Er zijn verschillende technische details die speciale aandacht behoeven tijdens de installatie van het ATOM-systeem. Optische beeldvorming presenteert een krachtige tool en cel gebaseerde analyse om (bijna) niet-invasieve de gedetailleerde ruimtelijke verdeling van veel cellulaire / subcellulaire componenten te visualiseren, waardoor een groot aantal morfologische, biofysische en biomoleculaire signaturen van cellen wordt ontdekt. Deze mogelijkheid om high-content informatie uit enkele cellen te verzamelen is echter in het algemeen gecompromitteerd wanneer een enorme en heterogene celcellen onderzocht moesten worden. Dit markeert een gemeenschappelijke afweging in celanalyses tussen meting doorvoer en inhoud. Een opvallend voorbeeld is dat het toevoegen van beeldcapaciteit om de cytometrie te stromen resulteerde in een afschaling van de doorvoer met ten minste 1-2 orders van grootte in vergelijking met die van de klassieke non-imaging flow cytometers. Hoewel het complexe morfologische single-cell analyse kan bieden die niet mogelijk is met standaard flow cytometers 1 , ontbreekt beeldstroomcytometrie over het algemeen onvoldoende doorvoer naar idCellulaire heterogeniteit met hoge statistische vertrouwen inhouden. Dit is nodig voor nieuwe ontdekkingen in de biologie en voor het begrijpen van de pathogenese van ziekten. De belangrijkste technische uitdaging ligt in de inherente snelheidsbeperking die wordt opgelegd door de gemeenschappelijke optische beeldvormingstrategieën: laserstraalscanning, ( bijvoorbeeld door galvanometrische spiegels) en / of beeldsensoren ( bijv. Laadkoppelapparaat (CCD) en complementair metaal- Oxide halfgeleider (CMOS)). De laserscansnelheid wordt intrinsiek beperkt door de mechanische inertie van de aftastspiegels, terwijl de beeldsnelheid van CCD of CMOS beperkt is door de fundamentele afweging tussen beeldsnelheid en gevoeligheid ( dwz het verhogen van de beeldsnelheidskanalen tot een verminderde signaaldetectiegevoeligheid , en vice versa). Op basis van een all-optisch, ultrafast beeldcoderend mechanisme is optische tijdreeksbeeldvorming aangetoond als een aantrekkelijk platform voor high-throughput imaging flow cytOmeters, zonder de conventionele beeldsensoren of mechanische laserscanning 2 , 3 . Gedetailleerde beschrijvingen van het werkprincipe van tijdreeksbeeldvorming zijn te vinden in referenties 4 , 5 , 6 , 7 . Kort samengevat bestaat het uit twee uitwisselbare mappingstappen: (i) spectrale codering (golflengte-ruimte mapping), waarbij de ruimtelijke coördinaten van het afgebeelde exemplaar worden toegewezen aan verschillende golflengten over het spectrum van de lichtpulse bundel 8 , 9 , En (ii) een dispersieve Fourier-transformatie (golflengte-time mapping) 9 , waarbij de golflengtecomponenten van individuele laserpulsen worden getransformeerd (uitgestrekt) via groepsnelheidsdispersie (GVD) in temporele golflengte-geveegde golfvormen ( Figuur 1 ). Een belangrijkeKenmerk van tijdreeksbeeldvorming is optische versterking, die een kritische rol speelt bij het bestrijden van het verlies van gevoeligheid door ultrafast fotodetectie en GVD-verlies, waardoor de beeldsignaal-ruisverhouding (SNR) wordt versterkt zonder dat het door het fotodetectorgeluid 9 wordt besmet . Aangezien elke laserpuls codeert voor een lijnscan van het afgebeelde monster, dat orthogonaal is voor de unidirectionele stroom van de cellen, wordt een effectieve lijnsnelheid bepaald door de laserherhalingstemperatuur, die typisch meer dan 10 MHz is. Deze ultrasnelle bediening maakt het mogelijk om onduidelijk beeldvorming met een cel op een doorsnede van 10.000-100.000 cellen / s te maken ( dwz 10-100 keer hoger dan conventionele beeldvormende flowcytometrie). Als gevolg daarvan kan tijdreeksbeelden unieke toepassingen vinden in high-throughput, single-cell, beeldgebaseerde screening, vooral wanneer er behoefte bestaat aan onbekende heterogeniteit of zeldzame / afwijkende cellen binnen een aanzienlijke populatie (duizenden tot miljoenen CEL<html> Ls), zoals zeldzame kankercelscreening 10 of microalgeclassificatie 11 . Tijdreeksbeeldvorming berust voornamelijk op beeldvorming op het gebied van de heldere veld (BF), waaruit het beeldcontrast wordt gegenereerd door lichtverdeling en absorptie van de cellen 2 , 3 , 9 , 10 , 11 . Zulke etiketvrije, single-cell imaging mogelijkheden kunnen de schadelijke effecten die verband houden met de fluorescerende labels, zoals