Jämfört med konventionella MS-baserad protein kvantifiering, använder iMALDI antikroppar för att berika analyter och rena dem från komplexa prover, därför gör det möjligt att kvantifiera proteiner eller peptider vid låga koncentrationer. Ett avgörande steg i protokollet iMALDI är immuno-anrikningen av målet peptiderna. För detta ändamål bör antikroppar med hög specificitet och affinitet väljas. Masspektrometri har blivit ett oumbärligt verktyg i kvantitativ proteomik. Masspektrometri kan bestämma massan av målet proteiner eller peptider, därför de erhållna analyt signalerna kan vara mycket specifik jämfört immunanalyser. Två jonisering metoder, elektrospray och MALDI, används oftast för att detektera proteiner och peptider1,2,3,4. En stor utmaning i MS-baserad protein kvantifiering ligger i detektion av låg-överflöd proteiner i komplexa prover vid ng/mL eller pg/mL koncentrationer i närvaro av hög-överflöd proteiner, och många kandidat protein biomarkörer hittade i human plasma är inom detta spänner5. Detta problem orsakas till stor del av sin natur brett dynamiskt omfång och komplexitet av mänskliga proteomet6.
För att övervinna utmaningarna upptäckt, immuno-MS metoder har utvecklats för att berika den målet proteiner eller peptider från provet lösningarna på en fast yta, följt av eluering av analyter och MS mätning7,8 , 9 , 10. genom immuno-anrikning, analyterna renas från komplexa prover och därför ion-suppression effekterna från andra molekyler minimeras. Bland olika fasta stöd används magnetiska pärlor för närvarande mest eftersom de har fördelarna med hög antikropp bindande kapacitet och enkel hantering. Magnetiska pärlor med olika functionalizations och storlekar har utvecklats och kommersialiseras för immunoprecipitation experiment. Hittills har immuno-anrikning på pärlor har varit kopplats ihop med både elektrospray jonisering (ESI) och MALDI-MS för protein och peptid mätning. I stabil isotop standarder och fånga av anti peptid antikroppar (SISCAPA) teknik rötas proteiner i proven, följt av inkubation med antikroppsbelagda pärlor för immuno-anrikning. I ”klassisk” SISCAPA, är de tagna proteotypic peptiderna elueras från pärlor och mäts av Liquid Chromatography-ESI-MS (LC-MS), eller genom direkt infusion ESI-flera reaktion övervakning-MS (ESI-MRM-MS)11,12. Immuno-anrikning förbättrades MRM analysens känslighet med 3-4 storleksordningar, att nå den låga ng/mL range13.
Jämfört med elektrospray-MS, MALDI-MS är snabbare och omfattar inte rengöring och förnyad Jämviktstiden LC kolumner så finns ingen överföring och kontaminering frågor, vilket gör den mer lämplig för hög genomströmning studier14. Immuno-MALDI teknik har utvecklats i vårt laboratorium att kombinera immuno-anrikning med VITEK-MS för känsligt och specifikt kvantifiering av peptider och proteiner (baserat på kvantitering av proteotypic peptider)15,16 ,17. Efter immuno-anrikning, pärlorna är deponeras på en MALDI målplatta, modellösning läggs till pärlor och plattan är redo för analys av en MALDI-TOF-MS efter torkning. Eluering av peptiderna från pärlorna utförs inte som ett separat steg, men affinitet-bundna analyter elueras av MALDI modellösning när det läggs till pärla fläckar, därigenom förenkla provpreparering och minimera prov förlust. Den iMALDI tekniken har använts i en mängd tillämpningar18,19, och nyligen har automatiserat och används för att mäta Angiotensin jag (Ang jag) för att fastställa plasma renin aktivitet (PRA)20. Detta protokoll kommer att demonstrera det förfarande som används för att utföra en automatiserad iMALDI analys. Tar PRA analysen som ett exempel, mellan dag CVs på mindre än 10% har uppnåtts genom automatisering, med möjlighet att mäta 744 prover per dag20.
IMALDI PRA analysen visat i detta manuskript kräver inte protein matsmältningen, som målmolekylen (Ang jag) är en peptid med en molekylvikt av 1295.7 Da. I andra analyser där protein matsmältningen är nödvändigt och en peptid används som surrogat för intakt protein, bör den valda peptiden för iMALDI unika och specifika för målproteinet, med en massa över 800 Da så att det kan lätt skiljas från c hemical buller från MALDI modellösning. Anti peptid antikroppar krävs för immuno-anrikningen av peptiderna. Protokollet för ett iMALDI test mäta PRA består av fyra steg: 1) generation av Ang jag i human plasma. (2) immuno-anrikning av Ang jag använder antikroppsbelagda pärlor; (3) överföring av pärlor till en MALDI målplatta och lägga modellösning; och 4) signal förvärv genom en MALDI-TOF-MS och data analys20.

peptide and protein quantification using automated immuno-maldi (imaldi)

Masspektrometri (MS) är en av de vanligaste teknikerna för att kvantifiera proteiner i komplexa prover, med utmärkta analysens specificitet till följd av direkt påvisande av massa-till-avgift förhållandet mellan varje målmolekylen. Men MS-baserad proteomik, som de flesta andra analystekniker, har en inriktning mot mäta hög-överflöd analyter, så det svårt för att uppnå upptäckt begränsar låga ng/ml eller pg/mL i komplexa prover, och detta är koncentrationsintervall för många sjukdom-relevanta proteiner i kroppsvätskor såsom humanplasma. För att hjälpa detektion av låg-överflöd analyter, har immuno-anrikning integrerats i analysen att koncentrera sig och rena analyten före MS mätning, avsevärt förbättra analysens känslighet. I detta arbete, är immuno - laser Laser Desorption/jonisering (iMALDI) tekniken presenteras för kvantifiering av proteiner och peptider i kroppsvätskor, baserat på immuno-anrikning på pärlor, följt av MALDI-MS mätning utan föregående eluering. Anti peptid antikropparna är functionalized på magnetiska pärlor, och inkuberas med prover. Efter tvätt, pärlorna överförs direkt till en MALDI målplatta och signaler mäts av en MALDI-tid på flyg (MALDI-TOF) instrumentet efter modellösning har tillämpats på pärlorna. Prov förberedelse förfarandet förenklas jämfört med andra immuno-MS analyser och MALDI mätningen är snabb. Hela provberedningen är automatiserad med en vätskehanterande system, med förbättrad analysreproducerbarhet och högre genomströmning.
