Eerder, de lokalisatie van een Golgi-eiwit onder de lichte microscopie was voornamelijk gekwantificeerd door de mate van correlatie of samenloop van het imago van het eiwit met de afbeelding van een Golgi-marker van bekende lokalisatie15,16, 17. De resulterende correlatie of overlappende coëfficiënt komt overeen met hoe dicht het testen eiwit is naar het Golgi-marker ruimtelijk. Er zijn ten minste drie waarschuwingen voor dez...

Het Golgi-complex speelt een essentiële rol in secretoire/endocytotische handel van eiwitten en lipiden (hierna cargos) in zoogdiercellen1,2,3. In het Golgi, zijn cargos niet alleen gesorteerd naar verschillende sub cellulaire compartimenten maar ook gewijzigd door verschillende soorten glycosylatie. Het zoogdieren Golgi-complex heeft talrijke lateraal verbonden Golgi stapels, die meestal bestaat uit 4-11 strak aaneengesloten en platte membraan zakjes genaamd cisternae. De serieel gestapelde Golgi-cisternae zijn verder gecategoriseerd, van het ene eind naar het andere, als cis, mediaal en trans-cisternae. Op de trans-kant van een Golgi-stack, de trans-meeste membraan OSS zich ontwikkelt tot een netwerk van buizen en reticulum membraan genoemd van de trans-Golgi network (TGN)4. Cargos afgeleid van het endoplasmatisch reticulum (ER) Voer in de secretoire traject, een Golgi-stack op de cis-zijde en vervolgens achter elkaar passeren mediaal en trans-cisternae. Cargos uiteindelijk verlaten het Golgi in de trans-Golgi of TGN wordt het plasmamembraan, de Endosomen of de secretoire korrels.
De moleculaire en cellulaire mechanismen van hoe cargos transit een Golgi-stack en hoe het Golgi onderhoudt haar cisternal organisatie blijven mysterieuze en momenteel nog steeds onder een verhit debat1. Een van de problemen op dit gebied is dat Golgi cisternae enkel worden onder de elektronenmicroscopie (EM) sinds de resolutie van een optische Microscoop opgelost kunnen (~ 200 nm) is onvoldoende om op te lossen van individuele Golgi cisternae (< 100 nm in beide cisternal dikte en afstand). Daarom worden de lokalisatie van de sub-Golgi van resident eiwitten en doorvoer cargos conventioneel bepaald door de immuno-goud EM. Echter, de immuno-goud-EM is zeer technisch veeleisend en het is dan het vermogen van de meeste cel biologie labs. Hoewel de resolutie van de EM sub nanometer kunnen, de resolutie die worden geboden door de immuno-goud EM wordt sterk belemmerd door de grootte van het antilichaam complexe (primaire en het secundaire antilichaam) en het gouden deeltje, en het kan slechter dan 20 nm. Bovendien, EM beelden worden verkregen 2D dun-profielen in plaats van een 3D totaalbeeld van het Golgi, die in onjuiste conclusies afhankelijk van de relatieve positie en oriëntatie van de 2D sectie5 resulteren kan. Bijvoorbeeld het bestuderen van een EM single-sectie is niet betrouwbaar onderscheiden van een blaasje van de orthogonale weergave van een zaadvormende aangezien beide identieke ronde membraan profielen kunnen weergeven. De recente komst van super resolutie microscopie technieken, zoals 3D-gestructureerde verlichting microscopie (3D-SIM), gestimuleerd emissie uitputting (STED), photoactivated lokalisatie microscopie (PALM) en stochastische optische wederopbouw microscopie) STORM), maakt het mogelijk om op te lossen van sub-Golgi structuren onder lichte microscopen6. Echter, er zijn ten minste vier nadelen die het gebruik ervan in de biologische studie van de cel van het Golgi aanzienlijk kunnen beperken. 1) huidige super resolutie technieken vereisen dure en speciale hardwareconfiguratie die buiten de meeste cel biologie labs. 2) speciale fluorescentie labeling protocollen zijn nodig voor sommige super resolutie technieken. 3) Hoewel, onder de beste conditie, beweren deze technieken 20-110 nm in ruimtelijke resolutie, is de praktische resolutie verkregen in echte monsters kunnen veel erger. 4) In vergelijking tot conventionele microscopie, deze super resolutie technieken nog steeds moeilijkheden ondervinden bij het uitvoeren van multicolor, 3D of live cel imaging, afzonderlijk of in combinatie. Waarschijnlijk belangrijker, zowel immuno-goud EM en de super resolutie microscopie technieken opleveren kwalitatieve in plaats van kwantitatieve lokalisatie gegevens.
Wilt gedeeltelijk het oplossen van problemen die hierboven vermeld, hebben we onlangs een conventionele de lichte microscopie van gebaseerde methode, die Golgi eiwit lokalisatie heet door imaging centra van massa (GLIM), systematisch en kwantitatief lokaliseren een Golgi ontwikkeld eiwit met een resolutie die gelijkwaardig is aan die van de immuno-goud EM7. Bij deze methode is het Golgi in gekweekte zoogdiercellen als Golgi mini-stacks verspreid door de behandeling van nocodazole, een microtubulus depolymerizing drug. Uitgebreide studies hebben aangetoond dat nocodazole-geïnduceerde Golgi mini-stacks (hierna Golgi mini-stacks) native Golgi stapels in zowel de organisatie als de cellulaire functies8,9,10lijken, 11. Het quotiënt (LQ) van de lokalisatie van een test-eiwit kan worden verkregen door middel van GLIM en het geeft de kwantitatieve sub-Golgi-lokalisatie. De numerieke waarden van LQs kunnen worden vergeleken en een LQ-database van meer dan 25 Golgi markers beschikbaar geweest.
In GLIM zijn Golgi mini-stacks triple-gelabeld door endogene of exogenously uitgesproken GM130, GalT-mCherry en het test-eiwit (x). GM130 en GalT-mCherry, cis- en trans-Golgi markeringen respectievelijk12,13, bieden referentiepunten. De drievoudige fluorescentie, rood (R), groene (G) en far-red (B), kunstmatig worden weergegeven als rood, groen en blauw, respectievelijk. Centrum van fluorescentie massa (hierna midden) is gekozen om sub pixelresolutie. Het Golgi-as wordt gedefinieerd als de vector vanuit het centrum van GM130 met die van GalT-mCherry. De mini Golgi-stack is gemodelleerd als een cilindrische structuur met oneindige draaisymmetrie rond de Golgi-as. Daarom kan een Golgi mini stack verder worden gemodelleerd als een eendimensionale structuur langs de Golgi-as. De LQ van het test-eiwit x wordt gedefinieerd als dx/d1, waarin dx de afstand van het centrum van x tot die van GM130, is terwijl d1 de afstand van het centrum van GalT-mCherry met die van GM130 is. Als het centrum van x af-as is, wordt de axiale projectie-afstand gebruikt voor de berekening. De variabelen, met inbegrip van Golgi as, axiale hoek, dx, d1, hoek α en β hoek, voor GLIM worden schematisch weergegeven in Figuur 1. LQ is onafhankelijk van het Golgi axiale hoek al Golgi mini-stacks willekeurig in een cel oriënteren.
Golgi mini-stacks verschijnen inhomogene in beelden. We ontwikkelden de drie criteria Schakel analyseren Golgi mini-stacks voor GLIM. 1) het signal-to-noise verhouding criterium, waarin de verhouding van de totale intensiteit van een Golgi mini stack naar de standaarddeviatie (SD) van de achtergrond ≥ 30 in elk kanaal is. Dit criterium is om de positie nauwkeurigheid van het massamiddelpunt, die afhangt van de signal-to-noise verhouding Golgi mini-stacks. 2) de axiale hoek of afstand criterium, waarvoor d1≥ 70 nm. d1 vermindert met de toename van de axiale Golgi-hoek. Wanneer de axiale hoek nadert 90° of verticaal, de mini stack wordt niet omgezet als d1 nadert 0. d1≥ 70 nm effectief in de buurt van verticale Golgi mini-stacks kunt uitsluiten. 3) het criterium van de co-lineariteit, waarin | tan α | of | tan β | ≤ 0,3 is. Dit criterium zorgt ervoor dat de drie centra van een mini stack voldoende co-lineaire voor onze eendimensionale model van het Golgi mini stack. Alle lichte microscopen last van chromatische aberratie die de relatieve posities van rode, groene en far-red fluorescentie centra ernstig kan verstoren. Chromatische aberratie van Microscoop systemen is experimenteel gekalibreerd door imaging 110 nm kralen, die triple-gelabeld door rode, groene en far-red fluorescentie zijn. Voor elke kraal afbeelding, het centrum van rood wordt gedefinieerd als de ware positie van de parel en chromatische-verschuivingen van groen en far-red centra door eerste-orde polynoom functies zijn gemonteerd. Centra van Golgi mini-stacks zijn onderworpen aan de polynomiale functies te corrigeren de chromatische-verschuivingen in groen en far-red kanalen.
Door middel van GLIM kunnen wij een resolutie van ~ 30 nm langs de as van het Golgi onder standaardomstandigheden. Nog belangrijker is, biedt het een planmatige methode kwantitatief kaart een Golgi-eiwit. GLIM kan worden uitgevoerd door conventionele microscopen, zoals breed-gebied of confocal microscopen, met behulp van gemeenschappelijke fluorescentie labeling protocollen. Beeldvorming en gegevensverwerking kunnen duren een uur zo kort. Door middel van GLIM, hebben we rechtstreeks aangetoond de geleidelijke overgang van de secretoire lading uit het cis- aan trans-kant van het Golgi-7.

