De VCCC heeft een aantal voordelen in termen van Recapitulerend MEJs in vitro, maar er zijn punten van discussie bij de vaststelling als de VCCC kan worden gebruikt voor specifieke toepassing. In kleine diameter weerstand slagaders scheidt een dunne interne elastische lamina (IEL) endothelial (EG) en de zachte spiercellen (SMC). De cellen kunnen project door openingen in deze elastische matrix en leveren van directe cytoplasmatische verbindingen via gap junction kanalen1,2. Deze unieke structuur is de myoendothelial kruising (MEJ) genoemd. De MEJ is een klein (ongeveer 0,5 µm x 0,5 µm, afhankelijk van het vasculaire bed), cellulaire projectie bestaat voornamelijk uit endotheliale cellen, maar kan voortkomen uit gladde spieren ook3,4. Een aantal onderzoekers hebben aangetoond dat de enorme complexiteit van de netwerken die zich selectief bij de MEJ voordoen, waardoor zij een bijzonder belangrijke locatie voor het vergemakkelijken van de bi-directionele signalen tussen endotheel en gladde spieren5 signalering ,6,7,8,9,10.
Een mechanistische dissectie van signaalroutes op de MEJ is echter moeilijk in een intact slagader. Omdat de MEJ een cellulaire projectie is, is het momenteel niet mogelijk om te isoleren een in vivo MEJ van de vasculaire muur. Om deze reden werd de VCCC model1 ontwikkeld. Nog belangrijker is, de VCCC repliceert fysiologische endothelial morfologie11 en polarisatie van de signalen tussen de apicale en MEJ gedeelten van de cel-12. Het was dit unieke model, dat de ontdekking dat alfa hemoglobine in de endothelial cel vergemakkelijkt is, gepolariseerde aan de MEJ. Dit is in tegenstelling tot conventioneel gekweekte endotheliale cellen die niet Alfa hemoglobine13 uitdrukken doen. Voor onderzoekers microvasculaire endotheel kochten, kan het dienstig meer gebruik van endotheliale cellen die hebben zijn gekweekt in de VCCC, vooral als de bedoeling is om te ontleden signaalroutes die zich in een kleine diameter weerstand slagaders voordoen.
Met behulp van een stevige plastic invoegen met een filter met een kleine diameter poriën (0.4 µm in diameter) te co cultuur twee verschillende celtypes voorkomt dat de cellen migreren tussen lagen. Het resulteert in een 10 µm-afstand tussen de cellen, die is aanzienlijk langer dan in vivo, maar nog steeds wordt gerepliceerd veel van de in-vivo -kenmerken van MEJs, waaronder eiwitten lokalisatie en tweede messenger signalering1 , 14. verder de VCCC staat voor het targeten van cel type-specifieke signalering via de toevoeging van een agonist of -antagonist tot de specifieke cellulaire compartiment. Bijvoorbeeld, laden van de EG met BAPTA-AM aan chelaat de calcium, en het stimuleren van de SMC met phenylephrine14. In tegenstelling tot andere beschrijvingen van co cultuur modellen15,16,17,18,19,20,21biedt dit instructies over het isoleren van de verschillende fracties voor mRNA en eiwit, met inbegrip van de verschillende MEJ breuk binnen de poriën van het filter. De toevoeging van deze techniek aan de VCCC voorziet in specifieke onderzoek naar veranderingen in mRNA lokalisatie of transcriptie22, proteïne fosforylatie12,23, en eiwit activiteit12. Het myoendothelial junction (MEJ), een unieke signalering microdomain in kleine diameter weerstand slagaders, vertoont lokalisatie van specifieke proteïnen en signalering processen die vasculaire Toon en bloeddruk kunnen. Als het is een projectie van hetzij de cel endothelial of gladde spieren, en als gevolg van zijn kleine formaat (gemiddeld een oppervlakte van ~ 1 µm2), de MEJ is moeilijk om te studeren in afzondering. Echter hebben we een cel cultuur model genaamd de vasculaire co celcultuur (VCCC) waarmee in vitro MEJ vorming, endothelial cel polarisatie en dissectie van signalering eiwitten en processen in de vasculaire muur van verzet slagaders. De VCCC heeft een veelheid van toepassingen en kan aangepast worden aan verschillende soorten cellen. Het model bestaat uit twee celtypen gegroeid aan weerszijden van een filter met 0.4 µm poriën waarin de in vitro MEJs kunnen vormen. Hier beschrijven we hoe maak je de VCCC via beplating van cellen en isolatie van endotheel, MEJ, en glad spierweefsel fracties, die vervolgens kunnen worden gebruikt voor isolatie van de eiwit- of activiteit testen. Het filter met intact cel lagen kan worden opgelost, ingesloten en verdeelde immunefluorescentie p.a.. Nog belangrijker is, zijn veel van de ontdekkingen van dit model bevestigd met behulp van intact weerstand slagaders, de fysiologische relevantie onderstrepen.

De filter-inzetstukken gebruikt voor de VCCC hebben kleine, 0.4 µm gaten. Figuur 1A, een weergave van de nl-gezicht van de VCCC filter inzet, toont de poriën waarin de MEJ in vitrokan vormen. Het schema toont de zachte spiercellen verguld aan de onderkant van het filter een dag voordat de endotheliale cellen zijn verguld aan de overkant (figuur 1B).
