Recente cumulatieve bewijzen over verschillende neuropsychiatrische stoornissen zoals de ziekte van Parkinson en schizofrenie stellen sterk voor dat hun pathofysiologie is gebaseerd op een kritische disfunctie van verlengde neuronale circuits die vaak corticale en subcortische structuren 1 , 2 , 3 betreffen . Volgens deze theorie ontstaan ​​de klinische manifestaties van de ziekten als gevolg van een verminderde informatieverwerkingscapaciteit van een netwerk van cellen in plaats van enkele cellen of specifieke neuronale elementen 1 , 2 , 3 . Om het begrip van deze complexe groep neuropsychiatrische ziekten te vergroten en nieuwe behandelingsopties te vinden, is het verplicht om de neuronale dynamiek van deze gedwongen netwerken in menselijke patiënten en in diermodellen in detail te karakteriseren. Een uitstekendMethode om grootschalige netwerken in levende onderwerpen te bestuderen, is elektrofysiologische opnames van extracellulaire potenties 4 . Met behulp van deze methode is het mogelijk om lokale veldpotentialen (LFP&#39;s) gelijktijdig te beoordelen, die voornamelijk de temporale summatie vormen van excitatoire en remmende postsynaptische stromingen en multi-unit activiteit (MUA), die wordt gegenereerd door presynaptische mogelijkheden 5 . De opname van extracellulaire potenties heeft meerdere voordelen ten opzichte van alternatieve methoden om netwerken te bestuderen, bijvoorbeeld functionele magnetische resonantiebeeldvorming en calciumbeeldvorming, omdat het een hogere temporale en ruimtelijke resolutie biedt en omdat het niet afhankelijk is van genetisch gemanipuleerde organismen 5 . Het gebruik van extracellulaire in vivo opnames is echter beperkt omdat het een invasieve techniek is die niet universeel toegepast kan worden. In vivo elektrofysiologische recOrdeningen kunnen zowel wakker als in verdovende dieren worden uitgevoerd 6 . Beide methoden worden gepaard gegaan met specifieke voor- en nadelen. Studies in wakker dieren maken het mogelijk om hersensignalen op te nemen tijdens de uitvoering van bepaalde gedragstaken, maar zijn gevoelig voor bewegingsgerelateerde en andere artefacten 7 , 8 . Opnames in verdovende dieren daarentegen bieden de mogelijkheid om LFP&#39;s en MUA te beoordelen met een minimum van artefacten bij zeer gedefinieerde corticale synchronisatiestaten, maar de resultaten verschillen ook tot op zekere hoogte in wat wakker is in vakken 9 , 10 , 11 . In de afgelopen jaren is aangetoond dat de bemonstering van LFP&#39;s bijzonder nuttig is om pathologische veranderingen van netwerkactiviteit te definiëren. Een prominente voorbeeld hiervan is onderzoek naar de pathofysiologie van PD in de menselijke patiëntS en diermodellen van de ziekte, waaruit bleek dat verbeterde bètoscillaties in de cortico-basale ganglia-lus gekoppeld zijn aan parkinsonische motorische symptomen 12 , 13 . Als gevolg van deze onderzoekslijn wordt momenteel onderzocht of bèta-oscillaties gebruikt kunnen worden als een online-feedback biomarker voor de gesloten-loop diepe hersenstimulatie 14 , 15 . In de onderhavige studie wordt een gedetailleerde beschrijving gegeven van acute multi-site in vivo elektrofysiologische opnames van LFP&#39;s en MUA bij ratten die met urethaan zijn verdoofd. Er wordt aangetoond hoe een volledig netwerk, zoals de hyperdirecte cortico-basale ganglia-route, elektrofysiologisch kan worden gekenmerkt door gebruik te maken van standaard en aangepaste elektroden en hoe deze elektroden kunnen worden gebouwd. Het wordt vooral benadrukt hoe een nauwkeurige targeting van basale ganglia kernen kan worden bereikt door coMbining stereotactische operatie samen met de online registratie van MUA&#39;s.

acute in vivo electrophysiological recordings of local field potentials and multi-unit activity from the hyperdirect pathway in anesthetized rats

Convergeert bewijs blijkt dat veel neuropsychiatrische ziekten moeten worden begrepen als aandoeningen van grootschalige neuronale netwerken. Om de pathofysiologische basis van deze ziekten beter te begrijpen, is het noodzakelijk om precies te karakteriseren op welke wijze de verwerking van informatie verstoord raakt tussen de verschillende neuronale delen van het circuit. Met behulp van extracellulaire in vivo elektrofysiologische opnamen is het mogelijk om neuronale activiteit nauwkeurig te definiëren binnen een neuronaal netwerk. De toepassing van deze methode heeft meerdere voordelen ten opzichte van alternatieve technieken, zoals functionele magnetische resonantiebeeldvorming en calciumbeeldvorming, omdat het een unieke temporale en ruimtelijke resolutie mogelijk maakt en niet vertrouwt op genetisch gemanipuleerde organismen. Het gebruik van extracellulaire in vivo opnames is echter beperkt omdat het een invasieve techniek is die niet universeel toegepast kan worden. In dit artikel wordt een eenvoudige en makkelijk te gebruiken methode gepresenteerd wMet welke het mogelijk is om extracellulaire potenties tegelijkertijd op te nemen, zoals lokale veldpotenties en multiunitactiviteit op meerdere sites van een netwerk. Het is gedetailleerd hoe een precieze targeting van subcortische kernen kan worden bereikt met behulp van een combinatie van stereotactische chirurgie en online analyse van multi-unit opnames. Zo wordt aangetoond hoe een compleet netwerk, zoals de hyperdirecte cortico-basale ganglia-lus, kan worden onderzocht in verdovende dieren in vivo .

Met de hierin gebruikte opnameelektroden is het mogelijk om LFP&#39;s uit de primaire motorcortex, de subthalamische kern en de substantia nigra pars reticulata en MUA van de STN en SNr te proeven. Aanvankelijk worden LFP&#39;s en multi-unit activiteit samen in een breedband signaal opgenomen.
