Om te beoordelen en optimaliseren van het protocol van de fabricage, gebruikten we een eenvoudige lay-out voor het patroon van de microfluidic met een grote rechthoekige kamer (5 x 2,5 mm grootte) in het centrum, twee kleine rechthoekige kamers (5,5 x 0,75 mm grootte) gescheiden van de belangrijkste circuit op het bovenste en onderste zijden, en 300 µm breed in-laat/uitgaand-let kanalen. Fourier Transform Infrared Spectro-microscopie (FTIR) is uitgebreid gebruikt als een etiket-vrije en niet-invasieve beeldvormende techniek gedetailleerde chemische informatie van een monster te verstrekken. Hierdoor is de winning van biochemische informatie te bestuderen van de chemie van biologische monsters, met een minimum bedrag van voorbereiding omdat het absorptiespectrum van het specimen de intrinsieke vingerafdrukken van haar chemische samenstelling1 draagt , 2. onlangs, FTIR is steeds meer toegepast op de studie van de levende biologische monsters, bijvoorbeeld, cellen3. Water, dat het medium voor de levende cellen in de meeste gevallen is, blijkt echter een sterke absorptie in de regio Midden-IR. Zelfs als een dunne laag, kan haar aanwezigheid volledig overweldigen de belangrijke structurele informatie van de specimens.
Jarenlang, was de gemeenschappelijke benadering vaststelling of drogen van de monsters om volledig uitsluiten het water absorptie signaal in het spectrum. Echter, deze aanpak staat geen voor real-time metingen op levende cellen, die is van essentieel belang om te bestuderen van de verandering van hun chemische samenstelling en cellulaire processen met de tijd. Een manier voor het verkrijgen van betrouwbare absorptiespectra van levende biologische monsters, is het beperken van de totale optische weglengte in het medium van de IR-straal tot minder dan 10 µm4.
Een gevestigde aanpak in levende cel experimenten geweest tot nu toe verzwakte totale reflectie (ATR)-FTIR imaging, waardoor metingen onafhankelijk van de dikte van het monster, waardoor cellen worden volgehouden in een dikkere laag van waterig medium. Metingen van de monsters wordt echter van het oppervlak van de ATR kristal5de kleine diepte van de penetratie van de vluchtig Golf beperkt tot alleen de eerste paar microns.
U kunt ook de water absorptie beperking heeft omzeild met de opkomst van verschillende systemen van de microfluidic, die zijn over het algemeen ingedeeld in twee grote groepen: open kanaal (waar één van de vloeistof oppervlakken wordt blootgesteld aan de atmosfeer) en gesloten kanaal (waar twee IR-transparante vensters worden gescheiden door een spacer met een gedefinieerde dikte).
Loutherback et al. ontwikkelde een open-kanaal membraan-apparaat waarmee lange termijn IR continumetingen van levende cellen voor maximaal 7 dagen-6. De methode vereist hoge luchtvochtigheid in de omgeving om te voorkomen dat de verdamping van het medium van het celoppervlak. Het systeem werkt het best met cellen die natuurlijk bij air-liquide interfaces, zoals epitheliale weefsels van de huid, longen, en ogen of microbiële biofilms7 groeien.
Een gesloten-kanaal configuratie beoogt te creëren van een uniform, dunne laag tussen twee parallelle IR-transparante vensters, waar cellen worden bijgehouden in hun waterige media. De dikte van deze holte is zodanig dat het water absorptie signaal hieronder verzadiging is. Water achtergrond kan vervolgens worden afgetrokken om te verkrijgen van de juiste monster spectra. Allermeest naar de gesloten-kanaal methoden maken gebruik van een plastic spacer scheiden van de twee vensters te vormen van een afneembare vloeibare kamer3,8,9. Een voordeel van deze methode is dat het niet nodig microfabrication; structuren die meer complex dan een meetkamer met in - en uitgaand - let kanalen zijn echter moeilijk te realiseren in de dunne spacer. Er is ook een probleem met de reproduceerbaarheid van de weglengte tussen IR metingen als gevolg van de afhankelijkheid van de mechanische klemmen. Met het oog op een meer nauwkeurige controle van de afstand voor een meer betrouwbare spectrum verwerving, optische lithografie methoden doorgevoerd naar patroon fotoresist bovenop het IR-substraat te definiëren van de spacer9,10 , 11 , 12. Hoewel dit het mogelijk voor meer complexe structuren worden gedefinieerd in de spacer maakt, de methode vereist toegang tot een microfabrication faciliteit voor de productie van het patroon op elke ondergrond.
