær ("ingen kortikal skade" gruppe) eller tilstedeværelse ("kortikal skade" gruppe) af kortikal skade, som en læsion i højre hjernehalvdel.
