Fröämnet clearance är en användbar metod för att upptäcka celldelning och utvärdera morfologi under embryogenes i Arabidopsis; med denna teknik, kan embryon observeras direkt under DIC mikroskopi5,12. Det kritiska steget för fröämnet clearance är steg 1.3.4. Den tid som krävs för fröämnet clearance i steg 1.3.4 är variabel. Embryogenes är den tidigaste händelsen i högre växt utveckling. En mogen embryo former från en zygot genom celldelning och celldifferentiering enligt strikt genetisk kontroll1,2. Arabidopsis embryon är en användbar modell för att studera kontroll av morfogenes eftersom sekvensen av celldelningar under embryogenes följer en förväntade mönster3,4. Ett Arabidopsis embryo som innehåller en till flera celler är dock för liten för att observeras och analyseras. Mutationen av vissa gener kan dessutom orsaka embryo dödlighet5, som anger nyckelroll som dessa gener i embryogenes. Karakterisering av mönster bildas i embryo-dödliga mutanter ger en grund för att förstå den molekylära mekanism varigenom viktiga gener reglerar embryots utveckling.
En detaljerad metod beskrivs här för karakterisering av embryo mönster bildas i modell växten Arabidopsis av direkt mikroskopisk observation. Metoden innebär fröämnet clearance följt av mikroskopisk observation av ett embryo. Karakterisering av ett embryo lethal mutant beskrivs också.
Morfologi av embryon i olika utvecklingsstadier kan bestämmas direkt genom mikroskopi med de fröämnen som har rensats med Hoyers lösning6,7. Sådan fröämnen uppvisar en hög brytningsindex; Detta fröämnet clearing metod är således särskilt fördelaktiga för exemplar som ska följas av differentiell störningar kontrast (DIC) mikroskopi.
De gener som uttrycks i en viss cell eller ett begränsat antal celler under embryogenes kan dessutom användas som markörer för att identifiera cellerna i ett embryo. Därför kan specifikationen cell identitet under embryogenes förses genom att övervaka uttrycket av GFP-märkta markör proteiner med confocal microscopy för laserscanning. Omvänt, observerar uttrycksmönstret av en okänd funktionell gen -GFP fusion under embryogenes kan ge en länk mellan embryo mallning och uttryck av denna gen, och underlätta identifieringen av nya gener som krävs för embryogenes i Arabidopsis.
Den metod som beskrivs här kan också användas att karakterisera fenotypen av ett embryo lethal mutant, och att fastställa påverkan av drabbade genen på embryogenes på cellnivå. Dessutom i kombination med en jämförelse av det uttryck pattern av markörgener under embryogenes mellan vildtyp och muterade växter, kan metoden ge ledtrådar om molekylära mekanismer och signalvägar involverade i embryogenes8,9. Faktiskt, metoden tillämpades på naa10 växter, och det konstaterades att onormala uppdelningen av hypofysen i mutant kan orsakas av en förändring i auxin fördelningen under embryogenes5.

Mycket förutsägbar med tanke på deras utveckling, har Arabidopsis embryon använts som modell för studier av morfogenes i växter. Dock tidigt skede växt embryon är små och innehåller några celler, vilket gör dem svåra att observera och analysera. En metod är beskrivs här karaktärisera mönster bildas i växten embryon under ett Mikroskop med modell organismen Arabidopsis. Efter avslutande av färska fröämnen Hoyers lösning, cell nummer i och morfologi av embryon kunde observeras, och deras utvecklingsstadier kunde fastställas med hjälp av differentiell störningar kontrast mikroskopi med hjälp av en 100 X olja nedsänkning linsen. Dessutom var uttrycket av specifika proteiner taggade med grönt fluorescerande Protein (GFP) övervakas för att kommentera cellen identitet specifikation under embryo mallning av confocal microscopy för laserscanning. Således kan denna metod användas att observera mönster bildas i vildtyp växt embryon på cellulär och molekylär nivå, och att karaktärisera rollen av specifika gener i embryo mönster genom att jämföra mönster bildas i embryon från vilda växter och embryo-dödliga mutanter. Metoden kan därför användas för att karaktärisera embryogenes i Arabidopsis.
