Macroautophagy (autophagy, heri) er et cellulært stress mekanisme, der er karakteriseret ved binding af bulk cytoplasma, proteiner og organeller i dobbelt membran vesikler kaldet autophagic vesikler. Gennem fusion af det ydre lag af den dobbelt membran, autophagic blærer og deres last er leveret til lysosomer og forringet deri1. Autophagy opstår på lav basal niveauer i alle celletyper og i alle organismer, udfører homeostatiske funktioner såsom protein nedbrydning og organelle (fx, mitokondrierne) omsætning. Under vilkår, som fører til cellulært stress, såsom sult, autophagy er hurtigt upregulated og tillader cellen at opretholde energi niveauer og basale stofskifte1,2,3.
Omkring 30 autophagy gener er blevet klonet fra gæren og deres proteinprodukter blev vist sig at spille en rolle i forskellige stadier af den autophagic proces, herunder vesikel Nukleering, ekspansion, vesikel fusion til sent endosome/lysosomet og last forringelsen 4 , 5. Undersøgelser i det sidste årti viste, at flere autophagy-relaterede proteinkomplekser og protein-protein interaktioner findes, og at de regulerer autophagy veje i en indviklet og kontrolleret måde. Vejkryds, sikkerhedskopier, feedback og feedforward mekanismer eksisterer, og de tillader cellen at koordinere autophagy med andre relaterede begivenheder (såsom vesikulære sekretion, lysosomet Biogenese, endosomal sortering og transport7, osv.) Immunofluorescens-baserede metoder er almindeligt anvendt til at overvåge protein-protein interaktioner. Protein-protein interaktioner er vigtige for forståelsen cellulær signalering kaskader og identificere roman pathway komponenter og protein dynamics. Fleste af cellulære aktiviteter kræver fysisk interaktion mellem proteiner. For at analysere og kortlægge disse interaktioner, blev forskellige eksperimentelle teknikker såvel som bioinformatik værktøjer udviklet. Autophagy er en cellulær genanvendelse mekanisme, der tillader cellerne til at håndtere forskellige stressfaktorer, herunder næringsstof afsavn, kemikalier og hypoxi. For bedre at forstå autophagy-relaterede signalering begivenheder og til at opdage nye faktorer, der regulerer proteinkomplekser i autophagy udført vi protein-protein interaktion skærme. Validering af disse screening resultater kræver brug af immunofluorescens og immunoprecipitation teknikker. I dette system, specifikke autophagy-relaterede protein-protein interaktioner, der opdagede vi blev testet i Neuro2A (N2A) og HEK293T cellelinjer.
