인간 관절 연골 자체를 복구하는 능력이 부족합니다. 따라서 연골 퇴행은 치유 적 치료가 아니라 보존 적 치료로 치료됩니다. 현재, 생체 외 팽창 된 연골 세포 또는 골수 유래 중간 엽 줄기 세포 (BMSC)로 손상된 연골을 재생시키기위한 노력이 진행되고있다. 그러나, 이들 세포의 생존력 및 불안정성이 제한적이어서 연골 재건에 대한 적용이 제한적이다. 인간 유도 pluripotent 줄기 세포 (hiPSCs) 재생 응용 프로그램에 대한 새로운 대안으로 과학적 관심을 받았다. 무제한자가 재생 능력과 다 기능성을 가진 hiPSCs는 연골 수리를위한 새로운 대체 세포원으로 주목 받았다. 그러나 고품질의 연질 펠릿을 대량으로 얻는 것은 임상 적용에 중요한 과제입니다. 이 연구에서 우리는 연쇄상 구별을 위해 배아 체 (EB) 유래 증식 세포를 사용했다. 성공적인 연골 형성은 PCR 및알 시안 블루, 톨루이딘 블루, 콜라겐 타입 I 및 II에 대한 항체 (각각 COL1A1 및 COL2A1)로 염색 하였다. 우리는 제대혈 단핵 세포 유래 iPSCs (CBMC-hiPSCs)를 연골 알약으로 분화시키는 방법을 상세히 제시한다.
