Dette manuskript beskrevet metoder for morfometrisk måling og cellulære analyse af murine mandibular condyles og hvaltænder. Radiografisk morfometrisk målingerne kan også bruges til at analysere andre knogler fra små forsøgsdyr. I TMJ er en unik bærende fælles beliggende i regionen kraniofacial og er dannet af fibrocartilage. MCC i TMJ er afgørende for ledfunktion, herunder uhindret kæbe bevægelse mens taler og masticating, men det er almindeligt påvirket af degenerative sygdomme, herunder slidgigt1. I TMJ har kapacitet til at tilpasse sig ydre stimuli og lastning ændringer, fører til strukturel og cellulære ændringer til komponenterne i MCC2,3,4,5. De bærende egenskaber af MCC kan forklares af vekselvirkninger mellem dets bestanddele, herunder vand, kollagen-netværk og tæt pakket proteoglycaner. The MCC har fire forskellige cellulære zoner, der udtrykker forskellige typer af kollagen og ikke-kollagen proteiner: 1) den overfladiske eller artikulære zone; 2) den proliferativ zone, der består af udifferentierede mesenchymale celler og der reagerer på lastning krav; 3) den prehypertrophic zone, der består af modne chondrocytter udtryk for kollagen type 2; og 4) den hypertrofisk zone, den region, hvor de hypertrofisk chondrocytter udtryk for kollagen type 10 dør og gennemgå forkalkning. Ikke-mineraliseret regionen er rig på proteoglycaner, som giver resistens over for trykstyrke styrker6.
Der er løbende mineralisering på Hypertrofiske zone af MCC, hvor overgangen fra chondrogenesis til osteogenesis opstår, garantere subchondral knogle af mandibular kondyl7robust mineral struktur. Cellulære ændringer i regionerne unmineralized og mineraliseret i sidste ende føre til morfologiske og strukturelle ændringer i mandibular kondyl og underkæben. Opretholdelsen af homøostase i alle cellulære regioner af MCC og mineralisering af subchondral del er afgørende for sundhed, bæreevne og integritet af TMJ.
Flere kollagen transgene musemodel (som beskrevet af Utreja et al.) 8 er et fantastisk værktøj til at bruge til at forstå ændringer i kollagen udtryk, fordi alle transgener udtrykkes i MCC. For en dybdegående histologisk vurdering, er histologiske pletter brugt til at studere matrix deposition, mineralisering, celleproliferation, og apoptose samt protein udtryk på de forskellige cellelag af MCC.
I dette bruges manuskript, histologiske og morfometrisk analyser til at evaluere cellulære og strukturelle ændringer i MCC og subchondral knoglen af den mandibular kondyl på mus. Derudover er en celle kvantificering metode, til at analysere fluorescerende histologiske billeder og for kortlægning af lys objektglas, beskrevet. Trykstyrke statisk TMJ lastning metode, som forårsager cellulære og morfologiske ændringer på MCC og subchondral knogle9, er også illustreret for at validere vores metoder.
De metoder, der beskrives her kan bruges til at bestemme morfometrisk og histologiske forandringer i mandibular kondyl og underkæben af gnavere eller at analysere andre regioner af endochondral ossifikation og morfologi af yderligere mineraliseret væv.

a morphometric and cellular analysis method for the murine mandibular condyle

Kæbeleddet (TMJ) har kapacitet til at tilpasse sig ydre stimuli, og indlæsning af ændringer kan påvirke placeringen af condyles, samt de strukturelle og cellulære komponenter af den mandibular kondylære brusk (MCC). Dette manuskript beskriver metoder til at analysere disse ændringer og en metode til at ændre lastning af TMJ i mus (dvs., trykstyrke statisk TMJ indlæsning). Den strukturelle evaluering illustreret her er en simpel morfometrisk tilgang, der bruger Digimizer software og er udført i røntgenbilleder af små knogler.
