I denne studien vi indusert HI hjerneskade i neonatal P7-10 CD-1 mus og identifisert hjernen lesjonen med relevante kognitive og motor underskudd. Under denne prosedyren var okklusjon av de ensidige høyre arteria carotis avgjørende. I dette trinnet kunne arterien bli skadet og revet. De fleste pups som opplevd en arterie rive døde. Omvendt, hvis forskere samskrevet en annen blod blodåre i stedet for den ensidige høyre arteria carotis, hjerne pup bare mildt ble skadet og ingen betydelig fenotypen kunne observeres<sup...

Neonatal HI hjerneskade oppstår under tidlig barndom (ca to pasienter per 1.000 barn)1,2,3,4,5. Studier om neonatal HI hjerneskade er viktig, og bruker en etablert neonatal HI hjernen skade musemodell kan lette i vivo prekliniske undersøkelser på HI hjerneskade.
Tradisjonelle HI modeller brukes på voksen rotter6. For den neonate modellen brukes ris-Vannucci metoden vanligvis på P7 rotter7,8. Men siden rotter og mus er litt annerledes9,10, selv om de begge gnagere, vi utført en endret ris-Vannucci metode på CD-1 pups på P7-10, basert på tidligere studier som viste at P7-10 er perioden med umoden oligodendrocytes, tilsvarer human frist P011,12. Neonatal HI musemodell er etablert gjennom både av ligation av den ensidige arteria carotis og eksponering for mus hypoksi med 8% oksygen i P7-10 pups.
Mus utsatt for prosedyren viste ulike grader av hjernen lesjoner i posterolateral-området i den høyre hjernehalvdelen. For å identifisere den kognitive og motor underskudd, neurobehavioral vurderingene er basert på PAT, ble stigen gå test, og grep styrke test utført. Forskjellene mellom ikke-opererte (dvs. vanlig) og HI mus ble analysert. Dette arbeidet gir grunnleggende informasjon om HI musemodell. neurobehavioral assessments in a mouse model of neonatal hypoxic-ischemic brain injury

Vi utførte ensidige arteria carotis okklusjon på CD-1 mus å opprette en neonatal hypoxic-iskemiske (HI) modell og undersøkt virkningene av neonatal HI hjerneskade ved å studere neurobehavioral funksjoner i disse musene sammenlignet med ikke-opererte (dvs. normal) mus. Studien, ble ris-Vannucci metoden brukt til å indusere neonatal HI hjerneskade i postnatal dag 7-10 (P7-10) mus. HI operasjonen ble utført på valpene ensidige arteria carotis ligatur og eksponering for hypoksi (8% O2 og 92% N2 for 90 min). En uke etter operasjonen, skadet hjernen ble evaluert med det blotte øye gjennom semi-transparent skallen og ble kategorisert undergrupper basert på fravær ("ingen kortikale skader" gruppe) eller tilstedeværelse ("kortikale skader" gruppe) av kortikale skader, som en leksjonen i høyre halvkule.
