Here we describe a method for the recovery of a bipartite infectious cDNA clone system for ZIKV. Previously described clones for ZIKV suffer from either attenuation or require the addition of introns, making plasmids larger and preventing rescue in insect cells. Infectious virus can be recovered using the two-plasmid clone system in either mammalian or insect cells (data not shown). Zika-virus (ZIKV; Family Flaviviridae : Genus Flavivirus ) is een muskietgedragen flavivirus die in 2013-14 in Brazilië is aangekomen en vervolgens geassocieerd werd met een massale uitbraak van febriele ziekte die zich verspreidde over heel Amerika 1 . Na spijsvertering en ligatie om de twee fragmenten aan te sluiten kan full-length virale RNA gegenereerd worden door in vitro transcriptie met T7 RNA polymerase. Na elektroporatie van getranscribeerde RNA in cellen werd virus hersteld die respectievelijk dezelfde in vitro groei kinetiek en in vivo virulentie en infectie fenotypen vertoonde in respectievelijk muizen en muggen.

Het hier beschreven protocol zorgt voor het herstel van infectieus kloon afgeleid Zika virus. Het manipuleren van het infectieuze kloonsysteem met twee plasmiden is rechtdoor wanneer het met zorg wordt uitgevoerd, in vergelijking met volledige lengteversies die zeer instabiel zijn (data niet getoond).
