Dit manuscript beschreven methoden voor de meting van Morfometrische en cellulaire analyse van lymfkliertest mandibulaire condyles en kaken. De radiografische Morfometrische metingen kunnen ook worden gebruikt voor het analyseren van andere beenderen van kleine proefdieren. Bovendien, de cellulaire analyse (cel kwantificering en kraakbeen afstand mapping) zijn niet beperkt tot de knaagdieren mandibulaire condyle, maar kan worden gebruikt voor het kwantificeren van de histologische secties van talrijke weefsels.

Het TMJ is een unieke dragende gezamenlijke gelegen craniofaciale regio en van fibrokraakbeen wordt gevormd. De MCC van de TMJ is essentieel voor gemeenschappelijke functie, met inbegrip van de onbelemmerde kaak verkeer tijdens het spreken en masticating, maar het wordt meestal beïnvloed door degeneratieve ziekten, met inbegrip van artrose1. Het TMJ heeft de capaciteit aan te passen aan externe stimuli en laden, wijzigingen leiden tot structurele en cellulaire veranderingen aan de componenten van de MCC2,3,4,5. De dragende eigenschappen van de MCC kunnen worden verklaard door de interactie tussen de onderdelen daarvan, met inbegrip van water, het collageen-netwerk, en de dichtbevolkte verpakt proteoglycans. De MCC heeft vier verschillende cellulaire zones die uitdrukking geven aan verschillende soorten collageen en niet-collageen eiwitten: 1) de oppervlakkige of articulaire zone; 2) de proliferatieve zone, samengesteld uit ongedifferentieerde mesenchymale cellen en die reageert op het laden van eisen; 3) de prehypertrophic zone, samengesteld uit volwassen chondrocyten uiting van collageen type 2; en 4) de hypertrofische zone, de regio waar de hypertrofische chondrocyten uiting van collageen type 10 sterven en het ondergaan van verkalking. De niet-gemineraliseerde regio is rijk aan proteoglycans waarmee de weerstand tegen druksterkte krachten6.
Er is continu mineralisatie in de hypertrofische zone van de MCC, waar de overgang van chondrogenesis naar osteogenesis optreedt, de robuuste minerale structuur van het subchondrale bot van de mandibulaire condyle7te garanderen. Cellulaire veranderingen in de unmineralized en gemineraliseerde gebieden uiteindelijk leiden tot morfologische en structurele veranderingen in de mandibulaire condyle en onderkaak. Onderhoud van de homeostase van alle cellulaire regio's van de MCC en de mineralisatie van de subchondrale gedeelte zijn essentieel voor de gezondheid, de draagkracht en de integriteit van de TMJ.
De meerdere collageen transgeen muismodel (zoals beschreven door Utreja et al.) 8 is een geweldig hulpmiddel gebruiken om wijzigingen in collageen expressie begrijpen omdat alle transgenen zijn uitgedrukt in de MCC. Voor een diepgaande histologische evaluatie, worden histologisch vlekken gebruikt om te studeren matrix depositie, mineralisatie, celproliferatie, en apoptose, evenals eiwit expressie in de cel van de verschillende lagen van de MCC.
In dit worden manuscript, histologisch en Morfometrische analyses gebruikt voor het evalueren van de cellulaire en structurele veranderingen in het MCC en subchondrale bot van de mandibulaire condyle van muizen. Daarnaast wordt een cel kwantificering, voor het analyseren van fluorescerende histologische beelden en voor het toewijzen van lichte Microscoop dia's, beschreven. De druksterkte statische TMJ laden methode, waardoor cellulaire en morfologische veranderingen op het MCC en subchondrale bot9, wordt ook geïllustreerd voor het valideren van onze methoden.
De hier beschreven methoden kunnen worden gebruikt om te bepalen Morfometrische en histologische veranderingen in de mandibulaire condyle en onderkaak van knaagdieren of andere regio's in endochondral ossificatie en de morfologie van extra gemineraliseerde weefsels te analyseren.

a morphometric and cellular analysis method for the murine mandibular condyle

Het kaakgewricht (TMJ)-gewricht heeft de capaciteit aan te passen aan externe prikkels, en laden van wijzigingen kan invloed hebben op de positie van condyles, alsook de structurele en cellulaire componenten van de mandibulaire Condylaire kraakbeen (MCC). Dit manuscript wordt beschreven methoden voor het analyseren van deze veranderingen en een methode voor het wijzigen van het laden van de TMJ in muizen (dat wil zeggen, druksterkte statische TMJ laden). De structurele evaluatie geïllustreerd hier is een eenvoudige Morfometrische-aanpak die gebruikt de Digimizer-software en wordt uitgevoerd in de röntgenfoto's van kleine beenderen. Bovendien, de analyse van cellulaire verandert leiden tot wijzigingen in collageen expressie, bot remodelleren, celdeling en Proteoglycaan distributie in het MCC wordt beschreven. De kwantificering van deze wijzigingen in histologische afdelingen - door het tellen van de positieve fluorescerende pixels met behulp van software en het meten van de afstand toewijzing image & gekleurd gebied met Digimizer - wordt ook aangetoond. De methoden die hier worden weergegeven zijn niet beperkt tot de lymfkliertest TMJ, maar kon worden gebruikt op de extra botten van kleine proefdieren en in andere regio's van endochondral ossificatie.

Beschrijvende statistiek werden te onderzoeken van de verdeling van de Morfometrische metingen (mandibulaire lengte, Condylaire lengte, Condylaire breedte) en histologische analyses uitgevoerd.
