Glioblastoom (GBM), de meest voorkomende en kwaadaardige hersenkanker, wordt gekenmerkt door een zeer diffuse invasie van het primaire tumorvolume in het omliggende gezonde hersenparenchym 1 , 2 . Deze diffuse invasie maakt de tumor bijzonder moeilijk om volledig te resecteren en de invasieve kankercellen die post-therapie blijven, is de meest voorkomende reden voor onvermijdelijk herhaling 2 , 3 , 4 . Vroeger vonden we de diffuse glioma-celinval te remmen om therapeutisch voordelig te zijn 5 , maar weinig bekend is over de complexe mechanismen die bijdragen aan GBM-invasie. De tumor micro-omgeving, of weefsel rondom de kanker, is betrokken bij de progressie van tumoren bij meerdere kankers 6 , 7 . De glioblastoom tumor micro-omgeving, in pArticulair, is relatief onderschat en is uniek complex, het vormen van meerdere gliale cellen, zoals astrocyten, microglia en oligodendrocyten, evenals extracellulaire matrix, oplosbare factoren en biofysische factoren. Experimenteel zijn astrocyten en microglia aangetoond dat de progressie van gliomen en invasie 8 , 9 , 10 toeneemt, maar de samenstelling van alle gliale cellen in de micro-omgeving van de menselijke hersenen is onbekend. We hebben eerder laten zien dat micro-componenten de overleving van patiënten kunnen voorspellen door cellulaire componenten van de glioblastoom micro-omgeving kwantitatief te analyseren en onze analyses te integreren in een proportioneel risico model 11 . Hier beschrijven we de kwantitatieve analysemethode voor het identificeren van de populaties van glialcellen binnen het tumorvolume en aangrenzende gebieden van tumorresecties van glioblastoom patients. We gebruikten chromogene immunohistochemie om de gliale celpopulaties en ImageJ te identificeren om de procentuele dekking van kleuring voor elke gliale populatie te analyseren. Het beoordelen van de procentuele dekking creëert een simpele meting voor het bepalen van de morfologische verschillen in cellen, met name die welke beïnvloed zijn door interacties met kankercellen. Eerdere studies voor het kwantificeren van histopathologische kleuring gebruiken standaard kleuring zoals hematoxyline en eosin 12 of Masson&#39;s trichroom 13 , die niet profiteren van de specificiteit van immunohistochemische kleuring op antilichamen. Onze methode is ontwikkeld om de glialpopulaties rechtstreeks te kwantificeren binnen glioblastoom patiënten tumor resecties, die we willen gebruiken om de complexe glioblastoom micro-omgeving te verhelderen.

Met de groeiende interesse in de micro-omgeving van de tumor, ontwikkelden we een methode om de micro-milieukomponenten specifiek te bepalen in patiëntenmonsters van glioblastoom, de dodelijkste en meest invasieve hersenkanker. Niet alleen zijn kwantitatieve methoden die gunstig zijn voor het nauwkeurig beschrijven van zieke weefsels, ze kunnen ook potentieel bijdragen tot een nauwkeuriger prognose, diagnose en de ontwikkeling van weefsel-gemanipuleerde systemen en vervangingen. Bij glioblastoom zijn gliale cellen, zoals microglia en astrocyten, onafhankelijk gecorreleerd met een slechte prognose gebaseerd op patholoog-gradering. Echter, de toestand van deze cellen en andere gliale celcomponenten is niet kwantitatief goed beschreven. Dit kan moeilijk zijn door de grote processen die deze glialcellen markeren. Bovendien richten de meeste histologische analyses op het algehele weefselmonster of alleen in het grootste deel van de tumor, in tegenstelling tot het kwantificeren van kwantificaties op basis van regio&#39;s witHin het zeer heterogene weefsel. Hier beschrijven we een methode voor het identificeren en kwantitatief analyseren van de populaties van gliale cellen in het tumorvolume en de aangrenzende gebieden van tumorresecties van glioblastoompatiënten. We gebruikten chromogene immunohistochemie om de gliale celpopulaties bij patiënten tumor resecties en ImageJ te identificeren om de percentuele dekking van kleuring voor elke gliale populatie te analyseren. Met deze technieken kunnen we de glialcellen beter omschrijven doorheen de regio&#39;s van de glioma tumor micro-omgeving.

Voor deze analyse werden twee regio&#39;s van belangen binnen onze tumorresecties - het primaire tumorvolume en de aangrenzende gebieden, hoofdzakelijk samengesteld uit gezond weefsel met diffuse invallende kankercellen ( Figuur 1A , 1B ) - geïdentificeerd door samenwerkende neuropathologen op hematoxyline en eosin-gekleurde patiënt samples.