quantitative localization of a golgi protein by imaging its center of fluorescence mass

Het Golgi-complex bestaat uit serieel gestapelde membraan-cisternae die verder kunnen worden onderverdeeld in sub-Golgi-regio's, met inbegrip van het GOS-Golgi, mediaal-Golgi, trans-Golgi en trans-Golgi-netwerk. Cellulaire functies van het Golgi worden bepaald door de karakteristieke verdeling van de resident eiwitten. De ruimtelijke resolutie van de lichte microscopie van conventionele is te laag om sub-Golgi structuur of cisternae te lossen. De elektronenmicroscopie immuno-goud is dus een methode van keuze te lokaliseren een eiwit op de sub-Golgi-niveau. Echter, de techniek en het instrument zijn dan het vermogen van de meeste cel biologie labs. We beschrijven hier onze onlangs ontwikkelde super resolutie methode genaamd Golgi eiwit lokalisatie door imaging centra van massa (GLIM) systematisch en kwantitatief lokaliseren een Golgi-eiwit. GLIM is gebaseerd op standaard fluorescentie labeling protocollen en conventionele breed-gebied of confocal microscopen. Het gaat om de kalibratie van chromatische-shift aberratie van de microscopische systeem, de Beeldacquisitie en de analyse na overname. De sub-Golgi-lokalisatie van een test-eiwit is kwantitatief uitgedrukt als het quotiënt van de lokalisatie. Er zijn vier belangrijkste voordelen van GLIM; het is snel, op basis van conventionele methoden en instrumenten, het resultaat localisatie is kwantitatieve en het biedt ~ 30 nm praktische resolutie langs de Golgi-as. Hier beschrijven we de gedetailleerd protocol van GLIM te lokaliseren een test Golgi eiwit.
