Here we describe a method for the recovery of a bipartite infectious cDNA clone system for ZIKV. Previously described clones for ZIKV suffer from either attenuation or require the addition of introns, making plasmids larger and preventing rescue in insect cells. Infectious virus can be recovered using the two-plasmid clone system in either mammalian or insect cells (data not shown). Enfeksiyöz cDNA klonları bir virüsün genetik manipülasyonuna izin verir, böylece aşılar üzerinde çalışma, patogenez, replikasyon, bulaşma ve viral evrimi kolaylaştırır. Burada Zika virüsü (ZIKV) için şu anda Amerika&#39;da patlayıcı bir salgına neden olan bulaşıcı bir klonun yapımını anlatıyoruz. Flavivirüs kaynaklı plazmidlerle sıkça görülen bakterilere toksisiteyi önlemek için genomu NS1 geninde ayıran ve mutasyonsuz başarıyla kurtarılamayan tam uzunluktaki yapılardan daha kararlı olan iki plazmid sistemi ürettik. İki parçaya katılmak için sindirim ve ligasyondan sonra, T7 RNA polimeraz ile in vitro transkripsiyon ile tam uzunluktaki viral RNA üretilebilir. Transkribe RNA&#39;nın hücrelere elektroporasyonundan sonra sırasıyla farelerde ve sivrisineklerde benzer in vitro büyüme kinetikleri ve in vivo virülans ve enfeksiyon fenotiplerini sergileyen virüs bulunmuştur.

Burada açıklanan protokol, bulaşıcı klon türevi Zika virüsünün iyileşmesine izin verir.
