El glioblastoma (GBM), el cáncer de cerebro más común y maligno, se caracteriza por una invasión altamente difusa desde el tumor primario al parénquima cerebral sano circundante 1 , 2 . Esta invasión difusa hace que el tumor sea particularmente difícil de resecar completamente, y las células cancerosas invasoras que permanecen después de la terapia es la razón más común para la recurrencia inevitable 2 , 3 , 4 . Anteriormente, se encontró inhibir la invasión difusa de células de glioma de ser terapéuticamente beneficioso [ 5] , sin embargo poco se sabe sobre los complejos mecanismos que contribuyen a GBM invasión. El microambiente tumoral, o tejido que rodea el cáncer, ha sido implicado en la progresión de tumores en múltiples cánceres [ 6 , 7] . El microambiente tumoral del glioblastoma, en pArticular, está relativamente sub-caracterizada y es singularmente compleja, compuesta de múltiples células gliales, tales como astrocitos, microglia y oligodendrocitos, así como matriz extracelular, factores solubles y factores biofísicos. Experimentalmente, se ha demostrado que los astrocitos y microglia aumentan la progresión e invasión de glioma 8 , 9 , 10 , pero se desconoce la composición de todas las células gliales en el microambiente humano nativo del cerebro. Anteriormente hemos demostrado que los componentes microambientales pueden predecir la supervivencia de los pacientes mediante el análisis cuantitativo de los componentes celulares del glioblastoma microambiente e incorporar nuestros análisis en un modelo de riesgos proporcionales [ 11] . En este sentido, describimos el método de análisis cuantitativo para identificar las poblaciones de células gliales dentro de la masa tumoral y regiones adyacentes de resecciones tumorales de pacientes con glioblastomaS Se utilizó inmunohistoquímica cromógena para identificar las poblaciones de células gliales y ImageJ para analizar el porcentaje de cobertura de tinción de cada población glial. La evaluación del porcentaje de cobertura crea una medición simple para determinar las diferencias morfológicas de las células, particularmente las afectadas por las interacciones con las células cancerosas. Los estudios previos para cuantificar la tinción histopatológica usan tinción estándar como la hematoxilina y la eosina 12 o el tricromo 13 de Masson, que no aprovechan la especificidad de la tinción inmunohistoquímica basada en anticuerpos. Nuestro método fue desarrollado para cuantificar directamente las poblaciones gliales dentro de las resecciones tumorales de pacientes con glioblastoma, que se pretende utilizar para elucidar el complejo microambiente glioblastoma.

Con el creciente interés en el microambiente tumoral, nos propusimos desarrollar un método para determinar específicamente los componentes del microambiente dentro de muestras de pacientes de glioblastoma, el cáncer de cerebro más mortífero y más invasivo. No sólo son métodos cuantitativos beneficiosos para describir con precisión los tejidos enfermos, sino que también pueden contribuir a un pronóstico más preciso, el diagnóstico y el desarrollo de sistemas de ingeniería de tejidos y reemplazos. En el glioblastoma, las células gliales, tales como microglia y astrocitos, se han correlacionado independientemente con un mal pronóstico basado en la clasificación de patólogos. Sin embargo, el estado de estas células y otros componentes de las células gliales no ha sido bien descrito cuantitativamente. Esto puede ser difícil debido a los grandes procesos que marcan estas células gliales.
