<html> 슬랩 겔 전기 영동 (SGE)은 DNA 단편 분리 1 , 2 , 3 , 4 , 5 에 가장 효과적인 방법이며 따라서 생화학 및 생물학적 분석에서 다용도 도구로 간주됩니다 6 , 7 , 8 . 그러나 많은 실험에서 SGE가 다음 네 가지 문제에 의해 제한된다는 것을 나타냅니다. (1) 분리에는 수 시간이 걸리고 심지어 며칠이 걸립니다. (2) 화학 물질 소비가 매우 높다. (3) 복잡한 장치 ( 예 : 2D 전기 영동 세포, 전기 영동 전원 및 겔 이미징 시스템)가 필요합니다. (4) 겔 영상 시스템은 실험이 끝났을 때 분리 된 DNA 조각만을 관찰 할 수있다. 또한, SGE 9 에서 일반적으로 사용되는 ethidium bromide (EtBr)10 = 돌연변이 유발 및 암 유발 11,12 . 따라서 EtBr이 들어있는 젤을 나눠 줄 때는 항상 장갑을 착용해야합니다. 모세관 전기 영동 (CE)은 자동 작동, 짧은 분리 시간, 낮은 소비 등과 같은 SGE에 비해 13 , 14 , 15 , 16 , 17의 다양한 장점을 가지고 있습니다. 그러나 CE 장비는 꽤 비쌉니다. 따라서 이러한 한계를 극복하기 위해 DNA 분리를위한 시스템이 개발되었습니다 ( 그림 1 ). 이러한 시스템은 화학 물질 소비를 크게 줄이고 SGE 실험 시간 (8 분 미만)을 절약 할 수있을뿐만 아니라 스마트 폰을 통해 아가로 오스 겔에서 DNA 분리 과정을 실시간으로 추적 할 수도 있습니다. 이 프로토콜에 설명 된 절차를 따르면, 학생들은SGE 칩을 설계 및 제작하고, 칩에서 아가 로스 겔을 준비하고, 스마트 폰으로 간단한 SGE 시스템을 설치하고 아가로 오스 겔에서 DNA 이동 과정을 기록 할 수 있어야한다.
