Esses métodos não são capazes de exibir com precisão todos os motivos de sinalização comumente encontrados in vivo. Tais mecanismos de sinalização incluem sequestrados e fatores de crescimento, sinalização celular, sinais bioquímicos espacialmente específicas. Além disso, a rigidez do substrato pode desempenhar um papel importante na diferenciação de comportamento e células-tronco de células e não é facilmente manipulado usando celular comum cultivo práticas1,2. Abordagens de biomaterial para oferecer uma nova plataforma para começar a explorar esses mecanismos de sinalização. Em particular, o hidrogel é excelentes candidatos para ajuste de substrato rigidez3,4, imobilização de proteínas e peptídeos5,6e criando padrões específicos espacialmente7, 8.
Hidrogel é comumente usados como andaimes em engenharia de tecidos devido a suas semelhanças bioquímicas e biofísicas com a matriz extracelular (ECM)9,10. Polímeros naturais são opções comuns para andaimes, que são biocompatíveis e são encontrados em muitos tecidos do corpo. A limitação do uso de polímeros naturais como substratos é que faltam partes químicos facilmente manipulados para bioconjugation. Por outro lado, o hidrogel sintético, como tal como PEG, é excelentes plataformas para químicos alvo11,12. Além disso, PEG hidrogel não eliciar uma resposta celular e, portanto, é usados como backbones de inertes para a criação de andaimes bioativos.
Para criar o hidrogel bioativos, tanto fotopolimerização e tiol-ene clique química reações são empregadas. Estes photoreactions requerem um fotoiniciador e uma fonte de luz UV. Quando Fotoiniciadores são introduzidas à luz UV, laços quebrar a radicais de formulário. Radicais de teses são necessários para iniciar a reação, mas podem afetar negativamente a proteína Bioatividade12,13. Portanto, é fundamental otimizar o fotoiniciador e tempos de exposição UV para manter a bioatividade de proteína.
Neste método, o hidrogel é sintetizados através de acrilato-acrilato fotopolimerização de crescimento de cadeia. Monômeros de PEG-diacrylate (PEGDA) reagem uns com os outros para formar redes de polímero ramificado responsável pela estrutura do hidrogel. A concentração de monómeros PEGDA dentro da solução de precursor de gel controlará a rigidez do substrato. Devido o pequeno pore tamanho do hidrogel, proteínas de ECM como fibronectina podem ser facilmente incorporadas dentro o hidrogel para efeitos de fixação da célula. Finalmente, estes hidrogel pode ser superfície-estampados com peptídeos bioativos ou proteínas através do thiol-ene clique em reações de química. Aqui, não tenha reagidos acrilatos livre dentro do sistema de hidrogel reagirá com tióis livre localizados na proteína ou peptídeo quando exposto à luz UV. Depois que as proteínas ou peptídeos são imobilizados na superfície de hidrogel, o hidrogel pode ser armazenado a 4 ° C durante várias semanas sem perder a bioatividade. Isto oferece conveniência, planejamento experimental flexíveis e a possibilidade de colaboração entre laboratórios. Abordagens de cultura celular geral dependem de fatores solúveis dentro o meio para controlar o comportamento de célula. No entanto, adições solúveis não podem imitar determinados motivos, tais como fatores de crescimento ligados a matriz de sinalização, sinalização celular e espaciais pistas bioquímicas, que são influências comuns em células. Além disso, as propriedades biofísicas da matriz, tais como a rigidez do substrato, desempenham importantes funções no destino da célula, que não é facilmente manipulado usando celular convencional, as práticas de cultivo. Neste método, descrevemos um protocolo simples para fornecer proteínas bioativas estampadas em hidrogel sintético polietileno glicol (PEG) usando fotoquímica.
