Tau werd oorspronkelijk geïdentificeerd als een warmte-stable microtubulus-geassocieerde proteïne, dat mede gezuiverde met tubuline1 was. Tau wordt uitsluitend uitgedrukt in hogere eukaryoten2,-3,4. De belangrijkste functie van tau is controle microtubulus vergadering1,5,6. Het draagt ook bij aan de polymerisatie van microtubuli7axonale vervoer8, veranderingen in axonale diameter9, vorming van neuroma polariteit en neurodegeneratie10. Tau fungeert ook als een eiwit steiger om te controleren enkele signaalroutes. Rat hersenen studies suggereren dat tau is neuron-specifieke en dat het voornamelijk in axonen11 lokaliseert. Omdat tau essentieel voor microtubuli polymerisatie en neuronale ontwikkeling is, was tau hypothetische om te spelen een belangrijke rol in axonale ontwikkeling in het centrale zenuwstelsel; deze hypothese werd later gecontroleerd door in vitro en in vivo experimenten. Naast neuronen, wordt tau uitgedrukt in verschillende non-neuronale cellen, met inbegrip van lever, nieren en spieren cellen12,13. Tau komt ook tot uiting in de menselijke borst, prostaat, colorectale, maag, en alvleesklier kanker cel lijnen en weefsels14,15,16,17,18, 19. tau wordt ook gevonden in opname-lichaam myositis als gedraaide tubulofilaments in integratie organen20.
Tau kan verschillende posttranslationele modificaties uitvoeren. Van alle posttranslationele modificaties is fosforylatie de meest voorkomende. Verhoogde tau fosforylatie vermindert zijn affiniteit voor de microtubuli, ten slotte destabiliserende het cytoskelet. Vijfentachtig fosforylatie sites zijn beschreven in tau-eiwit geïsoleerd uit menselijke Alzheimerpatiënten hersenen weefsels. Deze sites vormen 53% slechts 6% tyrosine residuen21,22,23, serine en threonine van 41%. Tau fosforylatie van invloed is op de lokalisatie, functie, bindende, oplosbaarheid en haar gevoeligheid voor andere posttranslationele modificaties. Meer dan de normale omvang ook tau fosforylatie (of volledig verzadigd met fosfaat groepen) staat bekend als hyperphosphorylation die structurele en functionele kenmerken van de ziekte van Alzheimer24repliceert. Tau onderhoudt goede werking van de axonale microtubuli en zorgt voor een normale neuronale werking onder fysiologische omstandigheden. Hyperphosphorylated tau mislukt echter om een overzichtelijk microtubulus-binding, waardoor neuronale verlies vanwege microtubulus demontage. Normale niveaus van tau fosforylatie zijn vereist voor juiste tau werking, maar tau mislukt om normaal te functioneren als haar karakteristieke fosforylatie niveau wordt gewijzigd en als het hyperphosphorylated25. In de ziekte van Alzheimer en sommige andere leeftijdsgebonden neurodegeneratieve aandoeningen, tau wordt hyperphosphorylated en vormt de gepaarde spiraalvormige filamenten en neurofibrillary klitten26,27. Dus, de methoden voor de bepaling van tau fosforylatie en microtubulus bindend zijn belangrijk.
Colorectaal carcinoom, een vergrijzing-geassocieerde kanker, is dat de derde meest voorkomende diagnose kanker en de derde prominente dood veroorzaken kanker voor mannen én vrouwen28. Colorectal kanker is één van de belangrijkste dood-veroorzaakt kanker in de westerse wereld29. Omdat zowel colorectale kanker en de ziekte van Alzheimer geassocieerd met vergrijzing worden en beide voornamelijk in de ontwikkelde landen waar de mensen genieten van soortgelijke voedingsgewoonten gebeuren, kunnen de twee ziekten een of andere manier worden gecorreleerd. Daarnaast reageren tau-positieve en tau-negatieve kankercellen anders op chemotherapeutische agenten, bijvoorbeeld, paclitaxel16.
Curcumine is een van de belangrijkste derivaten van Curcuma longa, de Indische specerij kurkuma30. Eeuwenlang hebben de Zuid-Aziatische populaties kurkuma in hun dieet op een dagelijkse basis verbruikt. Curcumine is gebruikt voor de behandeling van verschillende ziekten, met inbegrip van colorectal kanker, de ziekte van Alzheimer, diabetes, cystic fibrosis, inflammatoire darmziekte, artritis, hyperlipidemie, atherosclerose en ischemische hartziekte31, 32 , 33 , 34 , 35 , 36 , 37 , 38. lithium kan ook colorectal kankercellen doden of te voorkomen dat hun proliferatie39. Lithium kan ook worden gebruikt voor de behandeling van de ziekte van Alzheimer40 zoals het tau aggregatie vermindert en de hyperphosphorylation voorkomt zoals waargenomen in een transgene muis model41,42,43, 44.
Dit manuscript wil: 1) meet de totale tau en de phospho-tau expressie niveaus in de behandelde cellen; 2) beschrijven een fosfatase assay voor het meten van totale tau fosforylatie; 3) onderzoeken microtubuli-bindende voor tau; en 4) lokaliseren tau door confocale microscopie in colorectal kanker cellijnen behandeld met curcumine of LiCl. Resultaten blijkt dat de behandeling van de cel met curcumine, die een zogenaamd goede chemotherapeutische agent voor darmkanker is, en behandeling met LiCl uitdrukking van beide totale tau kan verminderen en phosphorylated tau in colorectal kanker cellijnen. Deze behandelingen kunnen ook leiden tot nucleaire translocatie van tau. Echter onverwacht, mislukt curcumine om de binding van tau tot microtubuli.

De microtubulus-geassocieerde eiwit tau is een neuronale eiwit dat meestal in axonen lokaliseert. Over het algemeen is tau essentieel voor normale neuronale functioneren omdat het is betrokken bij microtubulus vergadering en stabilisatie. Naast neuronen, wordt tau uitgedrukt in de menselijke borst, prostaat, maag, colorectale, en alvleesklier kanker waar het toont bijna vergelijkbare structuur en soortgelijke functies uitoefent als de neuronale tau. Het bedrag van tau en haar fosforylatie kunt wijzigen van haar functie als stabilisator van microtubuli, en leiden tot de ontwikkeling van de gepaarde spiraalvormige gloeidraden in verschillende neurodegeneratieve aandoeningen, zoals de ziekte van Alzheimer. Vaststelling van de staat van de fosforylatie van tau en de microtubuli-bindende eigenschappen ervan is belangrijk. Daarnaast is onderzoekt de intracellulaire lokalisatie van tau belangrijk in verschillende ziekten. Dit manuscript details standaardprotocollen voor het meten van tau fosforylatie en tau binding aan microtubuli in colorectal kankercellen met of zonder curcumine en LiCl behandeling. Deze behandelingen kunnen worden gebruikt om kanker celproliferatie en ontwikkeling te stoppen. Intracellulaire lokalisatie van tau wordt onderzocht met behulp van immunohistochemistry en confocale microscopie tijdens het gebruik van lage hoeveelheid antilichamen. Deze testen kunnen herhaaldelijk worden gebruikt voor het screenen van verbindingen die tau hyperphosphorylation of microtubulus bindende beïnvloeden. Roman therapeutics gebruikt voor verschillende tauopathies of verwante anticancer agents kunnen mogelijk worden gekarakteriseerd met behulp van deze protocollen.

Uitdrukking van totale tau en phospho-tau werd onderzocht na behandeling van de cellen met verschillende concentraties curcumine of LiCl (Figuur 1).
