Dit manuscript introduceert een eenvoudige in vitro zaadknop teelt protocol dat is efficiënt voor gebruik in de beeldvorming van de live-cel zygoten en embryo's te ontwikkelen.

Het plan van de basis lichaam van een organisme evolueert van een eencellige zygote. In de meeste bedektzadigen, zygotic divisie genereert een apicale en een basale cel, die uitgroeien tot de spruit en wortel, respectievelijk1. Daarom is het belangrijk om te begrijpen hoe het lichaam van de plant wordt gevormd tijdens de embryogenese, maar er is niet een doeltreffend instrument te observeren direct de dynamiek van levende zygoten en embryo's, omdat ze de ontwikkeling van diep in de bloem. In verschillende soorten monocot, zoals maïs en rijst, is een in vitro bevruchting methode gevestigde2,3. In deze methode, geïsoleerde zaadcellen en eicellen worden gesmolten elektrisch of chemisch en de gegenereerde cel kan uitgroeien tot een vruchtbare plant. In dicot planten is er echter geen in vitro bevruchting methode die goede embryo's, vermoedelijk vanwege de niet-gesynchroniseerde celcyclus staat voor mannelijke en vrouwelijke gameten4,5 produceren kan. Daarnaast speelt het embryo-omringende weefsel (endosperm) belangrijke rollen in de embryonale ontwikkeling6.
In een model dicot soorten, A. thaliana, werd een cultivatiemethode in vitro ontwikkeld door te focussen op de hele zaadknop, waarin zowel de embryo en endosperm7. Dit systeem werd met succes gebruikt voor het analyseren van de gevolgen van verschillende chemische reagentia voor embryogenese, maar het is niet geschikt voor time-lapse denkbaar, omdat er een laag overlevingspercentage. Dus, een roman in vitro zaadknop Cultuurstelsel werd ontwikkeld om te beginnen zo vroeg als de zygote fase en produceren van vruchtbare planten op een hoge verhouding8. Na verschillende proeven, bleek dat Nitsch medium en trehalose aanzienlijk verbeterd de overlevingskans van zaadknoppen8. Bovendien, omdat de zaadknop groeit als het groeit en dus vaak uit de buurt van het gebied van de observatie van de Microscoop beweegt, werd een PDMS apparaat ontwikkeld om op te lossen de zaadknop in de middelgrote9. Het PDMS apparaat ingeschakeld de langdurige beeldvorming voor 3-4 dagen, die voldoende is om te traceren van de ontwikkeling van een zygote tot een hart-fase embryo. Met deze methode wordt het mogelijk om te visualiseren van de dynamiek van zygote polarisatie en embryo patronen, niet alleen onder normale omstandigheden, maar ook in de aanwezigheid van chemische inhibitoren of in verschillende mutant achtergronden8,,10 ,11.

In de meeste bedektzadigen, de zygote en embryo zijn diep verborgen in het weefsel van de moeder, en dus het is al lang een mysterie van hoe ze de ontwikkeling van dynamisch; bijvoorbeeld, hoe de zygote polariseert de as van het lichaam en hoe het embryo geeft verschillende cel lot tijdens orgel vorming vast te stellen. Dit manuscript een in vitro zaadknop cultuur wordt methode beschreven voor het uitvoeren van live-cel beeldvorming van het ontwikkelen van zygoten en embryo's van Arabidopsis thaliana. Het medium van de geoptimaliseerde teelt kan zygoten of vroege embryo's uitgroeien tot vruchtbare planten. Door het te combineren met een poly(dimethylsiloxane) (PDMS) apparaat voor de matrix van de micropillar, wordt de zaadknop gehouden in de vloeistof in dezelfde positie. Deze fixatie is cruciaal voor het observeren van de dezelfde zaadknop onder een Microscoop voor verscheidene dagen vanaf de zygotic divisie tot het late stadium van het embryo. De resulterende beeldvorming van de live-cel kan worden gebruikt om te controleren de real-time dynamiek van zygote polarisatie, zoals nucleaire migratie cytoskelet omlegging, en ook de timing van de celdeling en cel lot specificatie tijdens embryo patronen. Bovendien, deze zaadknop Cultuurstelsel kan worden gecombineerd met remmer behandelingen voor het analyseren van de gevolgen van verschillende factoren op embryo-ontwikkeling, en met optische manipulaties zoals laser verstoring te onderzoeken van de rol van cel-cel communicatie.

Met behulp van deze zaadknop Cultuurstelsel, kan deze methode de levende dynamiek van zygote polarisatie en embryo patronen traceren. Dit is een prestatie, want eerder er geen techniek was te visualiseren het real-time gedrag van de zygote en embryo, die diep in het weefsel van de moeder zijn verborgen.
