Die jüngsten kumulativen Evidenz bei verschiedenen neuropsychiatrischen Erkrankungen wie Parkinson&#39;sche Krankheit (PD) und Schizophrenie deutet stark darauf hin, dass ihre Pathophysiologie auf einer kritischen Dysfunktion von erweiterten neuronalen Kreisen beruht, die oft kortikale und subkortikale Strukturen 1 , 2 , 3 beinhalten . Nach dieser Theorie entstehen die klinischen Manifestationen der Erkrankungen als Folge einer beeinträchtigten Informationsverarbeitungskapazität eines Netzwerkes von Zellen anstelle von einzelnen Zellen oder spezifischen neuronalen Elementen 1 , 2 , 3 . Um das Verständnis dieser komplexen Gruppe neuropsychiatrischer Erkrankungen zu verbessern und neue Behandlungsmöglichkeiten zu finden, ist es notwendig, die neuronale Dynamik jener ungeordneten Netzwerke bei menschlichen Patienten und in Tiermodellen im Detail zu charakterisieren. Eine excellEnt-Methode, um große Netzwerke in lebenden Probanden zu untersuchen, ist mehrfache elektrophysiologische Aufzeichnungen von extrazellulären Potentialen 4 . Mit dieser Methode ist es möglich, lokale Feldpotentiale (LFPs) gleichzeitig zu beurteilen, die in erster Linie die zeitliche Summierung von exzitatorischen und inhibitorischen postsynaptischen Strömen und Multi-Unit-Aktivität (MUA) darstellen, die durch präsynaptische Potentiale erzeugt wird. Die Aufzeichnung von extrazellulären Potentialen hat gegenüber alternativen Methoden, um Netzwerke zu untersuchen, zB funktionelle Magnetresonanztomographie und Calciumbildgebung, weil sie eine höhere zeitliche und räumliche Auflösung bietet und weil sie nicht auf gentechnisch veränderte Organismen angewiesen ist. Allerdings ist die Verwendung von extrazellulären In-vivo- Aufnahmen begrenzt, da es sich um eine invasive Technik handelt, die nicht universell angewendet werden kann. In vivo elektrophysiologische recOrden können sowohl bei wach als auch bei anästhesierten Tieren durchgeführt werden 6 . Beide Methoden werden von spezifischen Vor- und Nachteilen begleitet. Studien in wachen Tieren erlauben die Aufzeichnung von Hirnsignalen während der Durchführung von definierten Verhaltensaufgaben, sind aber anfällig für bewegungsbezogene und andere Artefakte 7 , 8 . Aufzeichnungen in anästhesierten Tieren bieten andererseits die Möglichkeit, LFPs und MUA mit einem Minimum an Artefakten in hoch definierten kortikalen Synchronisationszuständen zu beurteilen, aber die Ergebnisse unterscheiden sich auch in gewissem Maße von dem, was in den wachenden Fächern 9 , 10 , 11 gefunden werden kann . In den letzten Jahren wurde gezeigt, dass die Probenahme von LFPs besonders nützlich ist, um pathologische Veränderungen der Netzwerkaktivität abzugrenzen. Ein prominentes Beispiel dafür ist die Erforschung der Pathophysiologie der PD bei menschlichen PatientenS und Tiermodelle der Krankheit, wo es gezeigt werden konnte, dass erhöhte Beta-Oszillationen in der Cortico-Basal-Ganglien-Schleife mit Parkinson-Motorsymptomen 12 , 13 verknüpft sind. Als Konsequenz dieser Forschungsrichtung wird derzeit untersucht, ob Beta-Oszillationen als Online-Feedback-Biomarker für geschlossene tiefe Hirnstimulation 14 , 15 verwendet werden könnten. In der vorliegenden Studie wird eine detaillierte Beschreibung der akuten mehrstufigen in vivo elektrophysiologischen Aufzeichnungen von LFPs und MUA bei Ratten, die mit Urethan betäubt sind, bereitgestellt. Es wird gezeigt, wie ein komplettes Netzwerk, wie der hyperdirekte Cortico-Basalganglienweg, elektrophysiologisch mit Standard- und kundenspezifischen Elektroden charakterisiert werden kann und wie diese Elektroden aufgebaut werden können. Es wird besonders hervorgehoben, wie eine präzise Ausrichtung von Basalganglienkernen durch Co erreicht werden kannMbining stereotaktische Chirurgie zusammen mit der Online-Registrierung von MUAs.

Konvergente Beweise zeigen, dass viele neuropsychiatrische Erkrankungen als Störungen von großräumigen neuronalen Netzwerken verstanden werden sollten. Um die pathophysiologische Grundlagen dieser Erkrankungen besser zu verstehen, ist es notwendig, genau zu charakterisieren, in welcher Weise die Verarbeitung von Informationen zwischen den verschiedenen neuronalen Teilen der Schaltung gestört wird. Unter Verwendung von extrazellulären in vivo elektrophysiologischen Aufnahmen ist es möglich, die neuronale Aktivität innerhalb eines neuronalen Netzwerks genau abzugrenzen. Die Anwendung dieses Verfahrens hat gegenüber alternativen Techniken, zB funktionelle Magnetresonanztomographie und Calciumabbildung, mehrere Vorteile, da es eine einzigartige zeitliche und räumliche Auflösung ermöglicht und sich nicht auf gentechnisch veränderte Organismen stützt. Allerdings ist die Verwendung von extrazellulären In-vivo- Aufnahmen begrenzt, da es sich um eine invasive Technik handelt, die nicht universell angewendet werden kann. In diesem Artikel wird eine einfache und einfach zu bedienende Methode dargestelltMit denen es möglich ist, gleichzeitig extrazelluläre Potentiale wie lokale Feldpotentiale und Multifunktionsaktivitäten an mehreren Standorten eines Netzwerks aufzuzeichnen. Es wird detailliert, wie eine präzise Ausrichtung von subkortikalen Kernen durch eine Kombination von stereotaktischer Chirurgie und Online-Analyse von Multi-Unit-Aufnahmen erreicht werden kann. So wird gezeigt, wie ein komplettes Netzwerk wie die hyperdirekte Cortico-Basal-Ganglienschleife in anästhesierten Tieren in vivo untersucht werden kann .

Mit den hierin verwendeten Aufzeichnungselektroden ist es möglich, LFPs aus dem primären motorischen Kortex, dem subthalamischen Kern und den substantia nigra pars reticulata und MUA aus dem STN und SNr zu probieren. Anfänglich werden LFPs und Multi-Unit-Aktivitäten zusammen in einem Breitband-Signal aufgezeichnet.
