De methode die is beschreven in dit artikel voor het maken van een plant-on-a-chip-platform is uniek omdat het twee-laag ontwerpen grenzen de haren van de wortel tot een enkel imaging vliegtuig en het platform kan worden gedeconstrueerd en kan worden gebruikt als een basismateriaal voor hoge resolutie niet-optische beeldvorming . Fijne root functies vergroten water en verwerving van de nutriënten voor de plant, verkennen van nieuwe ruimtes van de bodem en het vergroten van de totale wortel oppervlakte De omzet van deze fijne root functies speelt een belangrijke rol bij het stimuleren van de ondergrondse voedselketen1 en het aantal fijne wortels in bepaalde plantensoorten is verwacht te verdubbelen onder verhoogde atmosferische kooldioxide2. Fijne wortels zijn over het algemeen gedefinieerd als kleiner dan 2 mm diameter, hoewel nieuwe definities pleiten voor het karakteriseren van de fijne wortels door hun functie3. Net als vele fijne wortels, root haren voorzien van de functie van de opname- en absorptie maar bezetten een veel kleinere ruimte met een bosdiameter volgorde van micron. Vanwege het kleine formaat, root haren zijn moeilijk te afbeelding in situ en zijn vaak over het hoofd gezien als een onderdeel van de algemene architectuur van de wortel in veld schaal experimenten en modellen.
Ex terra root haar onderzoeken, zoals zaailingen geteeld op agar platen, de wetenschappelijke gemeenschap hebben voorzien van waardevolle informatie over cellulaire groei en vervoer4,5. Terwijl agar platen toestaan wortelstelsel te worden beeld niet-destructieve wijze en in real time, bieden ze geen hoge omgevingscontrole voor de toevoeging van experimentele behandelingen zoals voedingsstoffen, plantenhormonen of bacteriën. Een opkomende oplossing tot het vergemakkelijken van hoge resolutie imaging terwijl de komst van microfluidic platforms voor plant studies ook bieden dynamische omgevingsbeheersing geweest. Deze platforms hebben ingeschakeld, het niet-destructieve groei en visualisatie van verschillende plantensoorten voor hoge throughput fenotypering6,7,8,9, geïsoleerde chemische behandelingen 10,11,,12van de metingen van de kracht en de toevoeging van micro-organismen13. Microfluidic platform ontwerpen hebben gericht op het gebruik van één open ruimte fluidic lagen waarin de wortels verspreiden kunnen, waardoor de haren van de wortel op drift in en uit optische focus tijdens groei of behandeling.
Hier presenteren we een procedure voor de ontwikkeling van een twee-laag microfluidic platform met behulp van foto- en soft-lithografie methoden die op eerdere plant-on-a-chip ontwerpen voortbouwt door het beperken van de zaailing root haren aan hetzelfde imaging vlak als de belangrijkste wortel. Dit kan we bijhouden root haar ontwikkeling in real time met hoge resolutie, en gedurende het proces van experimentele behandeling. Onze kweken methoden toestaan Arabidopsis thaliana zaailingen te worden ontkiemd uit zaad binnen het platform en gekweekte voor tot een week in een gehydrateerd en steriele omgeving, die het gebruik van spuit pomp apparatuur niet vereist. Zodra de time-lapse imaging experiment heeft gesloten, kan het platform die hier gepresenteerd worden geopend zonder verstoring van de positie van de fijnere root functies. Hierdoor is het gebruik van andere hoge resolutie beeldvormende methoden. Hier voorzien wij representatieve resultaten voor de kwantificering en visualisatie van root haar morfologie in dit platform door optische, scanning elektronen microscopie (SEM), en atomic force microscopie technieken (AFM).

Root haren verhogen wortel oppervlakte voor betere opname van water en nutriënten-absorptie door de plant. Omdat ze klein in grootte en vaak bedekt zijn door hun natuurlijke omgeving zijn, zijn root haar morfologie en functie moeilijk om te studeren en vaak uitgesloten van het plantenonderzoek. In de afgelopen jaren hebben microfluidic platformen een manier om te visualiseren wortelstelsel op hoge resolutie zonder verstoring van de wortels bij overdracht naar een imaging systeem aangeboden. Het microfluidic platform hier gepresenteerd bouwt op eerdere plant-on-a-chip onderzoek door het opnemen van een twee-laag-apparaat om te beperken van de belangrijkste wortel van Arabidopsis thaliana aan hetzelfde optische vlak als de haren van de root. Dit ontwerp maakt het mogelijk de kwantificering van root haren op een cellulaire en organel niveau en ook voorkomt dat de z-as drijven bij de toevoeging van experimentele behandelingen. We beschrijven hoe bewaart u de apparaten in een staand en gehydrateerd omgeving, zonder de noodzaak voor fluidic pompen, terwijl het handhaven van een milieu van de gnotobiotic voor de zaailing. Na de optische beeldvorming experiment, kan het apparaat worden gedemonteerd en gebruikt als een basismateriaal voor atomaire kracht of scanning elektronen microscopie terwijl fijn wortel structuren intact.

De twee-laag PDMS microfluidic apparaten hier beschreven hebben een hoge kanaal van 200 µm voor de belangrijkste wortel van Arabidopsis en een hoge kamer van 20 µm beperken lateraal groeiende root haren (figuur 1A).