In deze paper presenteren wij een eenvoudige productie-techniek van een IR-compatibele microfluidic apparaat, met als doel om de productie kosten en de eis van de toegang tot een microfabrication faciliteit. De methode hier gepresenteerd (Zie Figuur 1) gebruikt een gevestigde proces dat bekend staat als zachte lithografie. In dit geval zijn twee mallen vereist. De primaire mal bestaat uit een 4-inch silicium wafer met behulp van een standaard UV lithografie proces. De secundaire schimmel is de replica gemaakt van PDMS, die heeft een omgekeerde polariteit van het patroon in de primaire mal van silicium en fungeert als de meester mal voor latere apparaat fabricage.
Het apparaat heeft twee afzonderlijke lagen: een eerste laag met de lay-out voor een microfluidic (die in de voorgelegde geval bestaat uit het microfluidic-kanaal en in-laat/out-let een kamer observatie met een viewport CaF2 ), en een tweede laag met een vlakke ondergrond () die bestaat uit alleen een CaF2 viewport).
Hier is een UV-uithardende optische lijm, Norland optische lijm 73 (NOA73, afgekort als NOA voortaan), gebruikt om te vormen de belangrijkste kunststof behuizing van het apparaat. Er zijn verschillende voordelen van het gebruik van deze optische lijm: lage productie kosten, gemak van connectiviteit met externe systemen, goede optische transparantie, lage viscositeit en bovenal biocompatibiliteit13. CaF2 is een geschikte keuze als de viewport als gevolg van de biocompatibiliteit en een uitstekende IR-transparantie14.
Met deze nieuwe aanpak is toegang tot een microfabrication faciliteit strikt alleen vereist voor de fabricage van de primaire schimmel. Latere fabricage processen voor het kunststof microfluidic-apparaat kunnen worden uitgevoerd in een laboratorium dat uitgerust met een UV-verlichting-bron.

Infrarood (IR) spectro-microscopie van levende biologische monsters wordt belemmerd door de absorptie van water in het midden-IR-bereik en door het ontbreken van geschikte microfluidic apparaten. Hier, een protocol voor de fabrikatie van kunststof microfluidic apparaten wordt aangetoond, waar zachte lithografische technieken zijn gebruikt om te sluiten van transparante calciumfluoride (CaF2) weergave-poorten in verband met de observatie kamer (s). De methode is gebaseerd op een replica gieten aanpak, waar een Polydimethylsiloxaan (PDMS) mal is geproduceerd door middel van lithografische standaardprocedures en vervolgens gebruikt als sjabloon voor de productie van een kunststof apparaat. Het kunststof apparaat beschikt over ultraviolet/zichtbaar/infrarood (UV/Vis/IR) - transparante vensters van CaF,2 te voorzien van directe observatie met zichtbaar gemaakt en IR licht. De voordelen van de voorgestelde methode omvatten: een verminderde behoefte aan toegang tot een cleanroom micro-fabricage faciliteit, meerdere weergave-poorten, een gemakkelijke en veelzijdige verbinding met een externe pompsysteem via de plastic lichaam, flexibiliteit van het ontwerp, bv , open/gesloten kanalen configuratie, en de mogelijkheid geavanceerde functies zoals nanoporeuze membranen toe te voegen.

Figuur 6 presenteert de spectra doorlating van een gloednieuwe CaF2 in het venster de patroon helft van het apparaat, en de complete inrichting. Alle drie spectra vertonen uitstekende transparantie tot mid IR met doorlating groter zijn dan 80%.
