Ovulum Abstand ist eine nützliche Methode für die Erkennung der Zellteilung und Beurteilung der Morphologie in der Embryogenese in Arabidopsis; mit dieser Technik können Embryonen direkt unter DIC Mikroskopie5,12beobachtet werden. Embryogenese ist das früheste Ereignis höherer Pflanzenentwicklung. Ein Reifen Embryo bildet sich aus einer Zygote durch Zellteilung und Differenzierung unter strengen genetischen Kontrolle1,2. Arabidopsis Embryos sind ein nützliches Modell für die Kontrolle der Morphogenese zu studieren, weil die Reihenfolge der Zellteilungen in der Embryogenese eine erwartete Muster3,4 folgt. Embryo Arabidopsis , enthält ein bis mehreren Zellen ist jedoch zu klein, um beobachtet und analysiert werden. Darüber hinaus kann die Mutation bestimmter Gene Embryo Letalität5, zeigt die wichtige Rolle der Gene in der Embryogenese verursachen. Die Charakterisierung der Musterbildung in der Embryo-letale Mutanten bietet eine Grundlage für das Verständnis der molekularen Mechanismus, wobei wesentlichen Gene, Entwicklung des Embryos regulieren.
Eine detaillierte Methode wird hier für die Charakterisierung der Embryo Musterbildung in der Modellpflanze Arabidopsis durch direkte mikroskopische Beobachtung beschrieben. Die Methode beinhaltet Ovulum Abstand gefolgt von der mikroskopischen Beobachtung eines Embryos. Die Charakterisierung von einem Embryo-tödliche Mutant wird ebenfalls beschrieben.
Die Morphologie der Embryonen in unterschiedlichen Entwicklungsstadien kann direkt durch Mikroskopie mit den Eizellen die freigemachte mit Hoyers Lösung6,7bestimmt werden. Diese Eizellen weisen einen hohen Brechungsindex. Somit ist das Ovulum clearing-Methode besonders vorteilhaft für Exemplare, die von differential Interferenz Kontrast (DIC) Mikroskopie beachtet werden müssen.
Darüber hinaus können die Gene, die in einer bestimmten Zelle oder eine begrenzte Anzahl von Zellen in der Embryogenese exprimiert werden als Marker verwendet werden, um die Zellen in einem Embryo zu identifizieren. Daher kann die Zelle-Identität-Spezifikation in der Embryogenese durch Überwachung des Ausdrucks der GFP-Tags Markerproteine konfokale Laser-scanning-Mikroskopie mit annotiert werden. Umgekehrt kann beobachten das Expressionsmuster von einer unbekannten funktionale gen -GFP Fusion in der Embryogenese eine Verbindung zwischen Embryo Musterung und der Ausdruck dieses Gens und erleichtern die Identifizierung neuer Gene, die für die Embryogenese in Arabidopsiserforderlich sind.
Die hier beschriebene Methode kann auch verwendet werden, um den Phänotyp der eine Embryo-letale Mutanten zu charakterisieren und die Auswirkungen des betroffenen Gens auf Embryogenese auf zellulärer Ebene zu ermitteln. Darüber hinaus kann die Methode in Kombination mit einem Vergleich der Expressionsmuster von Markergenen in der Embryogenese zwischen Wildtyp und mutierte Pflanzen, Hinweise über die molekularen Mechanismen und Signalwege beteiligt Embryogenese8,9ergeben. In der Tat die Methode auf naa10 Pflanzen angewendet wurde, und festgestellt wurde, dass die abnorme Arbeitsteilung der Hypophyse in den Mutanten durch eine Veränderung der Auxin-Verteilung während der Embryogenese5verursacht werden kann.

Angesichts der sehr vorhersehbare Art ihrer Entwicklung, haben Arabidopsis Embryonen als Modell für Studien der Morphogenese in Pflanzen verwendet. Jedoch frühe Werk Embryonen sind klein und enthalten nur wenige Zellen, wodurch sie schwer zu beobachten und zu analysieren. Eine Methode wird hier beschrieben, zur Charakterisierung der Musterbildung im Werk Embryonen unter dem Mikroskop mit Hilfe der Modellorganismus Arabidopsis. Nach der Freigabe von frischen Eizellen mit Hoyer Lösung die Zellzahl in und Morphologie der Embryonen konnten beobachtet werden, und ihre Entwicklungsstufe konnte vom differential Interferenz Kontrast Mikroskopie mit 100 X Öl Immersion Objektiv festgestellt werden. Darüber hinaus war der Ausdruck der spezifischen Markerproteine getaggt mit grün fluoreszierenden Proteins (GFP) überwacht, um Zelle Identität Spezifikation beim Embryo Musterung mit Anmerkungen versehen durch konfokale Laser-scanning-Mikroskopie. Somit kann diese Methode, Musterbildung in Wildtyp Pflanze Embryonen auf zellulärer und molekularer Ebene zu beobachten und um die Rolle bestimmter Gene im Embryo Musterung zu charakterisieren, durch den Vergleich der Musterbildung in Embryonen aus Embryo-letale Mutanten und Wildtyp Pflanzen verwendet werden. Daher kann die Methode zur Charakterisierung der Embryogenese in Arabidopsisverwendet werden.
