Zaadknop klaring is een handige methode voor het opsporen van de celdeling en morfologie beoordelen tijdens de embryogenese Arabidopsis; met behulp van deze techniek, kunnen direct onder DIC microscopie5,12embryo's worden waargenomen. Embryogenese is de vroegste gebeurtenis in de ontwikkeling van hogere planten. Een volwassen embryo vormt van een zygote door celdeling- en differentiatie onder strenge controle van de genetische1,2. Arabidopsis embryo's zijn een nuttig model voor de controle op de voedselproductie studeren omdat de volgorde van de celdelingen tijdens de embryogenese een verwachte patroon3,4 volgt. Een embryo van Arabidopsis met één tot meerdere cellen is echter te klein om te worden waargenomen en geanalyseerd. Bovendien kan de mutatie van bepaalde genen embryo letaliteit5, met vermelding van de sleutelrol van die genen in embryogenese veroorzaken. De karakterisering van patroonformatie in de embryo-dodelijke mutanten biedt een basis voor het begrijpen van het moleculaire mechanisme waarbij essentiële genen embryo-ontwikkeling reguleren.
Een gedetailleerde methode wordt hier beschreven voor de karakterisatie van embryo patroonformatie in de model plant Arabidopsis met directe microscopische observatie. Bij deze methode wordt de zaadknop goedkeuring, gevolgd door de microscopische observatie van een embryo. De karakterisering van een embryo-dodelijke mutant wordt ook beschreven.
De morfologie van de embryo's op verschillende ontwikkelingsstadia kan direct door microscopie met behulp van de zaadknoppen die zijn gewist met7-Hoyer van oplossing6,worden bepaald. Dergelijke zaadknoppen vertonen een hoge brekingsindex; Deze zaadknop clearing methode is dus vooral voordelig voor specimens die moeten worden nageleefd door differentiële interferentie contrast (DIC) microscopie.
Daarnaast is de genen die worden uitgedrukt in een bepaalde cel of een beperkt aantal cellen tijdens de embryogenese inzetbaar als markeringen te identificeren van de cellen in het embryo. Daarom kan de cel identiteit specificatie tijdens de embryogenese geannoteerde worden door het toezicht op de expressie van marker GFP-gelabelde proteïnen met behulp van confocale laser scanning microscopie. Omgekeerd, observeren het expressiepatroon van een onbekende functioneel gen -GFP fusion tijdens de embryogenese kan beschikken over een koppeling tussen embryo patronen en de uitdrukking van dit gen, en vergemakkelijken de identificatie van nieuwe genen die vereist voor embryogenese Arabidopsis zijn.
De hier beschreven methode kan ook worden gebruikt om karakteriseren het fenotype van een embryo-dodelijke mutant, te bepalen en het effect van het betrokken gen op embryogenese op cellulair niveau. Bovendien kan de methode in combinatie met een vergelijking van het expressiepatroon van markers tijdens de embryogenese tussen wild-type en mutant planten, aanwijzingen over de moleculaire mechanismen en signaalroutes betrokken bij embryogenese8,9opleveren. Inderdaad, de methode werd toegepast op naa10 planten, en het bleek dat de abnormale verdeling van de hypophysis in de mutant kan worden veroorzaakt door een wijziging in de verdeling van de auxine tijdens de embryogenese5.

Gezien het zeer voorspelbare karakter van hun ontwikkeling, zijn Arabidopsis embryo's gebruikt als een model voor studies van genuitdrukking in planten. Echter vroeg stadium plant embryo's zijn klein en bevatten weinig cellen, waardoor ze moeilijk te observeren en te analyseren. Een methode is hier beschreven voor het karakteriseren van patroonformatie in plant embryo's onder een microscoop met behulp van de model-organisme Arabidopsis. Na de goedkeuring van de verse zaadknoppen met behulp van Hoyer de oplossing, het mobiele nummer in en morfologie van embryo's kon worden waargenomen en hun ontwikkelingsstadium door differentiële interferentie contrast microscopie met behulp van een 100 X olie-immersie objectief kan worden vastgesteld. Bovendien was de expressie van specifieke marker eiwitten gelabeld met Green Fluorescent Protein (GFP) gecontroleerd om te annoteren cel identiteit specificatie tijdens embryo patronen door confocale laser scanning microscopie. Dus, deze methode kan worden gebruikt om te observeren patroonformatie in wild-type plant embryo's de cellulaire en moleculaire niveau, en te karakteriseren van de rol van specifieke genen in embryonale patronen door het vergelijken van patroonformatie in embryo's van wild-type planten en embryo-dodelijke mutanten. Daarom kan de methode worden gebruikt om te karakteriseren embryogenese Arabidopsis.
