Vergeleken met conventionele op basis van MS eiwit kwantificering, iMALDI maakt gebruik van antilichamen te verrijken de analyten en hen te zuiveren van complexe monsters, dus waardoor het mogelijk is te kwantificeren van proteïnen of peptiden in lage concentraties. Een cruciale stap in het iMALDI-protocol is de immuno-verrijking van de doel-peptiden. Voor dit doel, moeten antilichamen met hoge specificiteit en affiniteit worden geselecteerd. Massaspectrometrie uitgegroeid tot een onmisbaar instrument in kwantitatieve proteomics. Massaspectrometrie kan bepalen de massa's van doel proteïnen of peptiden, dus de verkregen analyt signalen kunnen zeer specifiek t.o.v. immunoassay. Twee methoden van de behandeling door ionisering, electrospray en MALDI, worden meestal gebruikt voor het opsporen van de eiwitten en peptiden1,2,3,4. Een grote uitdaging in op basis van MS eiwit kwantificering ligt in het opsporen van lage-overvloed eiwitten in complexe monsters in ng/mL of pg/mL concentraties in de aanwezigheid van eiwitten van hoge-overvloed, en vele kandidaat-proteïne biomarkers gevonden in menselijk plasma zijn binnen dit bereik5. Dit probleem wordt grotendeels veroorzaakt door de inherent breed dynamisch bereik en de complexiteit van het menselijk proteoom-6.
Om te overwinnen van de uitdagingen van deze detectie, immuno-MS-methoden zijn ontwikkeld om te verrijken de doelgroep proteïnen of peptides van de monsteroplossingen op een effen oppervlak, gevolgd door elutie van de analyten en MS meting7,8 , 9 , 10. door immuno-verrijking, de analyten zijn gezuiverd van complexe monsters en daarom de effecten van de ion-onderdrukking van andere moleculen zijn geminimaliseerd. Onder verschillende solide ondersteunt, zijn magnetische kralen momenteel meest gebruikte zoals ze de voordelen van hoge antilichaam bindingscapaciteit en gebruiksgemak hebben. Magnetische kralen met verschillende functionalizations en maten zijn ontwikkeld en gecommercialiseerd voor immunoprecipitation experimenten. Tot op heden heeft immuno-verrijking op parels is geïnterfacet met electrospray ionisatie (ESI) én MALDI-MS voor eiwit en peptide meting. In stabiele isotoop normen en vangen door anti-peptide antilichamen (SISCAPA) technologie, zijn eiwitten in de monsters verteerd, gevolgd door incubatie met antilichaam-gecoate kralen voor immuno-verrijking. In "klassieke" SISCAPA, zijn de vastgelegde proteotypic peptiden geëlueerd van de kralen en gemeten door vloeibare chromatografie-ESI-MS (LC-MS), of door rechtstreekse infusie ESI-meerdere reactie Monitoring-MS (ESI-MRM-MS)11,12. Immuno-verrijking verbeterd de MRM assay gevoeligheid door 3-4 ordes van grootte, de lage ng/mL bereik13te bereiken.
Vergeleken met electrospray-MS, MALDI-MS is sneller, en gaat niet over het reinigen en opnieuw evenwichtsinstelling van LC-kolommen dus er zijn geen problemen met overdracht en verontreiniging, waardoor het meer geschikt voor high-throughput onderzoek14. Immuno-MALDI technologie is ontwikkeld in ons laboratorium immuno-verrijking combineren met MALDI-MS voor gevoelige en specifieke kwantificering van peptides en proteïnen (gebaseerd op de kwantificatie van proteotypic peptiden)15,16 ,17. Na immuno-verrijking, de kralen worden gestort op een MALDI doel plaat, de oplossing van de matrix is toegevoegd aan de kralen en de plaat is klaar voor analyse door een MALDI-TOF-MS na het drogen. Elutie van de peptides van de kralen wordt niet uitgevoerd als een afzonderlijke stap, maar affiniteit-gebonden analyten worden geëlueerd door de MALDI matrix oplossing wanneer het wordt toegevoegd aan de parel vlekken, daardoor vereenvoudiging van de bereiding van de monsters en het minimaliseren van monster verlies. De iMALDI-technologie is toegepast in een verscheidenheid van toepassingen18,19, en onlangs is geautomatiseerd en gebruikt voor het meten van angiotensine I (Ang ik) voor het bepalen van de plasma renine activiteit (PRA)20. Dit protocol zal de procedure gebruikt voor het uitvoeren van een geautomatiseerde iMALDI assay aantonen. De PRA test nemen als voorbeeld, Inter dag CVs van minder dan 10% hebben bereikt door middel van automatisering, met de mogelijkheid voor het meten van 744 monsters per dag20.
De iMALDI PRA assay aangetoond in dit manuscript vereist geen eiwit spijsvertering, als de doelmolecule (Ang ik) is een peptide met een moleculair gewicht van 1295.7 Da. In andere testen waar eiwit spijsvertering noodzakelijk is en een peptide wordt gebruikt als de surrogaat voor het intact eiwit, dient de geselecteerde peptide voor iMALDI uniek en specifiek voor de proteïne van de doelstelling, met een massa van meer dan 800 Da dus dat zij gemakkelijk kan worden onderscheiden van de c chemische lawaai van de MALDI matrix oplossing. Anti-peptide antilichamen zijn nodig voor de immuno-verrijking van de peptiden. Het protocol voor een bepaling van de iMALDI meten van PRA bestaat uit vier stappen: 1) generatie van Ang ik in menselijk plasma; 2) immuno-verrijking van Ang mij using antilichaam beklede kralen; 3) overdracht van parels aan een MALDI doel plaat en toevoegen matrix oplossing; en 4) signaal overname door een MALDI-TOF-MS en data analyse-20.

