Massespektrometri har blitt et uunnværlig verktøy i kvantitative Proteomikk. Massespektrometri kan bestemme massene av målet proteiner eller peptider, innhentet analytt signalene kan derfor være svært spesifikke forhold til immunanalyser. To ionisering metoder, electrospray og MALDI, brukes vanligvis for å oppdage proteiner og peptider1,2,3,4. En stor utfordring i baserte protein kvantifisering ligger i deteksjon av lav-overflod proteiner i komplekse eksempler på ng/mL eller pg/mL konsentrasjonen i nærvær av høy-overflod proteiner, og mange kandidat protein biomarkers i humant plasma er innenfor dette området5. Dette problemet skyldes i stor grad den iboende bredt dynamisk område og kompleksiteten av menneskelig proteom6.
For å overvinne disse oppdagelsen utfordringene, immuno-MS metoder har blitt utviklet for å berike målet proteiner eller peptider fra eksempel løsninger på en solid overflate, etterfulgt av elueringsrør analytter og MS måling7,8 , 9 , 10. gjennom immuno-berikelse, analytter er renset fra komplekse eksempler og derfor ion-undertrykking effekten fra andre molekyler er minimert. Magnetiske perler med forskjellige functionalizations og størrelser er utviklet og kommersialisert for immunoprecipitation eksperimenter. Hittil har immuno-berikelse på perler, tilkobles med både electrospray ionization (ESI) og MALDI-MS for protein og peptid måling. Stabil isotop standarder og fangst av anti-peptid antistoffer (SISCAPA) teknologi, er proteiner i prøvene fordøyd, etterfulgt av inkubasjon med antistoff perler for immuno-berikelse. I "klassisk" SISCAPA, er fanget proteotypic peptidene elut fra perler, og målt av flytende kromatografi-ESI-MULTIPLE Sclerosis (LC-MS), eller ved direkte infusjon ESI-flere reaksjon overvåking-MS (ESI-MRM-MS)11,12. Sammenlignet med electrospray-MULTIPLE Sclerosis, MALDI-MS er raskere, og innebærer ikke rengjøring og re balanse av LC kolonner slik at det ikke er noen carryover og forurensning problemer, noe som gjør det mer egnet for høy gjennomstrømming studier14. Immuno-MALDI teknologi har blitt utviklet i vårt laboratorium kombinere immuno-berikelse med MALDI-MS for sensitive og bestemt kvantifisering av peptider og proteiner (basert på kvantifisering av proteotypic peptider)15,16 ,17. IMALDI PRA analysen i dette manuskriptet krever ikke protein fordøyelse, som mål molekylet (Ang jeg) er et peptid med en molekylvekt på 1295.7 Da. peptide and protein quantification using automated immuno-maldi (imaldi)

Massespektrometri (MS) er en av de mest brukte teknologiene for kvantifisere proteiner i komplekse prøver, med utmerket analysen spesifisitet som følge av direkte deteksjon av masse-til-lade forholdet mellom hvert mål molekylet. Men baserte Proteomikk, som de fleste andre analytiske teknikker, har en bias mot måle høy-overflod analytter, så det er utfordrende for å oppnå oppdagelsen begrenser lav ng/ml eller pg/mL i komplekse eksempler, og dette er området konsentrasjon for mange sykdom relevante proteiner i biofluids som humant plasma. For å bistå i deteksjon av lav-overflod analytter, har immuno-berikelse blitt integrert i analysen å konsentrere seg og rense analytt før MS måling, betydelig bedre analysen følsomhet. I dette arbeidet, er negative Matrix-Assisted Laser desorpsjon/Ionization (iMALDI) teknologien presentert for kvantifisering av proteiner og peptider i biofluids, basert på immuno-berikelse på perler, etterfulgt av MALDI-MS måling uten forutgående elueringsrør. Anti-peptid antistoffer er functionalized på magnetiske perler, og inkubert med eksempler. Etter vasking, perler overføres direkte til en MALDI mål plate, og signalene er målt ved en MALDI-tid på fly (MALDI-TOF) instrument etter matrix løsningen brukes på perlene. Eksempel forberedelse prosedyren er forenklet sammenlignet med andre immuno-MS analyser, og MALDI målingen er rask. Hele utvalget forberedelsene er automatisert med en væske håndteringssystem, med forbedret analysen reproduserbarhet og høyere gjennomstrømming.