cytotoxiciteit en photobleaching, omzeilen en toch waardevolle informatie verschaffen voor single-cell analyse gebaseerd op de cellulaire en subcellulaire textuur en morfologie. Deze labelvrije parameters zijn bewezen effectief te zijn voor de diepbeeldclassificatie van cellen, vooral als er een enorme celbevolking beschikbaar is 11 ,&quot;Xref&quot;&gt; 12. In veel gevallen verzekert BF imaging echter niet voldoende contrast om de gedetailleerde morfologie van de labelvrije transparante cellen te onthullen. Verschillende labelvrije, fasecontrast-, tijdreeksafbeeldingsmodaliteiten zijn ontwikkeld om het imagingcontrast te verbeteren bij ultra snelle beeldsnelheden 13 , 14 , 15 . Onder deze technieken werd asymmetrische detectie-tijdreeks optische microscopie (ATOM) ontwikkeld om het fasegradiënt (differential-interference-contrast- (DIC) -achtig) contrast te tonen op basis van een concept vergelijkbaar met Schlieren photography, waardoor de label- Vrije, hoge contrast beeldvorming van enkele cellen met een ultrahoge microfluïdische snelheid (tot 10 m / s) 16 . Dit effect kan gemakkelijk worden gegenereerd door middel van schuine detectie of verlichting door het beeldgekodeerde straalpad gedeeltelijk te blokkeren of de balk voor de fotodetectie te kantelen. Een ander voordeel van ATOM is its aMogelijkheid om tegelijkertijd twee fase-gradiëntcontrasten te verkrijgen tegenover tegengestelde oriëntaties. Intensiteits subtractie en summatie van twee tegengestelde contrastbeelden geven het differentiaalfase-gradiëntcontrast en het absorptiecontrast respectievelijk van dezelfde lijnscanning. Dit werk bevat een gedetailleerd protocol waarin de implementatie van ATOM wordt beschreven, met inbegrip van de oprichting van de optische opstelling, de steekproefvoorbereiding en de dataverzameling en visualisatie. Specifiek, dit werk demonstreert de ATOM operatie met single-cell imaging van menselijke bloedcellen, kankercellen en fytoplankton (microalgen). Dit wijst op de toepasbaarheid van ATOM op het imaging van flowcytometrie, niet alleen in de biomedische arena, maar ook in onderzoek op zee- en biobrandstoffen 17 , 18 .

Het scaleren van het aantal meetbare parameters, waarmee multidimensionale data-analyse en daarmee hogere-vertrouwen statistische resultaten mogelijk zijn, is de belangrijkste trend in de geavanceerde ontwikkeling van flowcytometrie. Met name, het toevoegen van hoge resolutie beeldvorming mogelijkheden staat voor de complexe morfologische analyse van cellulaire / subcellulaire structuren. Dit is niet mogelijk met standaard flow cytometers. Het is echter waardevol voor het bevorderen van onze kennis van cellulaire functies en kan profiteren van wetenschappelijk onderzoek, klinische diagnostiek en milieu-monitoring. Het integreren van beeldcapaciteit in flowcytometrie maakt een compromis voor de analyse, vooral door de beperkingen op snelheid en gevoeligheid in de camera technologieën. Om deze snelheid of doorstroomuitdaging tegen beeldstroomcytometrie tegen te gaan terwijl de beeldkwaliteit wordt behouden, is de asymmetrische detectie-tijdoptische optische microscopie (ATOM) aangetoond om in hoge resolutie, single-cell imagMet een subcellulaire resolutie, bij een beeldvormende doorvoer zo hoog als 100.000 cellen / s. Op basis van het beeldvormende concept van conventionele time-stretch imaging, die afhankelijk is van all-optical image encoding en retrieval door het gebruik van ultrafast breedband laser pulsen, bevordert ATOM de beeldprestatie verder door het beeldcontrast van ongemerkt / niet-gestikte cellen te verbeteren. Dit wordt bereikt door toegang te krijgen tot de fase-gradiëntinformatie van de cellen, die spectraal gecodeerd wordt in single-shot breedbandpulsen. Bijgevolg is ATOM bijzonder gunstig in metingen met een hoge doorvoer van single-cell morfologie en textuurinformatie die indicatief is voor celtypes, staten en zelfs functies. Uiteindelijk zou dit een krachtig beeldvormend flow cytometry platform kunnen worden voor de biofysische fenotypering van cellen, die de huidige state-of-the-art biochemische markering gebaseerde cellulaire test aanvullen. Dit werk beschrijft een protocol om de sleutelmodules van een ATOM-systeem vast te stellen (van optische frontend naar data pRocessing en visualisatie backend), evenals de workflow van beeldstroom-cytometrie gebaseerd op ATOM, met behulp van menselijke cellen en microalgen als voorbeelden.
