Traditionelle cellekultur på flad todimensional (2D) overflader, såsom en kultur parabol eller multi godt plader, kan næppe fremkalde celle adfærd tæt på deres oprindelige stater. Præcis sammenfatning af indfødte cellulære microenvironments, som består af forskellige celletyper, ekstracellulære matricer og bioaktive opløselige faktorer i tre-dimensionelle (3D) arkitekturer1,2,3 ,4, er afgørende for at konstruere biomimicking væv i vitro til applikationer i tissue engineering, regenerativ medicin, grundlæggende biologi forskning og drug discovery5,6,7 ,8,9.
I stedet for 2D cellekultur, 3D cellekultur er almindeligt anvendt til at fremme biomimetiske mikro-arkitektoniske og funktionelle funktioner i celler dyrkes i vitro. En populær 3D celle kultur metode er at samlede celler i spheroids7,8,9,10. Cellulære spheroids kunne være injiceres til tilskadekomne væv med øget cellulær fastholdelse og overlevelse i forhold til injektion af spredte celler. Dog føre ikke ensartet klumpformet størrelser og uundgåelige mekanisk skade pålagt celler ved væske shear kraft under injektion til dårlig celle terapeutiske virkninger11,12,13. Ligeledes, den iboende ikke ensartethed under sammenlægning af spheroids har lavet deres oversættelse til 3D cellebaserede høj overførselshastighed stof screening udfordrende10.
En anden metode til 3D cellekultur opnås med bistand af biomaterialer, som typisk indkapsler celler i vandig hydrogels eller porøse stilladser. Det giver mulighed for større fleksibilitet i konstruere 3D arkitekturer. For terapi leveres celler indkapslet i bulk stilladser normalt til dyrs krop via kirurgisk implantation, som er invasive og traumatisk, dermed begrænser dens bred oversættelse til sengen. På den anden side aktiverer vandige hydrogels minimalt invasiv terapi ved at indsprøjte celler suspenderet i hydrogel forløber løsning i dyrs kroppe, så i situ gellation via thermo-, kemisk eller enzymatisk crosslinking11. Men som celler er leveret mens hydrogel prækursorer er stadig i en vandig tilstand, de også udsættes for mekanisk shear under injektion. Ikke kun så kan kemisk eller enzymatisk crosslinking under i situ gellation af hydrogel også pålægge skader på celler inden for. For narkotika-screening problemer biomateriale-assisteret cellekulturer med ensartethed, kontrollerbarhed og produktivitet. Ved hjælp af hydrogels er celler typisk involveret under gellation, hvorved processen kan påvirke cellernes levedygtighed og funktion. Gellation under celle såning også hæmmer brugen af de fleste høj overførselshastighed udstyr, da hydrogel kan skal holdes på is til at forhindre gellation før celle såning, og hydrogel kan jam udlevering tips, som er normalt meget tynd til at sikre nøjagtighed for høj overførselshastighed screening. Præformerede stilladser kunne potentielt adskille biomateriale fabrikation procedurer fra cellekultur, men de fleste stillads-baserede produkter er tilgængelige som bulk materialer med relativt lavere overførselshastighed14.
For at overvinde nogle af manglerne ved nuværende 3D kultur metoder, har vi udviklet en microfabrication-cryogelation integreret teknologi til at fabrikere en off-the-shelf og brugervenlige microcryogel array chip15. I denne protokol, er gelatine valgt at eksemplificere microcryogel fabrikation teknik, som det er biokompatible, nedbrydelige, omkostningseffektive, og ingen yderligere ændring er nødvendig for celle vedhæftet fil. Andre polymerer af naturlige eller syntetiske kilder kan også bruges til fabrikation, afhængigt af programmet. Via denne teknologi, kan vi fremstille miniaturized og meget elastisk microcryogels med kontrollerbare størrelse, form og layout. Når læsset med en række forskellige celletyper, kunne 3D microtissues blive dannet til forskellige applikationer. Disse unikke funktioner aktiverer ønskede injektionsydelse, cellebeskyttelse og site-directed opbevaring efter injektion i vivo for forbedret terapeutiske virkninger. Ikke kun så kunne microcryogels behandles yderligere for at danne 3D microtissue arrays, der er kompatible med fælles laboratorieudstyr og instrumenter til at indse høj overførselshastighed cellekultur for alsidig stof screening og andre cellulære assays. Heri, skal vi detalje fabrikationsproces af microcryogels og dens efterbehandling som individuelle 3D microtissues eller 3D microtissue arrays for to vigtige applikationer, celleterapi og stof screening, henholdsvis10,15 .

3d microtissues for injectable regenerative therapy and high-throughput drug screening

For at opgradere traditionelle 2D cellekultur til 3D cellekultur, har vi integreret microfabrication med cryogelation teknologi til at producere kombineret makroporøs individuel cryogels (microcryogels), som kan være fyldt med en lang række celletyper til at danne 3D microtissues. Heri, præsenterer vi protokollen for at fremstille alsidige 3D microtissues og deres applikationer i regenerativ terapi og stof screening. Størrelse og form-styrbar microcryogels kan være fabrikeret på en array chip, hvilket kan være fældet off-chip som enkelte celle-belæsset luftfartsselskaber for injicerbare regenerativ terapi eller være yderligere samlet på-chip til 3D microtissue arrays for høj overførselshastighed stof screening. På grund af høj elastisk karakteren af disse individuel cryogels udviser de 3D microtissues stor injektionsydelse for minimalt invasiv celleterapi ved at beskytte celler mod mekaniske shear kraft under injektion. Dette sikrer øget celle overlevelse og terapeutiske effekt i mus lemmer iskæmi model. I mellemtiden, samling af 3D microtissue arrays i et standard format, 384-multi-godt letter brugen af fælles laboratoriefaciliteter og -udstyr, gør det muligt høj overførselshastighed stof screening på denne alsidige 3D celle kultur platform.