peptide and protein quantification using automated immuno-maldi (imaldi)

De Spectrometrie van de massa (MS) is één van de meest gebruikte technologieën voor het kwantificeren van de eiwitten in complexe monsters, met uitstekende assay specificiteit als gevolg van de directe detectie van de verhouding van de massa-naar-charge van elke doelmolecule. Echter, op basis van MS proteomics, net als de meeste andere analytische technieken, heeft een voorkeur voor het meten van hoge-overvloed analyten, dus het is uitdagend om detectie beperkt voor lage ng/mL of pg/mL in complexe monsters, en dit is het concentratiebereik voor velen ziekte-relevante eiwitten in biofluids zoals menselijk plasma. Om te helpen bij de opsporing van lage-overvloed analyten, is immuno-verrijking geïntegreerd in de bepaling te concentreren en te zuiveren van de analyt voor MS meting, aanzienlijke verbetering van de gevoeligheid van de test. In dit werk, is de immuno - Matrix-Assisted Laser desorptie/ionisatie (iMALDI) technologie gepresenteerd voor de kwantificering van de eiwitten en peptiden in biofluids, op basis van immuno-verrijking op parels, gevolgd door meting van de MALDI-MS zonder voorafgaande elutie. De anti-peptide antilichamen zijn matiemaatschappij op magnetische kralen en geïncubeerd met monsters. Na het wassen, de kralen zijn rechtstreeks overgezet naar een MALDI doel plaat, en de signalen door een MALDI-Time van de vlucht (MALDI-TOF) instrument worden gemeten nadat de matrix-oplossing is vereffend met de kralen. De voorbereiding van de voorbeeldprocedure wordt vereenvoudigd ten opzichte van andere immuno-MS testen, en de MALDI-meting is snel. De hele monsterverwerking is geautomatiseerd met een vloeistof handling systeem, met verbeterde assay reproduceerbaarheid en een snellere doorgifte. In dit artikel, de bepaling van de iMALDI wordt gebruikt voor het bepalen van het peptide angiotensine I (Ang ik) concentratie in plasma, dat klinisch als uitlezing van plasma renine activiteit wordt gebruikt voor de screening van primaire aldosteronisme (PA).

Doel peptiden (endogene peptiden of peptiden uit verteerd eiwit) zijn verrijkt op anti-peptide magnetische kralen, en vervolgens de kralen zijn overgebracht naar een doel-plaat voor MALDI meting.
