Disse to programmene har svært ulike behov, ligger en felles mellom dem i behovet for en mer biomimetic dyrking tilstand å forbedre celle funksjoner19. Tradisjonelle cellekultur på flat todimensjonal (2D) flater, som en kultur parabol eller flere bra plater, kan knapt framprovosere celle atferd nær innfødt stater. Nøyaktig recapitulation av innfødte mobilnettet microenvironments, som består av ulike celletyper, ekstracellulære matriser og bioaktive løselig faktorer i tredimensjonale (3D) arkitekturer1,2,3 ,4, er avgjørende for å konstruere biomimicking vev i vitro for programmer i vev engineering, regenerativ medisin, grunnleggende biologi forskning og drug discovery5,6,7 ,8,9.
I stedet for 2D cellekultur, 3D cellekultur er mye brukt til å fremme biomimetic mikro-arkitektur og funksjonelle egenskaper celleområde kultivert i vitro. En populær 3D celle kultur metode er å samle celler i spheroids7,8,9,10. Mobilnettet spheroids kan injiseres skadet vev med forbedret mobilnettet oppbevaring og overlevelse i forhold til injeksjon av spredte celler. Imidlertid føre ikke-uniform spheroid størrelser og uunngåelig mekanisk skade pålagt celler med flytende skjær makt under injeksjon til dårlig celle terapeutiske effekter11,12,13. På samme måte, den iboende ingen-enhetlighet under samling av spheroids har gjort sin oversettelse til 3D cellebasert høy gjennomstrømming narkotikarelaterte screening utfordrende10.
En annen metode for 3D cellekultur oppnås med hjelp av biologisk materiale, som vanligvis omslutter celler i vandig hydrogels eller porøse stillaser. Det gir større flexibilities bygger 3D arkitekturer. For terapi leveres celler innkapslet i bulk stillaser vanligvis til dyr kroppen via kirurgisk implantasjon, invasiv og traumatisk, derfor begrense sin brede oversettelse til sengen. På den annen side, aktivere vandig hydrogels minimal invasiv behandling ved å injisere cellene suspendert i hydrogel forløper løsning i dyr organer, slik at i situ gelation via thermo-kjemisk eller enzymatisk crosslinking11. Men som cellene leveres mens hydrogel forløpere er fortsatt i vandig tilstand, er de også utsatt for mekaniske skjær under injeksjon. Ikke bare det, kjemiske eller enzymatisk crosslinking under i situ gelation av hydrogel kan også innføre skade celler innenfor. For narkotikarelaterte screening møter biomateriale-assistert cellekulturer problemer med ensartethet, kontrollerbarhet og gjennomstrømning. Bruker hydrogels, er cellene vanligvis involvert under gelation, som prosessen kan påvirke celle levedyktighet og funksjon. Gelation under cellen seeding hemmer også bruk av de fleste høy gjennomstrømming utstyr, siden hydrogel må holdes på is for å hindre gelation før cellen frø, og hydrogel kan jam dispensering tips, som er vanligvis svært tynn å sikre nøyaktighet for høy gjennomstrømming screening. Pre-formet stillaser kan potensielt skille biomateriale fabrikasjon prosedyrer fra cellekultur, men de fleste stillaset-baserte produkter er tilgjengelig som bulkgods med relativt lavere produksjon14.
For å overvinne noen av svakhetene ved dagens 3D kultur metoder, har vi utviklet en microfabrication-cryogelation integrert teknologi å utvikle en sokkel og brukervennlig microcryogel matrise chip15. I denne protokollen valgt gelatin formidle microcryogel fabrikasjon teknikk som det er biokompatible, nedbrytbare og kostnadseffektiv, og ingen ytterligere endringer er nødvendig for cellen vedlegg. Andre polymerer av naturlig eller syntetisk kilder kan også brukes for fabrikasjon, avhengig av programmet. Via denne teknologien, kan vi utvikle miniatyriserte og svært elastisk microcryogels med kontrollerbar størrelse, form og oppsett. Når lastet med en rekke celletyper, kan 3D microtissues genereres for forskjellige søknadene. Disse unike funksjoner aktiverer ønsket injectability, cellebeskyttelse og nettsted rettet oppbevaring etter injeksjon i vivo for forbedret terapeutiske effekter. Ikke bare det, microcryogels kan bearbeides videre for å danne 3D microtissue matriser som er kompatible med vanlige laboratorieutstyr og instrumenter å realisere høy gjennomstrømming cellekultur for allsidig narkotikarelaterte screening og andre cellulære analyser. Her, skal vi detalj fabrikasjon prosessen med microcryogels og dens etter behandling som individuelle 3D microtissues eller 3D microtissue matriser for to viktige programmer, cellen terapi og narkotikarelaterte screening, henholdsvis10,15 .

3d microtissues for injectable regenerative therapy and high-throughput drug screening

Du kan oppgradere tradisjonelle 2D cellekultur til 3D cellekultur, har vi integrert microfabrication med cryogelation teknologi for å produsere macroporous Mikroskala cryogels (microcryogels), som kan lastes inn med en rekke celletyper til 3D microtissues. Her presenterer vi protokollen for å dikte allsidig 3D microtissues og deres programmer i regenerativ therapy og narkotikarelaterte screening. Størrelse og form kontrollerbare microcryogels kan være laget på en rekke chip, som kan være høstet av prosessoren som individuelle celle-lastet bærer for injiserbare regenerativ therapy eller bli ytterligere montert på prosessoren til 3D microtissue matriser for høy gjennomstrømming narkotikarelaterte screening. I mellomtiden forenkler montering av 3D microtissue matriser i standardformat 384-multi-godt bruken av felles laboratoriefasiliteter og utstyr, slik at høy gjennomstrømming narkotikarelaterte screening på denne allsidige 3D celle kultur plattformen.
