Die "Frog-Community" ist jetzt an einem Wendepunkt angelangt wo ermöglicht grundlegende Genom Änderung Technologie das Studium der Genfunktion, praktisch nach Belieben. Programmierbare CRISPR/Cas9 jedoch haben die Mutagenese von Genen hocheffizient mit biallelische Mutationen möglich in den meisten Zellen der Tier4,5,6,7gemacht, 8,9,10,11. Diese Studien, untermalt von Bhattacharya Et al. 12 und Shigeta Et al. 13, haben gezeigt, dass die Funktion vieler Gene durch Mutagenese in F0 Mosaik Tiere untersucht werden kann. Dieser Ansatz hat viele Vorteile; Cas9-SgRNA-mikroinjiziert Embryonen werden jedoch oft Variable Phänotypen aufgrund unvollständiger Funktionsverlust (LOF) angezeigt. Noch bedeutsamer ist, die Generation der mutierten Linien ist ein großer Vorteil für einige Anwendungen – insbesondere bei der Gene, die einen mütterliche mRNA-Beitrag zu studieren. Mütterliche mRNAs und Proteine und ihre Einflüsse auf die Epigenetik, bleiben über einen längeren Zeitraum in Embryogenese14,15,16, Darstellung der frühen Entwicklungsstörungen Beiträge vieler Gene feuerfeste F0 Analysen. Daher sind andere LOF Ansätze erforderlich.
Bei der Suche nach mutierten Linien zu erstellen, muss der Pfad auf die Erlangung von homozygot LOF Embryonen mehrere Hindernisse. Erste, effiziente Mutagenese biallelische Mutationen produzieren kann nachteilig sein, weil Verlust von wesentlichen Genfunktionen scheitern, Geschlechtsreife, stören die Herstellung einer tragfähigen Linie überleben führt. Eine gemeinsame Lösung ist die sorgfältige Titration der Höhe der Cas9-SgRNA geliefert. Hier ist das Ziel, ein Gleichgewicht zwischen reduziert Letalität bei gleichzeitiger Maximierung der Keimbahn Mutagenese Effizienz auch. Ein zweites Problem ergibt sich aus der standard Zucht-Schema, wo phänotypische Analysen bis F-2 -Generation aufgeschoben. Mit dem standard-Ansatz, sexuell Reifen Sie F0 Tiere, die Mutante zu übertragen, die Allele durch die Keimbahn outcrossed sind zur Herstellung F1 heterozygote "Träger", die dann zur Geschlechtsreife angebaut werden. Zwei F-1 -heterozygot sind dann intercrossed, um F2 mutierten Embryonen mit einer erwarteten Mendelian Frequenz von 25 % zu produzieren. So sind zwei Generationen der Zucht notwendig für die Analyse der Mutanten Phänotypen. Mutierte Tiere könnte entweder homozygoten oder compound-heterozygot (d. h. nachkommen, enthält zwei verschiedene mutierten Allele, die abhängig von der Genotypen der Elterntiere in der F1 Intercross verwendet).
Diese Hindernisse können überwunden werden, durch die Beschränkung programmierbare Nuklease-vermittelten Mutagenese, Keimzellen, die eine neue Methode namens sprunghaften17zugrunde liegt. Leap Frogging besteht aus zwei Hauptkomponenten: (1) die Mikroinjektion von Cas-mRNA zusammen mit SgRNA oder Nuklease-SgRNA-komplexe (oder, im Prinzip, TALENs oder Zink-Finger-Nukleasen) Zeitpunkt der einzelligen effizient embryonale Genom mutagenize, gefolgt von (2) die Transplantation von PGCs auf Wildtyp Geschwister Embryonen, wo die endogenen PGCs entfernt wurden. Wenn beide Schritte sind effiziente und vollständige Keimbahn-Ersatz mit mutierten Keimzellen erhalten. Blackler bewiesen in den frühen 1960er Jahren, dass Xenopus PGCs zwischen den Embryonen im späten Neurula und frühen Tailbud Stufen18,19transplantiert werden konnte. Für Leap Frogging, wurde Blacklers Ansatz modifiziert durch Ausführen der Transplantationen im Blastozystenstadium Stufe17, wenn PGCs im Vegetal Pol der Embryo20,21lokalisiert sind. Transplantation vor Gastrulation hat zwei wesentliche Vorteile. Zunächst fand das Engraftment Transplantationen und die anschließende normale Entwicklung effizienter sein, wenn die Transplantation im Blastozystenstadium (unveröffentlichte Beobachtungen) durchgeführt wird. Zweitens durch Blastozystenstadium Transplantationen durchführen, kurz nachdem zygotic Transkription begonnen hat, vermeiden man die Letalität aus Entwicklungsstörungen Gen-Mutationen, die Gastrulation oder das stören sonst zu fehlerhaften späten Neurulae führen. PGC Transplantation-Lager ("leapfrogged") Embryonen sind gewachsen, Geschlechtsreife und Intercrosses zwischen diesen F0 Tieren haben gezeigt, dass in vielen Fällen 100 % der F-1 -Nachkommen den LOF-Phänotyp (die meisten werden zusammengesetzte anzeigen Heterozygoten), komplette Keimbahn Ersetzung durch gezielte Mutationen angibt.
