Par rapport à la quantification des protéines classiques basées sur, iMALDI utilise les anticorps d’enrichir l’analyser et de les purifier des échantillons complexes, rendant donc impossible de quantifier les protéines ou les peptides à de faibles concentrations. Une étape cruciale dans le protocole d’iMALDI est l’immuno-enrichissement des peptides cible. À cette fin, avec une grande spécificité et l’affinité des anticorps doivent être sélectionnés. Spectrométrie de masse est devenu un outil indispensable en protéomique quantitative. Spectrométrie de masse permet de déterminer les masses des peptides ou des protéines cibles, donc les signaux analyte obtenus peuvent être très spécifiques par rapport aux tests immunologiques. Deux méthodes d’ionisation electrospray et MALDI, sont plus couramment utilisés pour la détection des protéines et des peptides1,2,3,4. Un défi majeur dans la quantification des protéines basée sur MS réside dans la détection des protéines de faible abondance dans des échantillons complexes à des concentrations ng/mL ou pg/mL en présence de protéines de haute-abondance, et nombreux biomarqueurs protéiques de candidat dans le plasma humain sont au sein de cette gamme5. Ce problème est dû en grande partie par l’intrinsèquement une plage dynamique étendue et la complexité du protéome humain6.
Pour surmonter ces difficultés de détection, les méthodes immuno-SM ont été développés afin d’enrichir les protéines cibles ou des peptides dans les solutions d’échantillon sur une surface solide, suivie d’élution de l’analyser et MS mesure7,8 , 9 , 10. par immuno-enrichissement sans cause, les analytes sont purifiées à partir des échantillons complexes et donc les effets de la suppression des ions d’autres molécules sont réduites au minimum. Parmi les différents supports solides, billes magnétiques sont actuellement plus largement utilisés car ils ont les avantages de la capacité de liaison anticorps élevés et facilité de manipulation. Billes magnétiques avec différentes fonctionnalisation et tailles ont été développés et commercialisés pour des expériences d’immunoprécipitation. A ce jour, immuno-enrichissement sur perles a été interfacé avec ionisation par électrospray (ESI) et MALDI-MS pour la mesure de protéines et de peptides. Dans les normes des isotopes stables et la capture par la technologie anticorps anti-peptide (SISCAPA), les protéines dans les échantillons sont digérées, suivie d’une incubation avec perles revêtus d’anticorps pour immuno-enrichissement. Dans SISCAPA « classique », les peptides proteotypic capturés sont éluées de talons et mesurées par Liquid Chromatography-ESI-MS (LC-MS), ou par perfusion directe ESI-Multiple réaction Monitoring-MS (MRM-ESI-MS)11,12. Immuno-enrichissement amélioré la sensibilité de l’éssai MRM de 3-4 ordres de grandeur, atteignant le rang faible ng/mL13.
Par rapport à electrospray-MS, MALDI-MS est plus rapide et n’implique pas le nettoyage et la rééquilibration des colonnes LC donc il n’y a aucun problème de report et contamination, plus adapté aux études haut débit14. Immuno-MALDI technologie a été développée dans notre laboratoire à combiner immuno-enrichissement avec MALDI-MS pour la quantification sensible et spécifique des peptides et des protéines (basés sur le dosage des peptides proteotypic)15,16 ,,17. Après enrichissement immuno, les perles sont déposés sur une plaque de mire MALDI, la solution de matrice est ajoutée aux billes et la plaque est prête pour l’analyse par un MALDI-TOF-MS après séchage. L’élution des peptides de la perle n’est pas exécutée comme une étape distincte, mais affinité lié aux analytes sont éluées par la solution de matrice MALDI lorsqu’il est ajouté aux spots perle, ce qui simplifie la préparation de l’échantillon et réduisant au minimum la perte de l’échantillon. La technologie iMALDI a été appliquée dans une variété d’applications18,19et récemment a été automatisée et utilisée pour la mesure de l’angiotensine I (Ang je) pour la détermination de l’activité (PRA) rénine plasmatique20. Ce protocole fera la démonstration de la procédure utilisée pour effectuer un test automatique iMALDI. Prendre le dosage de la PRA à titre d’exemple, la inter-journée CVs de moins de 10 % ont été obtenus grâce à l’automatisation, la capacité de mesurer 744 échantillons par jour20.
Le test PRA iMALDI démontré dans ce manuscrit ne nécessite pas de digestion des protéines, comme la molécule cible (Ang I) est un peptide avec un poids moléculaire de 1295.7 Da. Lors d’autres essais où la digestion de protéines est nécessaire et un peptide est utilisé comme substitut pour la protéine intacte, le peptide sélectionné pour iMALDI doit être unique et spécifique à la protéine cible, avec une masse plus de 800 Da alors qu’elle peut être aisément distinguée de la c bruit chimique de la solution de matrice MALDI. Anticorps anti-peptides sont requis pour l’immuno-enrichissement des peptides. Le protocole de test iMALDI PRA de mesure se compose de quatre étapes : 1) génération de Ang j’ai dans le plasma humain ; 2) immuno-enrichissement d’Ang I à l’aide de petites perles revêtus d’anticorps ; 3) transfert de perles à une plaque de mire MALDI et ajout solution de matrice ; et 4) acquisition de signal par une analyse de MALDI-TOF-MS et données20.

peptide and protein quantification using automated immuno-maldi (imaldi)

Spectrométrie de masse (MS) est une des technologies plus couramment utilisées pour quantifier les protéines dans des échantillons complexes, avec dosage excellente spécificité à la suite de la détection directe du ratio masse à charge de chaque molécule cible. Cependant, protéomique axée sur les MS, comme la plupart des autres techniques analytiques, a un biais de mesure haute-abondance analytes, donc il est difficile d’atteindre les seuils de détection de basse ng/mL ou pg/mL chez des échantillons complexes et c’est la plage de concentration pour beaucoup maladie-les protéines liquides comme le plasma humain. Pour aider à la détection d’analytes de faible abondance, immuno-enrichissement a été intégré dans l’essai de se concentrer et de purifier l’analyte avant MS mesure, améliorer significativement la sensibilité. Dans ce travail, la technologie immuno - Matrix-Assisted Laser désorption-ionisation (iMALDI) est présentée pour la quantification des protéines et des peptides dans les liquides, issue des immuno-enrichissement sur perles, suivie par MALDI-MS mesure sans préalable d’élution. Les anticorps anti-peptides sont fonctionnalisés sur billes magnétiques et incubées avec les échantillons. Après le lavage, les perles sont directement transférées sur une plaque de mire MALDI, et les signaux sont mesurés par un MALDI-temps des instruments de vol (MALDI-TOF) après que la solution de la matrice a été appliquée aux talons. La procédure de préparation d’échantillon est simplifiée par rapport aux autres tests immuno-MS, et la mesure de la MALDI est rapide. La préparation de l’ensemble de l’échantillon est automatisée avec un liquide de système, de la manutention avec la reproductibilité du dosage améliorée et un débit plus élevé. Dans cet article, le test d’iMALDI est utilisé pour déterminer l’angiotensine peptide j’ai (Ang I) concentration dans le plasma, qui est utilisé cliniquement comme indicateur de l’activité rénine plasmatique pour la projection d’hyperaldostéronisme primaire (PA).

Une procédure automatisée d’iMALDI pour mesurer les Ang I est montré dans la Figure 1.
