Dessa studier omfattar beteende analyser, kalcium imaging studier eller ens filmvisningar för specifika fenotyper, vilket resulterar i mer exakta mätningar av experimentella resultat1,2,3,4, 5,6. Kalcium imaging studier använder genetiskt kodade kalcium indikatorer (GECIs) uttryckta i målceller för att mäta neural dynamiken i dessa celler i realtid8,9,10,11. Transparent beskaffenhet C. elegans tillåter inspelning av fluorescerande nivåerna av dessa proteiner i levande djur. Traditionellt GECIs är beroende av grönt fluorescerande protein (GFP)-baserat sensor GFP-Calmodulin-M13 peptid (GCaMP), men nyare studier har anpassat dessa sensorer som möjliggör bättre signal-brus-förhållanden och röd-skiftat excitation profiler. Efter utvecklingen av GCaMP3, proteiner med dessa specifikationer har varierat, inklusive sensorer såsom GCaMP6s och GCaMP6f (långsamma och snabba fluorescens off-priser, respektive), samt RFP-Calmodulin-M13 peptid (troppy), som har en röd-skiftat aktivering profil. Kombinationen av dessa GECIs med C. elegans -specifika promotorn gensekvenser kan rikta celler av intresse, särskilt sensoriska nervceller12,13,14,15 , 16.
Medan C. elegans användarvänlighet i ultrakalla studier framgår, har nästan alla studier fokuserat på hermafroditer. Trots hanar endast står för 0,01-0,02% av vildtyp befolkningen, ovärderlig fynd kan uppstå från deras karakterisering. Medan den fysiska connectome hermafrodit nervsystemet har mappats fullt för årtionden17, förblir den manliga connectome ofullständig, särskilt i regionen huvud av djur18. Användning av kalcium imaging i män hjälper till att skapa förståelse för manliga nervsystemet och de skillnader som uppstår mellan de två könen. Den mindre storleken C. elegans vuxna män förhindrar effektivt och tillförlitligt svällning i lastningen hamnar i traditionella olfactory anordningar som är avsedda för större hermafroditer. För att åtgärda detta, en modifierad version av Chronis Olfactory Chip19 utvecklades med en smalare lastning hamn, en lägre kanal höjd, och vänder i masken lastningshamnen (som rotera djuret), vilket möjliggör visualisering av bilaterala vänster/höger neuronala par. Denna design tillåter: (1) effektiv fångst av unga vuxna män, (2) en mer pålitlig orientering av djuret för visualisering av båda medlemmar av bilaterala ihopkopplade neuroner, och (3) exakt avbildning av neural aktivitet i manliga nervceller.
Allt fler visar studier att C. elegans hanar svara annorlunda än hermafroditer på en mängd ascarosides (ascr) eller nematod feromoner20,21,22,23 ,24. Därför blivit utveckla en förståelse för de neurala dynamics och representationer inom den manliga connectome ännu mer relevant. Manliga C. elegans innehåller 87 kön-specifika nervceller inte närvarande i den hermafrodita25,26, förändra connectome i som-ännu obestämt sätt. Att kunna bild dessa unika neurala dynamics ger oss möjlighet att bättre förstå kön-specifika svaren och neurala representationer.
Det här protokollet beskriver användningen av en hane-anpassad olfactory chip för den neurala avbildning av manliga C. elegans chemosensation. Användningen av kalcium indikatorer har kraftigt ökat vår förståelse av neurala dynamics och förordning. Nematoden Caenorhabditis elegans, med dess helt mappade nervsystemet och transparent anatomi, presenterar en idealmodell för att förstå realtid neurala dynamics använder kalcium indikatorer. I kombination med mikroflödessystem teknik och experimentell design utförs kalcium-imaging studier med dessa indikatorer i både fri-flytta och fångade djur. De flesta tidigare studier utnyttjar svällning enheter, till exempel lukt chip beskrivs i Chronis et al., har dock utrustning avsedd för användning i det vanligare hermafroditt, liksom den mindre vanliga manlign både morfologiskt och strukturellt olika. En anpassad olfactory chip ritades och fabricerade för ökad effektivitet i manliga neuronala imaging med unga vuxna djur. En sväng införlivades masken laddar port att rotera djuren och tillåta för separation av enskilda nervceller inom ett bilateralt par i 2D imaging. Maskar är utsatta för ett kontrollerat flöde av luktämnet inom mikroflödessystem enheten, som beskrivs i föregående hermafrodit studier. Kalcium transienter analyseras sedan med hjälp av öppen programvara ImageJ. Det förfarande som beskrivs häri bör möjliggöra en ökad mängd hane-baserade C. elegans kalcium imaging studier, fördjupa vår förståelse av kön-specifika nervcellernas signalering mekanismer.