Es wird erwartet, dass Leap Frogging gentechnische Ansätze im Xenopusbeschleunigen wird. Leap Frogging bietet auch eine Alternative zu den "host-Transfer" Methode22 für die LOF-Analyse von Genen mütterlicherseits-Effekt (unveröffentlichte Beobachtungen). In der aktuellen Publikation präsentiert eine detaillierte Beschreibung der Methode, vor allem mit Schwerpunkt auf PGC Transplantation, in X. tropicalis (mit geringfügigen Änderungen für laevis). Die Transplantation von PGCs beweist hier um einen schnelleren Transfer dieser Technologie an andere Laboratorien arbeiten mit Xenopuszu erleichtern. Die Prinzipien dieser Methode sollte erfolgreich in anderen Amphibien (z.B. Urodeles) und Organismus-spezifische Änderungen in der Methodik dürfen für die Anwendung auf vielen anderen Tieren, die in der effizienten Mutagenese erreicht werden kann und PGCs sind leicht transplantierbaren.

Die Schaffung von mutierten Linien von Genom-Bearbeitung beschleunigt genetische Analyse in vielen Organismen. CRISPR/Cas9 Methoden wurden für den Einsatz in der afrikanischen krallenbewehrten Frosch, Xenopus, langjährige Modellorganismus für die biomedizinische Forschung angepasst. Klassische Züchtung Regelungen für die Erstellung von homozygoter Mutante Linien mit CRISPR/Cas9-gezielte Mutagenese haben mehrere zeitaufwändige und mühsame Schritte. Zur Erleichterung der Schaffung von mutierten Embryonen, besonders um die Hindernisse, die Gene, die für die Embryogenese, ausschlagen zugeordnet wurde eine neue Methode namens Leap Frogging entwickelt. Diese Technik nutzt die Robustheit des Xenopus Embryonen "Ausschneiden und einfügen" embryologischen Methoden. Leap Frogging nutzt die Übertragung der primordialen Keimzellen (PGCs) aus effizient mutagenisierter Spender Embryonen in PGC abgetragen Wildtyp Geschwister. Diese Methode ermöglicht die effiziente Mutation des wesentlichen Gene durch die Schaffung von Chimären Tiere mit Wildtyp-Körperzellen, die ein mutiertes Keimbahn tragen. Wenn zwei F0 Tiere tragen "leapfrog Transplantationen" (d. h. mutierten Keimzellen) sind intercrossed, sie produzieren homozygote oder compound heterozygote, null F1 Embryonen, spart eine ganze Generationszeit, phänotypische Daten zu erhalten. Leap Frogging bietet auch einen neuen Ansatz für die Analyse von mütterlicher Effekt Gene, die refraktär gegenüber F0 phänotypische Analyse nach CRISPR/Cas9 Mutagenese sind. Nach Transplantation sollte die qualitative Bestimmung der Wirksamkeit von CRISPR/Cas9 Mutagenese vor Aufwendung den Aufwand bei der Tierhaltung zu wachsen und pflegen die Tiere zur Geschlechtsreife durchgeführt werden.
