Partiell hepatectomy är en etablerad teknik för utredande lever regenerering och överdriven hepatectomy rapporteras att efterlikna AHF modellen. För närvarande föreslås hepatocyte transplantation som ett alternativ till hela organtransplantation för behandling av patienter med allvarliga leversjukdomar. Man tror att det kan överbrygga patienter till hela organtransplantation1. Förutom de prövningar hepatocyter2utreds xenogen hepatocyter3 och hepatocyter härrör från stamceller4 också i djurmodeller. I detta sammanhang, homing och engraftment är potential de transplanterade cellerna i mottagaren ett viktigt kriterium för cellterapi baserat i akut leversvikt (AHF).
För att utreda transplantation av hepatocyter eller hepatocyte-liknande celler5, skapas AHF i en djurmodell av kirurgiska6 eller farmakologiska7 förfaranden, följt av omplantering celler. Partiell hepatectomy är en mångsidig och reproducerbar metod att studera lever regenerering och effekten av cellbaserade therapeutics i olika sjukdomstillstånd. Partiell hepatectomy underlättar också ökad engraftment och spridningen av transplanterade celler av accelererande kärlnybildning och cellmigration mot levern. Här beskriver vi ett enkelt protokoll för utför 30% hepatectomy och transplantation av celler i mjälten hos en icke-feta diabetiker/svår kombinerad nedsatt immunförsvar nicka. SCID (NICKA. Två små snitt görs i den här proceduren. Första snittet är att exponera och resect den vänstra LOB i levern, och en annan liten snitt att exponera mjälte för intrasplenic transplantation av celler. Detta förfarande kräver inte någon specialiserad kirurgisk kompetens, och det kan fyllas i 5-7 minuter med mindre stress och smärta, snabbare återhämtning och bättre överlevnad.
