Biofilm eksistere og vokse inden for en meget hydreret tilstand. Dette er en stor udfordring i at bruge vakuum-baserede overflade analyseteknikker såsom time of flight sekundære ion massespektrometri (ToF-SIMS) på grund af vanskeligheden ved at studere flygtige væsker i vakuum. Som et resultat, har vakuum-baserede overflade analyseteknikker været begrænset næsten udelukkende til at studere biofilm prøver på kun deres tørret tilstand. Bakterielle biofilm er overflade-associerede Fællesskaber, der studeres langt for at forstå deres egen-producerede ekstracellulære polymere stoffer (EPS) og deres roller i miljøet Mikrobiologi. Denne undersøgelse beskriver en metode til at dyrke biofilm vedhæftet fil system til analyse på flydende vakuum Interface (SALVI) og opnå i situ kemiske kortlægning af en levende biofilm ved time of flight sekundære ion massespektrometri (ToF-SIMS). Dette gøres gennem dyrkning af bakterier både uden for og inden for SALVI kanalen med vores specialiserede opsætning samt gennem optiske billeddannelse teknikker til at opdage biofilm tilstedeværelse og tykkelse før ToF-SIMS analyse. Vores resultater viser karakteristiske toppene af Shewanella biofilm i sin naturlige hydreret tilstand, fremhæve ved sin lokaliserede vand klyngemiljø samt EPS-fragmenter, der er drastisk anderledes fra den samme biofilm dehydreret stat. Disse resultater viser gennembrud evne til SALVI, der giver mulighed for i situ biofilm imaging med en vakuum-baserede kemiske imaging instrument.

Disse repræsentative resultater tjene til at vise hvordan den kemiske profil af den vedlagte biofilm kan identificeres og fortolket, som opnås gennem ToF-SIMS. Efter plotte massespektre fra ToF-SIMS dataopsamling, fremhævet kortvarigt i afsnittet procedurer bør peak identifikation gennemføres for at tildele hver respektive m/z værdi identiteter.
